Esta pesquisa recebeu aprovação do Comitê de Ética em Experimentação Animal da Kunming Medical University (kmmu20220945). Todos os experimentos seguiram as diretrizes estabelecidas para os cuidados com os animais. Células endoteliais linfáticas leptomeníngeas (LLECs) foram coletadas de camundongos machos C57Bl/6J com idade entre 6-8 semanas e peso entre 20-25 g. Esses camundongos foram adquiridos da Universidade de Medicina de Kunming, em Kunming, na China. Todo o procedimento experimental foi conduzido sob rigorosas condições estéreis. Todas as etapas de centrifugação são realizadas à temperatura ambiente, salvo especificação em contrário.
1. Preparação de reagentes e instrumentos
NOTA: Todas as etapas que envolvem soluções devem ser conduzidas dentro de um gabinete de risco biológico classe II.
<ol><li>Preparar o tampão de lavagem misturando solução salina tamponada com fosfato (PBS) com cálcio e magnésio, 10% de soro fetal bovino (FBS) e 1% de penicilina-estreptomicina (P/S) (ver Tabela de Materiais). Mantenha este tampão frio a 4 °C. É essencial prepará-lo fresco no dia do uso e desgaseificar o tampão.</li>
	<li>Preparar as enzimas digestivas combinando 10 mL de PBS (sem cálcio e magnésio) com 2 mL de tripsina a 0,25%, 1 mL de papaína 20 mg/mL e 200 μL de 1 mg/mL de colagenase I (ver Tabela de Materiais). NOTA: Garantir o uso de alíquotas de volumes apropriados para evitar ciclos repetidos de congelamento-descongelamento. Conservar estas alíquotas a -20 °C.</li>
	<li>Preparar o meio de cultura utilizando o kit de meio de crescimento de células endoteliais disponível comercialmente, que inclui VEGF-C (100 ng/mL) e 1% P/S (ver Tabela de Materiais).</li>
	<li>Prepare o tampão de parada suplementando o meio de Eagle modificado (DMEM) de Dulbecco com 10% de FBS para interromper o processo de digestão.</li>
	<li>Prepare instrumentos cirúrgicos estéreis: esses instrumentos devem incluir tesouras, pinças de ponta serrilhada e pinças de ponta fina.</li>
</ol>2. Revestimento do frasco
<ol><li>Cobrir o balão T25 com uma solução de fibronectina a 100 μg/ml (ver Tabela de Materiais) e incubar durante a noite a 37 °C. Antes de usar, remova a solução de revestimento usando uma pipeta.</li>
	<li>Lavar o frasco três vezes com PBS, aspirar posteriormente o PBS e deixar o frasco T25 secar ao ar.</li>
</ol>3. Dissociação das leptomeninges
NOTA: Utilize sempre tampões e soluções pré-arrefecidos a 4 °C.
<ol><li>Anestesiar os camundongos com inalação excessiva de isoflurano a 4% e, em seguida, sacrificá-los por decapitação rápida usando tesouras previamente limpas com etanol 70%.</li>
	<li>Inciso cuidadosamente a pele ao longo da linha média, começando pela abertura na parte de trás do crânio e estendendo-se em direção à área frontal.</li>
	<li>Remova delicadamente o crânio, levantando-o suavemente com a tesoura para evitar danificar as leptomeninges, garantindo que todo o cérebro, incluindo as leptomeninges, seja obtido.</li>
	<li>Submerja todo o cérebro em um tampão de lavagem e lave-o suavemente para remover o sangue da superfície.</li>
	<li>Transfira o cérebro para uma placa de Petri estéril sem cortar e extraia as leptomeninges da superfície do cérebro usando pinças de ponta fina sob um microscópio.</li>
	<li>Cortar o tecido das leptomeninges em fragmentos usando microtesouras estéreis.</li>
</ol>4. Digestão enzimática das leptomeninges
<ol><li>Adicionar 10 ml da mistura enzimática (passo 1.2) aos fragmentos e incubar a 37 °C durante 15 minutos. Agite suavemente para garantir que os fragmentos se desprendem do fundo do tubo. Depois, adicionar 10 ml de tampão de paragem (passo 1.4) para interromper a digestão.</li>
	<li>Centrifugar a 300 x g durante 5 minutos a 4 °C e remover cuidadosamente o sobrenadante utilizando uma pipeta.</li>
	<li>Adicione 10 mL de PBS frio e passe a mistura através de um filtro de 70 μm em um tubo estéril de 50 mL para filtrar quaisquer aglomerações.</li>
</ol>5. Expansão celular
<ol><li>Placa 1 x 10 5 células por cm 2 em frasco T25 revestido com fibronectina (passo2) com5 mL de meio de cultura.</li>
	<li>Incubar as células a 37 °C com 5% de CO2 durante 24 horas. Depois, remova o meio para eliminar as células não aderidas.</li>
	<li>Manter a cultura substituindo 50% do meio a cada dois dias. Repita esse processo duas ou três vezes.</li>
</ol>6. Seleção de células magnéticas
NOTA: Trabalhe rapidamente, mantenha a frieza da célula e utilize soluções pré-resfriadas para evitar a marcação celular não específica.
<ol><li>Preparação de instrumentos e reagentes: fixar um separador magnético a um suporte separador magnético (ver Tabela de Materiais). Conecte uma coluna de seleção ao separador magnético e posicione um filtro de células de 70 μm no topo da coluna de seleção. Coloque um tubo de 50 mL abaixo da coluna de seleção para coletar o fluxo.</li>
	<li>Digestão enzimática: uma vez que as células atinjam 80% de confluência, aspirar o meio e enxaguar as células com PBS. Adicionar tripsina a 0,25% para destacar as células aderentes e incubar a 37 °C por 5 min. Pare a digestão adicionando o buffer de parada. Centrifugar as células a 300 x g durante 5 min à temperatura ambiente e remover o sobrenadante.</li>
	<li>Incubação de anticorpos: ressuspender 1 x 107 células totais em 100 μL de PBS e adicionar 10 μL de anticorpo LYVE-1 (ver Tabela de Materiais). Misture bem e incube durante 30 minutos no escuro a 4 °C.<ol><li>Gire as células a 300 x g por 5 min e descarte o sobrenadante. Enxaguar as células introduzindo 1 mL de PBS e centrifugando a 300 x g por 5 min. Remova completamente o sobrenadante.</li>
	</ol></li>
	<li>Marcação de microesferas : ressuspender as células em 100 μL de PBS e adicionar 20 μL de Microesferas (ver Tabela de Materiais). Misture bem e incube durante 30 minutos no escuro a 4 °C.<ol><li>Gire as células a 300 x g por 5 min e decante o sobrenadante. Em seguida, limpar as células com 1 mL de PBS através de centrifugação a 300 x g por 5 min. Por fim, remova completamente o sobrenadante.</li>
	</ol></li>
	<li>Exclusão magnética negativa: ressuspender as células em 4 mL de PBS. Passe as células através de um filtro celular de 70 μm para eliminar aglomerações. Prepare a coluna de seleção (consulte Tabela de Materiais) enxaguando-a com 3 mL de PBS e, em seguida, aplique a suspensão de células na coluna de seleção.<ol><li>Execute as etapas de lavagem com 3 mL de PBS e colete as células negativas para LYVE-1 em um tubo de 50 mL posicionado abaixo da coluna de seleção para permitir a sua passagem.</li>
	</ol></li>
	<li>Seleção positiva magnética: pipetar 6 mL de PBS na coluna de seleção. Elimine instantaneamente as células marcadas magneticamente empurrando firmemente o êmbolo para dentro da coluna de seleção para obter LLECs positivos para LYVE-1. Observação : sempre aguarde até que o reservatório da coluna de seleção está vazio antes de prosseguir para a próxima etapa.</li>
</ol>7. Cultura de LLECs
<ol><li>Centrifugar os LLECs positivos para LYVE-1 a 300 x g por 5 minutos e, em seguida, remover cuidadosamente o sobrenadante.</li>
	<li>Placa 1 x 10 5 células por cm2 em frasco T25 revestido com fibronectina com5 mL de meio de cultura.</li>
	<li>Manter a cultura substituindo 50% do meio a cada dois dias. Quando as células atingirem 80% de confluência, desprender as células com 0,25% de tripsina e realizar a passagem celular. Utilizar as células para experimentos subsequentes após 2-3 passagens.</li>
</ol>8. Análise por citometria de fluxo
OBS: A análise por citometria de fluxo foi realizada seguindo o procedimento descritoanteriormente15.
<ol><li>Adicionar tripsina a 0,25% para destacar as células aderentes e incubar a 37 °C por 5 min. Em seguida, adicione o tampão de parada para interromper a digestão.</li>
	<li>Centrifugar as células a 300 x g por 5 min e aspirar completamente o sobrenadante. Ressuspender 1 x 106 células em 100 μL de PBS.</li>
	<li>Adicionar 1 μL do anticorpo LYVE-1. Misture bem e incube por 10 min no escuro à temperatura ambiente.</li>
	<li>Ressuspender as células em uma quantidade apropriada de tampão PBS para análise usando citometria de fluxo e software FlowJo (consulte Tabela de Materiais).</li>
</ol>9. Coloração por imunofluorescência
OBS: A coloração por imunofluorescência foi realizada seguindo o procedimento descrito anteriormente16.
<ol><li>Lave as células com PBS três vezes. Fixar as células com paraformaldeído (PFA) a 4% durante 10 minutos à temperatura ambiente. Descarte o PFA e lave as células com PBS três vezes.</li>
	<li>Aplique o tampão de bloco durante 1 h à temperatura ambiente. Remova o buffer de bloco das lamínulas.</li>
	<li>Adicione o anticorpo LYVE-1 no tampão de coloração às células e incube no escuro. Lave as células três vezes com PBS.</li>
	<li>Adicione um anticorpo secundário apropriado (ver Tabela de Materiais) no tampão de coloração às células e incube no escuro. Lave as células três vezes com PBS.</li>
	<li>Contracoloração com 4',6-diamidino-2-fenilindol (DAPI). As células já estão prontas para a microscopia de imunofluorescência.</li>

As recém-descobertas células endoteliais linfáticas leptomeníngeas (LLECs) formam uma malha de células individuais dentro das leptomeninges, exibindo um padrão de distribuição distinto quando comparadas às células endoteliais linfáticas periféricas 1,2. As funções celulares e as implicações clínicas associadas aos LLECs permanecem em grande parte território desconhecido. A fim de abrir caminho para a pesquisa funcional em LLECs, é imperativo estabelecer um modelo in vitro para o seu estudo. Portanto, este estudo elaborou um protocolo abrangente para o isolamento e cultura primária de LLECs.
Camundongos são o modelo animal preferido devido à sua adequação para manipulação genética em pesquisas de doenças. Estudos prévios isolaram com sucesso células endoteliais linfáticas de vários tecidos de camundongos, incluindo linfonodos3, tecido mesentérico4, tecido dérmico5, linfáticos coletores6 e tecido pulmonar7. Esses procedimentos de isolamento têm se baseado principalmente em técnicas como a classificação de células ativadas por magnetismo (MACS) e a classificação por citometria de fluxo 8,9,10. Além disso, esforços de pesquisa levaram ao estabelecimento de linhagens de células aracnoides de ratos e linhagens de células capilares linfáticas deratos11,12. Apesar da existência de técnicas de cultura de explantes para leptomeninges13, existe uma necessidade urgente de um protocolo padronizado para o isolamento e cultivo de LLECs. Consequentemente, este estudo colheu e cultivou com sucesso LLECs dissociando meticulosamente leptomeninges sob a orientação de um microscópio e promovendo a expansão de LLECs através do uso do fator de crescimento endotelial vascular-C (VEGF-C). O marcador distintivo das células endoteliais linfáticas é o receptor hialurônico dos vasos linfáticos-1 (LYVE-1)14. Este protocolo de várias etapas isola seletivamente LLECs LYVE-1-positivos usando MACS e, posteriormente, verifica sua pureza através de análise por citometria de fluxo e coloração por imunofluorescência.
As etapas primárias deste protocolo de várias etapas podem ser resumidas da seguinte forma: revestimento do frasco, dissociação de leptomeninges, digestão enzimática de leptomeninges, expansão celular, seleção de células magnéticas e subsequente cultura de LLECs. Finalmente, a pureza dos LLECs isolados é confirmada através de análise por citometria de fluxo e coloração por imunofluorescência. O objetivo geral deste estudo é apresentar um protocolo reprodutível em várias etapas para o isolamento de LLECs de leptomeninges de camundongos e seu subsequente cultivo in vitro . As células endoteliais linfáticas leptomeníngeas (LLECs) são uma população celular intracraniana recentemente descoberta com uma distribuição única claramente distinta das células endoteliais linfáticas periféricas. Sua função celular e implicações clínicas permanecem em grande parte desconhecidas. Consequentemente, a disponibilidade de um suprimento de LLECs é essencial para a realização de pesquisas funcionais in vitro. No entanto, atualmente não existe um protocolo para colheita e cultivo de LLECs in vitro.

The newly discovered leptomeningeal lymphatic endothelial cells (LLECs) form a meshwork of individual cells within the leptomeninges, exhibiting a distinct distribution pattern when compared to peripheral lymphatic endothelial cells1,2. The cellular functions and clinical implications associated with LLECs remain largely uncharted territory. In order to pave the way for functional research on LLECs, it is imperative to establish an in vitro model for their study. Therefore, this study has devised a comprehensive protocol for the isolation and primary culture of LLECs.
Mice are the preferred animal model due to their suitability for genetic manipulation in disease research. Previous studies have successfully isolated lymphatic endothelial cells from various mouse tissues, including lymph nodes3, mesenteric tissue4, dermal tissue5, collecting lymphatics6, and lung tissue7. These isolation procedures have primarily relied on techniques such as magnetic-activated cell sorting (MACS) and flow cytometry sorting8,9,10. Additionally, research efforts have led to the establishment of rat arachnoid cell lines and rat lymphatic capillary cell lines11,12. Despite the existence of explant culture techniques for leptomeninges13, there exists an urgent need for a standardized protocol for the isolation and culture of LLECs. Consequently, this study has successfully harvested and cultured LLECs by meticulously dissociating leptomeninges under the guidance of a microscope and promoting LLECs expansion through the use of vascular endothelial growth factor-C (VEGF-C). The distinctive marker for lymphatic endothelial cells is lymphatic vessel hyaluronic receptor-1 (LYVE-1)14. This multi-step protocol selectively isolates LYVE-1-positive LLECs using MACS and subsequently verifies their purity through flow cytometric analysis and immunofluorescent staining.
The primary steps of this multi-step protocol can be summarized as follows: flask coating, dissociation of leptomeninges, enzymatic digestion of leptomeninges, cell expansion, magnetic cell selection, and subsequent culture of LLECs. Finally, the purity of the isolated LLECs is confirmed through flow cytometric analysis and immunofluorescent staining. The overarching aim of this study is to present a reproducible, multi-step protocol for the isolation of LLECs from mouse leptomeninges and their subsequent in vitro culture. This protocol is poised to greatly facilitate investigations into the cellular functions and clinical implications of LLECs.

Leptomeningeal lymphatic endothelial cells (LLECs) are a recently discovered intracranial cellular population with a unique distribution clearly distinct from peripheral lymphatic endothelial cells. Their cellular function and clinical implications remain largely unknown. Consequently, the availability of a supply of LLECs is essential for conducting functional research in vitro. However, there is currently no existing protocol for harvesting and culturing LLECs in vitro.
This study successfully harvested LLECs using a multi-step protocol, which included coating the flask with fibronectin, dissecting the leptomeninges with the assistance of a microscope, enzymatically digesting the leptomeninges to prepare a single-cell suspension, inducing the expansion of LLECs with vascular endothelial growth factor-C (VEGF-C), and selecting lymphatic vessel hyaluronic receptor-1 (LYVE-1) positive cells through magnetic-activated cell sorting (MACS). This process ultimately led to the establishment of a primary culture. The purity of the LLECs was confirmed through immunofluorescence staining and flow cytometric analysis, with a purity level exceeding 95%. This multi-step protocol has demonstrated reproducibility and feasibility, which will greatly facilitate the exploration of the cellular function and clinical implications of LLECs.

Mouse Leptomeninges Dissociation and Enzymatic Digestion for Efficient Culturing of Leptomeningeal Cells

To begin, carefully incise the skin of the sacrificed mouse, starting from the skull's opening and extending towards the frontal area. Using scissors, delicately lift and remove the skull without damaging the leptomeninges, collecting the entire brain. Submerge the whole brain in the washing buffer, and gently flush it to remove surface blood.Transfer the brain into a sterile Petri dish without chopping. Then, under a microscope, use fine-point tweezers to extract the leptomeninges from the brain's surface. Using sterile microscissors, cut the leptomeninges tissue into fragments.Repair the digestive enzyme mix using the given components. Add 10 milliliters of mix to the fragments and incubate at 37 degrees Celsius for 15 minutes. Gently agitate to detach the fragments from the bottom of the tube.Then, add 10 milliliters of stopping buffer. Centrifuge the suspension at 300 g for five minutes at four degrees Celsius. And using a pipette, carefully remove the supernatant.Add 10 milliliters of cold PBS, and filter any clumps through a 70 micron strainer into a sterile 50 milliliter tube. Plate 10 to the fifth cells per square centimeter into a fibronectin-coated T25 flask with five milliliters of culture medium, and incubate at 37 degrees Celsius with 5%carbon dioxide for 24 hours. After incubation, replace the medium with a fresh medium to eliminate non-attached cells.

Establishing Primary In Vitro Culture of Leptomeningeal Lymphatic Endothelial Cells (LLECs)

Begin by attaching a magnetic separator to its stand. Connect a selection column to the magnetic separator and place a 70 micron cell strainer on top of the selection column. Place a 50 milliliter tube beneath the selection column to collect the flow.Once the mouse brain cells reach 80%confluency, aspirate the medium and rinse the cells with PBS. Add 0.25%trypsin to detach the adherence cells and incubate at 37 degrees Celsius for five minutes. Then add a stopping buffer.Centrifuge the cell suspension at 300 G for five minutes, and remove the supernatant. For antibody staining, resuspend 10 to the seven cells in 100 microliters of PBS. Then add 10 microliters of LYVE-1 antibody solution and mix thoroughly.Incubate the mixture for 30 minutes in the dark at four degrees Celsius. After incubation, centrifuge the cells and discard the supernatant. Then rinse the cells by adding one milliliter of PBS and centrifuging at 300 G for five minutes.For microbead labeling, resuspend the pellet in 100 microliters of PBS and 20 microliters of microbeads. Make swell and incubate for 30 minutes in the dark at four degrees Celsius. Then centrifuge the suspension and wash the pellet with one milliliter of PBS.Again, centrifuge and resuspend the pellet in four milliliters of PBS for magnetic negative exclusion. Pass the cell suspension through a 70 micron strainer to eliminate clumps. Prepare the selection column by rinsing it with three milliliters of PBS.Then add cell suspension into the selection column. Wash the column with three milliliters of PBS and collect LYVE-1 negative cells into a 50 milliliter tube. For magnetic positive selection, pipette six milliliters of PBS into the selection column.Then flush the magnetically labeled cells by firmly pushing the plunger into the selection column to obtain LYVE-1 positive LLECs. Next, centrifuge the positive cell suspension and remove the supernatant. Plate 10 to the fifth cells per square centimeter into a T25 flask with five milliliters of culture medium.Maintain the culture by replacing 50%of the medium every alternate day. When the cells reach 80%confluency, detach them as demonstrated previously before performing cell passage. Flow cytometry analysis revealed that the percentage of LYVE-1 positive cells in passage two was not significantly different from passage three after MACS, indicating a purity greater than 95%Immunofluorescent staining confirmed co-staining of LYVE-1 with PDPN, VEGFR-3 and PROX1, establishing the identity of LLECs.LYVE-1 positive cells did not express F48 and platelet derived growth factor beta, effectively distinguishing LLECs from macrophages and fibroblasts. Leptomeningeal cells before MACS displayed heterogeneous morphology ranging from round spheres to fused fiber shapes. However, post MACS, LLECs exhibited typical endothelial-like features such as spindle and cobblestone shapes.
