Alle Verfahren müssen mit geeigneter persönlicher Schutzausrüstung durchgeführt werden.
1. Herstellung der TCPP-Stammlösung, 1,0 mg/ml
HINWEIS: Dies kann monatlich erstellt werden.
<ol><li>Wiegen Sie mit einer Analysenwaage, einem Spatel und einem Wiegepapier 49,0 bis 50,9 mg TCPP ab. Runden Sie das Gewicht auf 1/10 Milligramm. Stellen Sie die gemessene Menge an TCPP lichtgeschützt beiseite. Anmerkungen: Verwenden Sie eine statische Pistole, wenn die Gewichtsanzeige instabil ist.</li>
	<li>Bestimmen Sie die erforderlichen Mengen an gereinigtem Wasser und Isopropanol (IPA) aus Tabelle 1 auf der Grundlage der in Schritt 1.1 gewogenen TCPP-Menge. Geben Sie das gereinigte Wasser und Isopropanol in ein 100-ml-Glasbecherglas und bedecken Sie es mit Parafilm, um es vor Verdunstung zu schützen.</li>
	<li>Bestimmen Sie die erforderliche Menge an Natriumbicarbonat aus Tabelle 1 auf der Grundlage der in Schritt 1.1 gewogenen TCPP-Menge.</li>
	<li>Wiegen Sie mit der Analysenwaage, dem Spatel und dem Wiegepapier die in Schritt 1.3 ermittelte erforderliche Menge an Natriumbicarbonat ab. Runden Sie das Gewicht auf 1/10 Milligramm. Anmerkungen: Verwenden Sie eine statische Pistole, wenn die Gewichtsanzeige instabil ist.</li>
	<li>Geben Sie das Natriumbicarbonat in das 100-ml-Becherglas mit gereinigtem Wasser und Isopropanol. Decken Sie die Lösung mit Parafilm ab, um sie vor Verdunstung zu schützen.</li>
	<li>Geben Sie die Lösung aus Schritt 1.5 auf eine Rührplatte und rühren Sie, bis sie sich aufgelöst hat (ca. 10 min)</li>
	<li>Messen Sie den pH-Wert, um sicherzustellen, dass die natriumbicarbonathaltige Lösung aus Schritt 1.6 einen pH-Wert zwischen 9 und 10 hat.</li>
	<li>Fügen Sie das in Schritt 1.1 gewogene TCPP langsam zur Lösung aus Schritt 1.7 hinzu und rühren Sie weiter, bis es sich aufgelöst hat (~30 min). Schützen Sie sich während dieses Schritts vor Licht.</li>
	<li>In einem Glas- oder Polypropylenbehälter bei Raumtemperatur und lichtgeschützt aufbewahren.</li>
</ol>2. Herstellung von 2-(N-Morpholino )-ethansulfonsäure (MES) und Heminatriumsalz-Pufferlösung, 0,1 M, pH 6,0-6,2 ("MES-Puffer")
Anmerkungen: Dies muss am Tag der Verwendung zubereitet und bei Raumtemperatur aufbewahrt werden.
<ol><li>Wiegen Sie 2,50 g MES-Heminatriumsalz ab und geben Sie es in eine 150-ml-Plastikflasche.</li>
	<li>Fügen Sie 121 ml gereinigtes Wasser hinzu und lösen Sie es durch manuelles Schütteln auf, bis kein Feststoff mehr sichtbar ist.</li>
	<li>Messen Sie den pH-Wert des MES-Puffers, um sicherzustellen, dass er zwischen 6 und 6,2 liegt.</li>
	<li>Bei Raumtemperatur aufbewahren, um am selben Tag verwendet zu werden.</li>
</ol>3. N-(3-Dimethlyaminopropyl)-N'-ethylcarbodiimid (EDC)-Pulver
<ol><li>Nehmen Sie das EDC-Pulver aus dem Gefrierschrank und lassen Sie es bis zur Verwendung in Schritt 5 bei Raumtemperatur ruhen.</li>
</ol>4. Kombination von Amin-funktionalisierten Polystyrolkügelchen mit TCPP-Lösung
<ol><li>Geben Sie 4,3 ml der in Schritt 2 hergestellten 0,1 m MES-Pufferlösung in ein 15-ml-Polypropylenröhrchen.</li>
	<li>Wirbeln Sie die Amin-funktionalisierte Polystyrol-Kügelchensuspension (10 μm, 2,5 % w/v) für 60 s bei maximaler Geschwindigkeit.</li>
	<li>Geben Sie 288 μl dieser frisch gewirbelten Perlensuspension in den MES-Puffer aus Schritt 4.1.</li>
	<li>Wirbeln Sie die MES/Wulst-Lösung 15 s lang bei maximaler Geschwindigkeit.</li>
	<li>Die in Schritt 1 hergestellte 1 mg/ml TCPP-Lösung wird 60 s lang bei maximaler Geschwindigkeit aufgewirbelt.</li>
	<li>Geben Sie 1,20 ml dieser frisch vortexed TCPP-Stammlösung in die MES/Bead-Suspension aus Schritt 4.4.</li>
	<li>Wirbeln Sie die MES-/Wulst-/TCPP-Aufhängung 15 s lang bei maximaler Geschwindigkeit auf.</li>
	<li>Decken Sie das Röhrchen mit Folie ab, während die EDC-Lösung zubereitet wird.</li>
</ol>5. Herstellung von N-(3-Dimethlyaminopropyl)-N'-ethylcarbodiimid (EDC)-hydrocholorid (HCl) Stammlösung
Anmerkungen: Die EDC-Lösung ist verderblich und sollte sofort nach der Zubereitung verwendet werden.
<ol><li>Geben Sie 20,0 ml gereinigtes Wasser in ein neues konisches 50-ml-Röhrchen.</li>
	<li>Wiegen Sie 200 mg EDC HCl aus Schritt 3 ab und geben Sie es in das Wasser (Schritt 5.1).</li>
	<li>Wirbeln Sie die EDC HCL 15 s lang mit maximaler Geschwindigkeit auf, um eine klare Lösung zu erzeugen.</li>
</ol>6. Vorbereitung der EDC HCl/MES-Arbeitslösung
HINWEIS: Die EDC HCl/MES-Lösung ist verderblich und sollte sofort nach der Zubereitung verwendet werden.
<ol><li>Geben Sie 54,0 ml MES-Pufferlösung (hergestellt in Schritt 2) in eine 150-ml-Plastikflasche.</li>
	<li>6,0 ml der EDC-HCl-Stammlösung (hergestellt in Schritt 5) in die MES-Pufferlösung geben und 10 s lang unter Schütteln mischen.</li>
</ol>7. Beschriftung der Perlen mit TCPP
<ol><li>Geben Sie 4,5 ml EDC-Arbeitslösung (aus Schritt 6) in das 15-ml-Polypropylenröhrchen, das die Perlen und TCPP in MES-Puffer enthält (Schritt 4.7).</li>
	<li>Legen Sie das Rohr für 16 h bei Raumtemperatur und lichtgeschützt in einen Umkehrrotator bei 35 U/min.</li>
	<li>Zentrifugieren Sie das Röhrchen bei Raumtemperatur für 10 min bei 1.000 × g.</li>
	<li>Aspirieren Sie den Überstand und resuspendieren Sie die Beads in 0,8 ml Hanks' ausgewogener Salzlösung (HBSS).</li>
	<li>Übertragen Sie die Perlenlösung mit einer 1-ml-Pipette in eine durchstechflasche aus braunem Polypropylen und lagern Sie sie bis zur weiteren Verwendung bei 4 °C. HINWEIS: Die Perlen sind zu diesem Zeitpunkt mindestens 3 Monate haltbar.</li>
</ol>8. Qualitätskontrolle (QC) der TCPP-Beads mittels Durchflusszytometrie
HINWEIS: Die Qualitätskontrolle sollte sich darauf konzentrieren, ob die mittlere Fluoreszenzintensität (MFI) der TCPP-Kügelchen für ihren Verwendungszweck hell genug ist und wie viel Partikel durch das Verfahren erzeugt werden. Weitere Informationen finden Sie im Abschnitt "Repräsentative Ergebnisse".
<ol><li>Beschriften Sie ein 5-ml-Styroporröhrchen als "TCPP-negative Beads". HINWEIS: Die negativen Beads unterscheiden sich von den Amin-funktionalisierten Beads, die für die Beschriftung verwendet werden. Siehe Materialtabelle.</li>
	<li>Beschriften Sie ein anderes Röhrchen als "TCPP-positive Beads".</li>
	<li>Beschrifte eine andere Tube mit "Regenbogenperlen".</li>
	<li>Aliquotieren Sie 300 μl eiskaltes HBSS in die Röhrchen, die mit "TCPP negative Beads" und "TCPP positive Beads" gekennzeichnet sind.</li>
	<li>Geben Sie 500 μl eiskaltes HBSS in das Röhrchen mit der Bezeichnung "Regenbogenperlen".</li>
	<li>Die nicht funktionalisierte Polystyrol-Bead-Suspension (unmarkiert) kurz mit maximaler Geschwindigkeit (2-3 s) aufwirbeln und 10 μL davon in das Röhrchen mit der Aufschrift "TCPP negative Beads" geben.</li>
	<li>Die TCPP-markierte Bead-Suspension (finalisiert in Schritt 7.5) kurz mit maximaler Geschwindigkeit aufwirbeln und 3 μL davon in das "TCPP positive beads"-Röhrchen geben.</li>
	<li>Wirbeln Sie die Rainbow-Perlenaufhängung kurz mit maximaler Geschwindigkeit und geben Sie zwei Tropfen davon in das "Rainbow Beads"-Röhrchen.</li>
	<li>Bewahren Sie alle Röhren abgedeckt und lichtgeschützt auf Eis auf.</li>
	<li>Leiten Sie die entsprechenden täglichen Startvorgänge des Durchflusszytometers ein und führen Sie eine Qualitätskontrolle durch, um die optimale Flüssigkeits- und Laserausrichtung zu überprüfen. HINWEIS: Für diesen Teil des Protokolls wird davon ausgegangen, dass der Bediener in der Verwendung des verfügbaren Durchflusszytometers geschult ist, einschließlich der Verfahren zur Standardisierung der Lichtstreuung und Fluoreszenzintensität sowie der Grundprinzipien zur Berechnung der korrekten Kompensationsmatrix.</li>
	<li>Führen Sie die Rainbow-Perlen und die TCPP-Perlen aus, ohne die Spannungseinstellungen zwischen den verschiedenen Durchläufen zu ändern.<ol><li>Laufe und sammle 10.000 Events der Regenbogenperlen.</li>
		<li>Führen Sie eine Spülung mit Wasser durch und sammeln Sie 10.000 Ereignisse der TCPP-negativen Beads.</li>
		<li>Führen Sie eine Spülung mit Wasser durch und sammeln Sie 10.000 Ereignisse der TCPP-positiven Beads.</li>
		<li>Führen Sie eine 1-minütige Wasserspülung durch. Anmerkungen: Es ist wichtig, nach dem Ausführen der TCPP-Perlen eine Spülung mit Wasser durchzuführen. Wenn das TCPP nicht aus den Leitungen im Zytometer gespült wird, besteht die Möglichkeit, dass verbleibendes TCPP Zellen im nächsten zu erfassenden Röhrchen markieren kann.</li>
		<li>Führen Sie die entsprechenden Reinigungs- und Abschaltprotokolle durch, die für die Anweisungen des Herstellers für das Zytometer spezifisch sind. HINWEIS: Repräsentative Ergebnisse finden Sie in Abbildung 1.</li>
	</ol></li>
</ol>9. Perlenfiltration
HINWEIS: Wenn die Qualitätskontrolle der Beads durch Durchflusszytometrie (Schritt 8) einen hohen Anteil an Partikeln (70 % oder mehr) zeigt, sollten Sie die Beadsuspension mit dem folgenden Protokoll filtern (Abbildung 2).
<ol><li>Fügen Sie 3,20 ml eiskaltes HBSS zu 0,8 ml der in Schritt 7.5 abgeschlossenen TCPP-Perlensuspension hinzu (wodurch eine fünffache Verdünnung entsteht).</li>
	<li>Wirbeln Sie die verdünnte Wulstsuspension 15 s lang bei maximaler Geschwindigkeit vor.</li>
	<li>Entfernen Sie den Kolben aus einer 5-ml-Einwegspritze.</li>
	<li>Setzen Sie die Spritze mit einem Glasfaserfilter (5 μm, 13 mm Durchmesser) ein.</li>
	<li>Geben Sie 4 ml HBSS in die Spritze.</li>
	<li>Fügen Sie 0,5 ml der vortexed verdünnten Perlensuspension hinzu (Schritt 9.2).</li>
	<li>Verwenden Sie den Kolben, um die Suspension mit ca. 2 Tropfen/s durch die Spritze/Filtereinrichtung zu filtern.</li>
	<li>Waschen Sie die Kügelchen, indem Sie 5 ml frisches HBSS mit ca. 2 Tropfen/s durch den Filter in die Spritze ziehen.</li>
	<li>Schieben Sie das HBSS mit ca. 2 Tropfen/s wieder in den Abfallbehälter.</li>
	<li>Um die Perlen aus dem Filter zu entfernen, ziehen Sie weitere 5 ml frisches HBSS durch den Filter in die Spritze.</li>
	<li>Entfernen Sie vorsichtig den Filter aus der Spritze.</li>
	<li>Werfen Sie die Perlensuspension aus der Spritze in ein konisches 50-ml-Zentrifugenröhrchen.</li>
	<li>Setzen Sie den Filter wieder auf die Spritze und wiederholen Sie die Schritte 9.10-9.12 noch viermal. Entsorgen Sie dann den Filter und die Spritze.</li>
	<li>Wiederholen Sie die Schritte 9.2-9.13, bis alle Perlen aus Schritt 9.1 gefiltert sind. Verwenden Sie jedes Mal eine frische Spritze und filtern Sie.</li>
	<li>Die filtrierten Kügelchensuspensionen werden 10 min bei 1.000 × g bei Raumtemperatur zentrifugiert.</li>
	<li>Aspirieren Sie den Überstand jedes 50-ml-Röhrchens und resuspendieren Sie die Kügelchen vorsichtig, indem Sie sie in 0,5 ml frischem HBSS kombinieren.</li>
	<li>Übertragen Sie die Kügelchen mit einer p1.000-Mikropipette in ein neues Durchstechflasche aus Braun-, Glas- oder Polypropylen und lagern Sie sie bei 4 °C.</li>
	<li>Wiederholen Sie Schritt 8, um festzustellen, ob der Anteil der TCPP-bezogenen Partikel in der gefilterten Perlensuspension abgenommen hat. Repräsentative Ergebnisse finden Sie in Abbildung 3.</li>

Trotz der zahlreichen Anwendungen von Porphyrinen in der Krebsdiagnostik und -therapie2 gibt es nur begrenzte Literatur über ihre potenzielle Verwendung als durchflusszytometrisches Reagenz zur Identifizierung von krebsartigen und nicht-krebsartigen Zellpopulationen in primären menschlichen Geweben24,25,26. Porphyrine sind aufgrund ihrer Fluoreszenz- und Tumor-Targeting-Eigenschaften seit vielen Jahren im biomedizinischen Bereich von Interesse 1,2,3. Therapeutische Anwendungen wie die photodynamische Therapie (PDT) und die sonodynamische Therapie (SDT) beinhalten die systemische Verabreichung eines Porphyrins an einen Krebspatienten, die Akkumulation des Medikaments im Tumor und die lokale Exposition des Tumors mit einem Laserlicht einer bestimmten Wellenlänge oder Ultraschall. Die Bestrahlung mit Laserlicht oder Ultraschall führt zur Bildung reaktiver Sauerstoffspezies durch das Porphyrin und in der Folge zum Zelltod 4,5. In der photodynamischen Diagnostik (PDD) wird die Porphyrinfluoreszenz verwendet, um Krebszellen von normalen Zellen zu unterscheiden6. In diesem Zusammenhang wird Protoporphyrin IX, ein natürliches fluoreszierendes Porphyrin, das sich bei systemischer oder lokaler Injektion seiner Vorstufe, der 5-Aminolävulinsäure (5-ALA), in Tumoren anreichert, zur Identifizierung von gastrointestinalen Stromatumoren, Blasenkrebs und Hirntumoren verwendet 7,8. In jüngerer Zeit wurde die 5-ALA-Behandlung als Ansatz zur Erkennung einer minimalen Resterkrankung beim Multiplen Myelomuntersucht 9. Unser Labor verwendet das Tetraarylporphyrin TCPP (5,10,15,20-tetrakis-(4-carboxyphenyl)-21,23 H-porphin) wegen seiner Fähigkeit, selektiv Lungenkrebszellen und krebsassoziierte Zellen in menschlichen Sputumproben zu färben, eine Eigenschaft, die in objektträgerbasierten und durchflusszytometrischen diagnostischen Assays ausgenutzt wurde10.
Einige Porphyrine sind insofern bifunktional, als sie als therapeutische und diagnostische Mittel eingesetzt werden können 2,11. In der biomedizinischen Forschung werden solche bifunktionalen Porphyrine verwendet, um zu bewerten, wie ihre Fähigkeit, Krebszellen selektiv anzugreifen und abzutöten, von ihrer Struktur abhängt und wie sie durch das Vorhandensein anderer Verbindungen beeinflusst wird 12,13,14,15,16. Sowohl die zelluläre Aufnahme von Porphyrinen als auch ihre Zytotoxizität können auf einer durchflusszytometrischen Plattform im Hochdurchsatz gemessen werden. Die Absorptions- und Emissionsspektren von fluoreszierenden Porphyrinen sind komplex, aber die meisten durchflusszytometrischen Plattformen sind in der Lage, sie korrekt zu identifizieren. Das Absorptionsspektrum fluoreszierender Porphyrine zeichnet sich durch eine starke Absorptionsbande im Bereich von 380-500 nm aus, die als Soret-Bande bezeichnet wird. Zwei bis vier schwächere Absorptionsbanden werden im Allgemeinen im Bereich von 500-750 nm (Q-Banden) beobachtet17. Ein blauer 488-nm-Laser, der in den meisten Durchflusszytometern vorhanden ist, oder ein violetter Laser (405 nm) können Licht der entsprechenden Wellenlänge erzeugen, um Porphyrine anzuregen. Die Emissionsspektren von Porphyrinen zeigen typischerweise Spitzen im Bereich von 600-800 nm18, was zu einer sehr geringen spektralen Überlappung mit Fluoresceinisothiocyanat- oder Phycoerythrin (PE)-Fluorophoren, aber zu einer erheblichen Überlappung mit anderen häufig verwendeten Fluorophoren, wie Allophycocyanin (APC), sowie Tandem-Fluorophoren, wie PE-Cy5 und anderen, führt. Daher sind bei der Verwendung von Porphyrinen in mehrfarbigen Durchflusszytometrie-Assays Einzelfluorophorkontrollen unerlässlich, um den Spillover der Fluoreszenz in anderen Kanälen als dem, der für die Messung der Fluoreszenz des Porphyrins vorgesehen ist, angemessen zu korrigieren.
Idealerweise sollten die Einzelfluorophor-Kontrollen, die zur Berechnung der Spillover-Matrix für ein Panel von Fluorophoren verwendet werden (auch "Kompensationskontrollen" genannt), aus denselben Zelltypen wie die Probe bestehen. Die Verwendung der Probe für diesen Zweck ist jedoch nicht optimal, wenn es zunächst nur sehr wenige Proben gibt oder wenn die Zielpopulation innerhalb der Probe sehr klein ist (z. B. wenn man minimale Resterkrankungen oder Krebszellen in frühen Stadien der Krankheit betrachten möchte). Eine nützliche Alternative zu Zellen sind Beads, die mit demselben Fluorophor gekoppelt sind, der zur Analyse der Probe verwendet wird. Viele dieser Perlen sind im Handel erhältlich; Diese Beads sind entweder mit dem gewünschten Fluorophor vormarkiert (vormarkierte Fluorophor-spezifische Beads)19,20, oder es kann ein fluoreszenzmarkierter Antikörper an sie angehängt werden (Antibody Capture Beads)20,21. Während für viele Fluorophore kommerzielle Kompensationskügelchen zur Verfügung stehen, sind solche Beads für Porphyrine trotz ihres zunehmenden Einsatzes in der Grundlagen- und klinischen Forschung nicht verfügbar.
Neben der Probenkonservierung und der angemessenen Größe positiver und negativer Populationen sind die weiteren Vorteile der Verwendung von Beads als Kompensationskontrollen die einfache Präparation, die geringe Hintergrundfluoreszenz und die ausgezeichnete Stabilität über die Zeit22. Der potenzielle Nachteil der Verwendung von Beads als Kompensationskontrolle besteht darin, dass sich das Emissionsspektrum des fluoreszierenden Antikörpers, der auf Beads eingefangen wird, von dem desselben Antikörpers unterscheiden kann, der zur Markierung der Zellen verwendet wird. Dies kann bei Verwendung eines spektralen Durchflusszytometers20 von besonderer Bedeutung sein. Daher muss die Entwicklung von Beads als Kompensationskontrolle auf dem Durchflusszytometer durchgeführt werden, das für den Assay verwendet wird, für den die Beads entwickelt werden. Darüber hinaus muss die Entwicklung der Kügelchen einen Vergleich mit Zellen beinhalten, die mit demselben fluoreszierenden Färbereagenz markiert sind.
In dieser Arbeit beschreiben wir die Herstellung von TCPP-Amin-funktionalisierten Polystyrol-Kompensationsbeads, deren mediane Fluoreszenzintensität im Detektionskanal mit der von TCPP-markierten Zellen im Sputum vergleichbar war, und deren Verwendung als Kompensationskontrollen für die Durchflusszytometrie. Die Autofluoreszenz von äquivalenten, nicht-funktionalisierten Beads war niedrig genug, um sie als negative Fluoreszenzkompensationskontrollen zu verwenden. Darüber hinaus zeigten diese Perlen eine Lagerstabilität von fast 1 Jahr.

porphyrin-modified beads for use as compensation controls in flow cytometry

Die Durchflusszytometrie kann verschiedene Zellpopulationen auf der Grundlage von Fluoreszenzmessungen schnell charakterisieren und quantifizieren. Die Zellen werden zunächst mit einem oder mehreren fluoreszierenden Reagenzien gefärbt, die jeweils mit einem anderen fluoreszierenden Molekül (Fluorophor) funktionalisiert werden, das aufgrund ihrer phänotypischen Eigenschaften, wie z. B. der Antigenexpression auf der Zelloberfläche, selektiv an Zellen bindet. Die Intensität der Fluoreszenz von jedem Reagenz, das an Zellen gebunden ist, kann auf dem Durchflusszytometer mit Kanälen gemessen werden, die einen bestimmten Wellenlängenbereich erfassen. Wenn mehrere Fluorophore verwendet werden, schwappt das Licht einzelner Fluorophore oft in unerwünschte Detektionskanäle über, was eine Korrektur der Fluoreszenzintensitätsdaten in einem Prozess erfordert, der als Kompensation bezeichnet wird. 
Kompensationskontrollpartikel, in der Regel Polymerkügelchen, die an einen einzelnen Fluorophor gebunden sind, werden für jeden Fluorophor benötigt, der in einem Zellmarkierungsexperiment verwendet wird. Daten von Kompensationspartikeln aus dem Durchflusszytometer werden verwendet, um eine Korrektur der Fluoreszenzintensitätsmessungen vorzunehmen. Dieses Protokoll beschreibt die Herstellung und Aufreinigung von Polystyrol-Kompensationskügelchen, die kovalent mit dem fluoreszierenden Reagenz Meso-Tetra(4-carboxyphenyl)porphin (TCPP) funktionalisiert sind, und deren Anwendung in der Durchflusszytometrie-Kompensation. In dieser Arbeit wurden aminfunktionalisierte Polystyrolkügelchen mit TCPP und dem Amid-Kupplungsreagenz EDC (N-(3-Dimethylaminopropyl)-N′-ethylcarbodiimidhydrochlorid) bei pH 6 und bei Raumtemperatur für 16 h unter Rühren behandelt. Porphyrins have been of interest for many years in the biomedical field owing to their fluorescence and tumor-targeting properties1,2,3. Therapeutic applications such as photodynamic therapy (PDT) and sonodynamic therapy (SDT) entail the systemic administration of a porphyrin to a cancer patient, the accumulation of the drug in the tumor, and the localized exposure of the tumor to a laser light of a specific wavelength or ultrasound. The exposure to laser light or ultrasound leads to the generation of reactive oxygen species by the porphyrin and subsequent cell death4,5. In photodynamic diagnosis (PDD), porphyrin fluorescence is used to distinguish cancer cells from normal cells6. In this context, protoporphyrin IX, a natural fluorescent porphyrin that accumulates in tumors upon the systemic or local injection of its precursor, 5-aminolevulinic acid (5-ALA), is used to identify gastrointestinal stromal tumors, bladder cancer, and brain cancer7,8. More recently, 5-ALA treatment was explored as an approach to detect minimal residual disease in multiple myeloma9. Our laboratory has been using the tetraaryl porphyrin TCPP (5,10,15,20-tetrakis-(4-carboxyphenyl)-21,23H-porphine) for its ability to selectively stain lung cancer cells and cancer-associated cells in human sputum samples, which is a property that has been exploited in slide-based and flow cytometric diagnostic assays10.
Some porphyrins are bifunctional in that they can be used as therapeutic and diagnostic agents2,11. In biomedical research, such bifunctional porphyrins are used to evaluate how their ability to selectively target and kill cancer cells is a function of their structure as well as how it is affected by the presence of other compounds12,13,14,15,16. Both the cellular uptake of porphyrins and their cytotoxicity can be measured on a flow cytometric platform in a high-throughput manner. The absorption and emission spectra of fluorescent porphyrins are complex, but most flow cytometric platforms are equipped to correctly identify them. The absorption spectrum of fluorescent porphyrins is characterized by a strong absorption band in the 380-500 nm range, known as the Soret band. Two to four weaker absorption bands are generally observed in the 500-750 nm range (Q bands)17. A blue&#160;488 nm laser, present in most flow cytometers, or a violet laser (405 nm) can generate light of the appropriate wavelength to excite porphyrins. The emission spectra of porphyrins typically display peaks in the 600-800 nm range18, which results in very little spectral overlap with fluorescein isothiocyanate or phycoerythrin (PE) fluorophores but considerable overlap with other often-used fluorophores, such as allophycocyanin (APC), as well as tandem fluorophores, such as PE-Cy5 and others. Therefore, when using porphyrins in multi-color flow cytometry assays, single-fluorophore controls are essential to adequately correct the spillover of fluorescence in channels other than the one designated to measure the porphyrin&#39;s fluorescence.
Ideally, the single-fluorophore controls used to calculate the spillover matrix for a panel of fluorophores (also called &#34;compensation controls&#34;) should consist of the same cell type(s) as the sample. However, using the sample for this purpose is not optimal if there is very little sample to begin with or if the target population within the sample is very small&#160;(for example, if one wants to look at minimal residual disease or cancer cells at the early stages of the disease). A useful alternative to cells is beads coupled with the same fluorophore that is used to analyze the sample. Many such beads are commercially available; these beads are either prelabeled with the desired fluorophore (prelabeled fluorophore-specific beads)19,20, or a fluorescently labeled antibody can be attached to them (antibody capture beads)20,21. While commercial compensation beads are available for many fluorophores, such beads are unavailable for porphyrins, despite their increasing use in basic and clinical research.
In addition to sample preservation and appropriately sized positive versus negative populations, the other advantages of using beads as compensation controls are the ease of preparation, low background fluorescence, and excellent stability over time22. The potential disadvantage of using beads as a compensation control is that the emission spectrum of the fluorescent antibody captured on beads may differ from that of the same antibody used to label the cells. This may be of specific importance when using a spectral flow cytometer20. Therefore, the development of beads as a compensation control needs to be performed on the flow cytometer that will be used for the assay for which the beads are developed. Moreover, the development of the beads needs to include a comparison with cells labeled with the same fluorescent staining reagent.
Here, we describe the preparation of TCPP amine-functionalized polystyrene compensation beads, whose median fluorescence intensity in the detection channel was comparable to that of TCPP-labeled cells in sputum, and their use as compensation controls for flow cytometry. The autofluorescence of equivalent, non-functionalized beads was sufficiently low for their use as negative fluorescence compensation controls. In addition, these beads demonstrated stability in storage for nearly 1 year.

We developed polystyrene beads modified with the porphyrin TCPP as compensation controls and multicolor flow cytometry. The beads used in this protocol have fluorescence intensity comparable to TCPP stain cells and still passed through the flow cytometer. We accomplished that and are currently working on better methods to remove the particulate byproduct.Porphyrin-modified compensation beads were preferable to TCPP stained cells as compensation controls. These speeds were stable over months without significant loss of fluorescence. Porphyrins are known to stain cancer cells selectively.The research combines the selectivity of porphyrins with quantitative flow cytometry as a powerful platform for high-throughput analysis of human samples.

Flow cytometry can rapidly characterize and quantify diverse cell populations based on fluorescence measurements. The cells are first stained with one or more fluorescent reagents, each functionalized with a different fluorescent molecule (fluorophore) that binds to cells selectively based on their phenotypic characteristics, such as cell surface antigen expression. The intensity of fluorescence from each reagent bound to cells can be measured on the flow cytometer using channels that detect a specified range of wavelengths. When multiple fluorophores are used, the light from individual fluorophores often spills over into undesired detection channels, which requires a correction to the fluorescence intensity data in a process called compensation. 
Compensation control particles, typically polymer beads bound to a single fluorophore, are needed for each fluorophore used in a cell labeling experiment. Data from compensation particles from the flow cytometer are used to apply a correction to the fluorescence intensity measurements. This protocol describes the preparation and purification of polystyrene compensation beads covalently functionalized with the fluorescent reagent meso-tetra(4-carboxyphenyl) porphine (TCPP) and their application in flow cytometry compensation. In this work, amine-functionalized polystyrene beads were treated with TCPP and the amide coupling reagent EDC (N-(3-dimethylaminopropyl)-N&#8242;-ethylcarbodiimide hydrochloride) at pH 6 and at room temperature for 16 h with agitation. The TCPP beads were isolated by centrifugation and resuspended in a pH 7 buffer for storage. TCPP-related particulates were observed as a byproduct. The number of these particulates could be reduced using an optional filtration protocol. The resultant TCPP beads were successfully used on a flow cytometer for compensation in experiments with human sputum cells labeled with multiple fluorophores. The TCPP beads proved stable following storage in a refrigerator for 300 days.

The protocol describes how porphyrin-based compensation beads for flow cytometry are prepared by the reaction of amine-functionalized polystyrene beads with the porphyrin TCPP and the amide coupling reagent EDC. A filtration procedure is used to reduce the particulate byproducts.

Preparation of Meso-Tetra(4-Carboxyphenyl) Porphine (TCPP) Stock Solution

To begin, weigh 49.0 to 50.9 milligrams of TCPP using an analytical balance. Round the weight to 1/10 of a milligram. Set the measured amount of TCPP aside, protected from light.Based on the amount of TCPP, add the required amounts of purified water and isopropanol to a 100 milliliter glass beaker and cover it with parafilm to protect it from evaporation. Next, weigh the required amount of sodium bicarbonate based on the amount of TCPP and add it to the 100 milliliter beaker containing purified water and isopropanol. Cover the solution with parafilm to protect it from evaporation, and place it on a stir plate for approximately 10 minutes.Once the contents are dissolved, ensure that the pH of the solution is between nine and 10. Next, carefully add the weighted TCPP, protect the solution from light, and stir it for approximately 30 minutes until dissolved. Store the solution in a polypropylene container at room temperature protected from light.

Combining Amine-Functionalized Polystyrene Beads with TCPP Solution

Begin by adding 4.3 milliliters of 0.1 molar MES buffer solution to a 15 milliliter polypropylene tube. Then add 288 microliters of the vortexed amine functionalized polystyrene bead suspension to the 15 milliliter polypropylene tube containing MES buffer. Vortex the MES bead solution for 15 seconds at maximum speed.Next, vortex the one milligram per milliliter TCPP solution for 60 seconds at maximum speed. Then add 1.2 milliliters of this solution to the MES bead suspension. After vortexing the resultant suspension for 15 seconds, cover the tube with foil.

Labeling Amine-Functionalized Polystyrene Beads with TCPP

To begin, add freshly prepared 4.5 milliliters of EDC working solution to a 15 milliliter polypropylene tube containing the amine-functionalized polystyrene beads and TCPP in MES buffer. Place the tube in an inverting rotator at 35 RPM for 16 hours at room temperature protected from light. Then, centrifuge it for 10 minutes.Aspirate the supernatant and re-suspend the beads in 0.8 milliliters of Hank's Balanced Salt Solution. Then, using a one milliliter pipette, transfer the bead solution to an amber polypropylene vial and store it at four degrees Celsius. The relationship between the particulate formation, the median, fluorescence intensity or MFI, and the bead TCPP/EDC ratio showed that with increasing ratios, the MFI increased until it formed a plateau.The particulates appeared only when EDC was used as a coupling reagent during the labeling procedure. Labeling the beads with TCPP alone resulted in much lower fluorescence intensity. When TCPP-labeled beads were tested after 300 days, no significant change in the MFI or in the particulate content was observed.The coupling of TCPP to the beads had a minimum effect on its fluorescence emission spectrum. However, its spectrum differed more from TCPP-labeled A549 lung cancer cells. In a multi-flora four labeled sputum sample, the TCPP-labeled beads worked equally well as the TCPP-stained A549 cells.

Quality Check (QC) of the TCPP Beads by Flow Cytometry

Begin the quality check of the beads labeled as meso-tetra 4-carboxyphenyl porphine or TCPP beads in the flow cytometer by running and collecting 10, 000 events of the rainbow beads. Then perform a rinse with water and collect 10, 000 events of the TCPP negative beads. After rinsing with water again, collect 10, 000 events of the TCPP positive beads.Then perform a one minute water rinse before appropriate cleaning and shut down the flow cytometer. A representative outcome of the TCPP bead labeling process determined by flow cytometry is shown here with a standardized profile of rainbow beads. These beads serve as quality checks for standardizing the laser voltages to detect TCPP.The light scatter profile of amine functionalized polystyrene TCPP labeled beads was observed. The light scatter profile of non-functionalized beads was used as a negative bead control to calculate the compensation matrix. The population indicated by the red rectangle in the TCPP labeled beads was absent in the unlabeled bead suspension representing that the TCPP related particulates were not formed in the absence of TCPP and EDC.The fluorescence intensity of the labeled beads selected by the black gate was several logs higher than that of the unlabeled beads.

TCPP Bead Filtration

Begin the TCPP bead filtration by diluting 0.8 milliliters of the TCPP bead suspension with 3.20 milliliters of ice-cold Hanks'Balanced Salt Solution, or HBSS. Vortex the diluted bead suspension at maximum speed for 15 seconds. Then, remove the plunger from a disposable five-milliliter syringe and fit the syringe with a five-micrometer glass fiber tip filter having a 13-millimeter diameter.Next, add four milliliters of HBSS to the syringe before adding 0.5 milliliters of the vortexed diluted bead suspension. Using the plunger, filter the bead suspension through the syringe filter at approximately two drops per second. Then, wash the beads by drawing five milliliters of fresh HBSS into the syringe through the filter and pushing the HBSS out into the waste container at approximately two drops per second.To remove the beads from the filter, draw another five milliliters of fresh HBSS into the syringe through the filter. Then, remove the filter carefully from the syringe. After ejecting the bead suspension from the syringe into a 50-milliliter conical centrifuge tube, place the fresh filter back on the syringe and repeat the five-milliliter HBSS wash four times.Once done, discard the filter and syringe. Centrifuge the filtered bead suspensions at room temperature for 10 minutes at 1, 000g. Aspirate the supernatant from each 50-milliliter tube.Gently resuspend the beads and 0.5 milliliters of fresh HBSS. Transfer the combined beads with a P1000 micropipette to a new amber-colored glass vial, and store at four degrees Celsius. The filtration effect on a bead suspension comprising approximately 66%particulates is seen here.After filtration, the particulate content decreased to approximately 25%The adverse effects of filtration are the loss of beads and a slight loss in fluorescence intensity.
