pseudofracture: an acute peripheral tissue trauma model

A seguito di un trauma è uno dei primi iper-reattiva risposta infiammatoria che può portare a disfunzione d&#39;organo multipla e alta mortalità nei pazienti con trauma, questa risposta è spesso accompagnata da una immunosoppressione ritardato che aggiunge le complicazioni cliniche di infezione e può anche aumentare la mortalità 1-9. Molti studi hanno cominciato a valutare questi cambiamenti nella reattività del sistema immunitario dopo un trauma. 10-15 Gli studi immunologici sono fortemente sostenute attraverso la grande varietà di transgenici e topi knockout per la modellazione in vivo;. Aiuto di questi ceppi nelle indagini dettagliate per valutare i meccanismi molecolari coinvolti nella risposta immunologica 16-21 La sfida di modellazione trauma sperimentale murino è un&#39;indagine a lungo termine, come le tecniche di fissazione della frattura nei topi, può essere complesso e non facilmente riproducibili. 22-30 Questo modello pseudofracture, un modello di trauma facilmente riproducibili, supera queste difficoltà, immunologicamente mimando un ambiente frattura degli arti, consentendo libertà di movimento per gli animali e la sopravvivenza a lungo termine senza il continuo, l&#39;uso prolungato di anestesia. L&#39;intento è quello di ricreare le caratteristiche delle fratture delle ossa lunghe; muscoli feriti e dei tessuti molli sono esposti a osso danneggiato e midollo osseo senza rompere l&#39;osso nativo. Il modello pseudofracture compone di due parti: un accordo bilaterale di lesione da schiacciamento muscolare alla arti posteriori, seguito da iniezione di una soluzione di osso in questi muscoli feriti. La soluzione osso è preparata dalla raccolta delle ossa lunghe da entrambe le zampe posteriori di età e peso corrispondenti donatore singenici. Queste ossa vengono poi pigiate e risospesi in tampone fosfato per creare la soluzione di osso. Frattura del femore bilaterale è un modello di uso comune e consolidata di traumi degli arti, ed è stato il modello comparativo durante lo sviluppo del modello pseudofracture. Tra le varietà di modelli di frattura a disposizione, abbiamo scelto di utilizzare un metodo chiusa di frattura con lesione dei tessuti molli come il nostro confronto al pseudofracture, come volevamo una sterile ancora proporzionalmente grave trauma dei tessuti periferici modello. 31 Shock emorragico è un reperto comune nel contesto di gravi traumi, e l&#39;ipoperfusione globale aggiunge un elemento molto rilevante per un modello di trauma. Pseudofracture 32-36 Il modello può essere facilmente abbinato ad un modello di shock emorragico per un modello di traumi multipli di gravità elevata 37.
