When handling mouse blood vessels, it is important to remember that the endothelium is fragile and that any excessive mechanical force will damage endothelial cells. For example, endothelial cells break or detach from the vessel wall if the vessel is perfused too forcefully, which can easily happen when the vasculature is perfused using a hand-operated syringe.

For histopathological studies by light microscopy, three dimensional pieces of biological tissues are routinely processed for paraffin embedment followed by sectioning and staining. A tissue sample that has been paraffin-embedded may be several millimeters in all three dimensions. However, for the purpose of light microscopy, it must be first sectioned so that light can pass through and then stained so that the thin section yields enough contrast for imaging. Because sectioned specimens are usually 5-10 &#181;m in thickness, one sees only a very small fraction of the whole specimen in two dimensions at a time. It is possible to collect sequential sections and, after imaging each section individually, perform computer-assisted reconstruction of the 3D images, but this is a tedious job indeed. Histopathology of blood vessels, especially for studying the pathogenesis of atherosclerosis, presents unique problems. Atherosclerosis is a focalized disease that develops locally in areas where disturbed blood flow occurs. Furthermore, the disease is initiated within the intima, a thin tissue consisting of a monolayer of endothelial cells and extracellular matrix, of large arteries. For these reasons, it is a challenge to locate and study early lesions using sectioned blood vessels because one can easily miss sectioning the lesion. Even if a section does include a diseased area, one will see only a 5-10 &#181;m portion containing endothelial cells and other vascular wall cells in the media and adventitia.
Whole mount en face (pronounced &#228;n &#712;f&#228;s) preparations allow us to survey a wide area of the blood vessel surface such as the entire aorta from the aortic root all the way down to the common iliac arteries. Using such a specimen stained with specific antibodies and other specific probes, one can pinpoint the location of lesions and also where various molecular events occur in endothelial cells in conjunction with atherogenesis such as changes in the expression, localization, and posttranslational modifications of proteins. In addition to studying atherogenesis, the endothelial cell shape observed in en face preparations is used as an indicator of the regional time-averaged blood flow pattern. Such data are important for studying mechanosignaling of endothelial cells in situ. For this purpose, routine histological cross-sectioned blood vessels are not useful. Thus, for vascular medicine and biology, it is especially important to acquire a technique for making en face preparations of blood vessels that allows one to observe a wide area of the vessel surface as well as the deeper subsurface areas of the vessel.
As reviewed by Jelev and Surchev1, vascular biologists have developed various methods to observe the lining of blood vessels en face. Some ingenious methods were developed in the 1940&#39;s and 1950&#39;s. Using these methods, they were able to study the fundamental organization of endothelial cells that line the inner surface of blood vessels. However, because of the way these en face preparations are prepared (the so-called Hautchen method2,3,4 or peeling off of the vessel surface5) and the way the specimen was stained, it was not always possible to obtain uninterrupted morphological information from the vessel surface into the deeper areas of the blood vessel wall. Whole mount en face vessel preparation combined with immunofluorescence staining allowed us to not only study endothelial cell morphology and protein expression and localization in these cells, but also to extend such studies to the subendothelial region of the vessel wall. Early studies using blood vessel en face preparations stained immunoflurescently began to appear in the 1980&#39;s6,7. With the advent of laser scanning confocal microscopy and more recently multiphoton microscopy, one can now obtain clear in-focus images of the blood vessel wall structure in immunofluorescently stained en face vessel samples as well as the vascular network in live animals8,9,10,11. These computer-based imaging techniques create in-focus optical sectioned images, and by stacking up such images, one can obtain reconstructed 3D images of the vessel wall and the vascular network in tissues. In addition, one can generate images of a section made along the Z-axis of the reconstructed image12,13.
In this article, we will illustrate a method for preparing en face preparations of the mouse aorta and the carotid artery for immunofluorescent staining. En face preparations can be made even after these vessels have been experimentally manipulated. For example, a carotid artery may be partially ligated and then an en face preparation made after such a surgery. For this reason, we will also describe in this article how we do a partial ligation on the carotid artery. Compared with making similar preparations from larger animals such as rats, rabbits, and humans, mouse vessels are small in size and more fragile, thus requiring added care for handling during surgical isolation of vessels and preparing them for antibody staining and microscopy. Because the most commonly used animal model for genetic modification is the mouse, it becomes critical for many investigators to handle mouse vessels without damaging them. In this manuscript, we will describe how to handle mouse blood vessels when making en face preparations of the mouse aorta and carotid artery. For the purpose of demonstration, we will use wild type C57/b6 mice.

Sections of paraffin embedded tissues are routinely used for studying tissue histology and histopathology. However, it is difficult to determine what the three-dimensional tissue morphology is from such sections. In addition, the sections of tissues examined may not contain the region within the tissue that is necessary for the purpose of the ongoing study. This latter limitation hinders histopathological studies of blood vessels since vascular lesions develop in a focalized manner. This requires a method that enables us to survey a wide area of the blood vessel wall, from its surface to deeper regions. A whole mount en face preparation of blood vessels fulfills this requirement. In this article, we will demonstrate how to make en face preparations of the mouse aorta and carotid artery and to immunofluorescently stain them for confocal microscopy and other types of fluorescence-based imaging.

A typical en face immunofluorescence image of the endothelium is shown in Figure 3. This image shows a single optical section of a mouse aorta taken near the opening of an intercostal artery (the large dark egg-shaped area). En Face Preparation of Mouse Blood Vessels

A procedure for making en face preparations of the mouse carotid artery and aorta is described. Such preparations, when immunofluorescently stained with specific antibodies, enable us to study localization of proteins and identification of cell types within the entire vascular wall by confocal microscopy.

The overall goal of this procedure is to prepare the mouse aorta and carotid artery for en face immuno staining after left carotid artery ligation. This method allows us to study organization, each protein expression, and possible state of antral cells and other vascular cells in immuno stain methods. En face preparations allow the analysis of large areas across blood vessels for the assessment of regional difference in the expression of various normal and possible cell parameters.Demonstrating the procedure will be Dr.Quaico a micro surgeon in our group. Begin by placing the anesthetized mouse onto a surgical board in the supine position. After confirming a lack of response to toe pinch tape the fore paws to the board in the out stretched position and both hind legs to the right side of the mouse to expose to left side of the neck.Remove the hair around the cervical area and disinfect the exposed skin. Cover the mouse with a sterilized surgical drape with a hole over the surgical region and apply ointment to the animal's eyes. Move the mouse under the dissecting microscope, and make a ventral mid line incision around the cervical area.Reposition the salivary glands that cover the blood vessels to the left side of the animal to expose the left common carotid artery, which bifurcates into the left internal carotid artery and the left external carotid artery. Gently remove the connective tissue around and below the left internal carotid artery and use a forceps to pass a 2.5 centimeter length of pre cut six O silk suture under the artery, using a second forceps to pull the suture roughly one third of its length to the other side of the vessel. Ligate the left internal carotid artery.Then remove the connective tissue around the left external carotid artery as just demonstrated, and make a ligation proximal to the left superior thyroid artery. When all of the arteries except the occipital artery have been ligated, return the salivary glands to their original positions and hydrate the surgical field with two to three drops of saline. Then use six O coated vicryl sutures to close the skin and place the mouse in the pre warmed cage with monitoring until full recumbency.At the termination of the experiment secure the mouse in the supine position on a dissecting board and use iris scissors to expose the abdominal cavity with a mid line incision. Cut the ribs laterally to the sternum to expose the thoracic cavity. Then nick the femoral artery and insert a 26 gauge needle attached to a gravity profusion set up into the apex of the left ventricle to profuse the circulatory system with saline solution supplemented with heparin.Continue the profusion until the saline flowing from the cut becomes clear. Then switch the profusion system from saline to 4%paraformaldehyde in PBS. After five minutes move the mouse under the dissecting microscope and use blunt end scissors and forceps to harvest the aorta and the left and right carotid arteries.Next, transfer the tissues into a Petri dish of PBS and carefully remove the fat and connective tissues, followed by the separation, and longitudinal splitting of the aorta and carotid arteries to expose the endothelium. The single most important thing to remember when making en face blood vessel preparations is that the endothelial cells are easily damaged by rough handling. Do not stretch the vessels at any time, especially during the harvesting and cleaning steps.Then transfer each vessel into individual wells of a 12 well plate containing 0.5 milliliters of permeabilizing solution per well. Permeabilize the blood vessels for 10 minutes with rocking at room temperature, followed by a brief wash in PBS. Block any nonspecific binding with a 30 minute incubation in 10%normal serum from the same species as the planned secondary antibodies in TTBS, with rocking at room temperature.Then label the samples with the primary antibodies of interest diluted in TTBS with 10%normal serum overnight with rocking at four degrees Celsius. The next morning, rinse the blood vessels with three 10 minute washes in TTBS with rocking at room temperature, followed by incubation in the appropriate secondary antibodies diluted in TTBS and 10%normal serum and dappy. Return the samples to the rocker and cover to protect from light.After one hour at room temperature with rocking wash the samples three times in fresh TTBS as demonstrated, followed by a brief rinse in PBS, and place one drop of anti fade reagent per vessel onto individual 22 by 50 millimeter cover glasses. Under a microscope, place one vessel into each drop of reagent with the endothelium facing down, and cover the vessels with one 22 by 75 millimeter slide glass per sample. Place the slides on a clean laboratory wipe and cover them with two pieces of laboratory wipe and 3.5 kilograms of weight to flatten the vessels.After a maximum of five minutes, remove the weight, and wipe the excess solution from around the cover slips. Then apply nail polish to the four corners of the cover slips and place the slides in a slide box, cover slip side up. The next morning use more nail polish to seal the cover slips completely and image the vessels as soon as the nail polish is dry.Here a typical en face immuno florescence image of endothelial double stained with anti VE cadherin and anti vascular cell adhesion molecule one antibodies and illustrating a single optical section of a mouse aorta taken near the opening of an intercostal artery is shown. Note the green linear outline staining at the adherence junction of the endothelial cells and the strong anti vascular cell adhesion molecule one staining at the opening of the intercostal artery where the disturbed blood flow is known to occur. In these images the en face staining of a partially ligated left carotid and a control unligated right artery one day after the surgery can be observed, with an increased anti vascular cell adhesion molecule one staining apparent in the ligated vessel.After watching this video you should have a good understanding of how to make a partial left carotid artery ligation and to make en face preparations in mouse blood vessels. Take this with your optical to other small animals. The use of en face preparations is now limited to immuno florescence staining, but can also be used with other methods, such as for study of those whole regions after oriole staining, or for ex vivo counter experiments.

Preparation of Dissociated Mouse Cortical Neuron Cultures

This video will guide you through the process for generating cortical neuronal cultures from late embryo and early postnatal mouse brain. These cultures can be used for a variety of applications including immunocytochemistry, biochemistry, electrophysiology, calcium and sodium imaging, protein and/or RNA isolation. These cultures also provide a platform to study the neuronal development of transgenic animals that carry a late embryonic or postnatal lethal gene mutation. The procedure is relatively straight forward, requires some experience in tissue culture technique and should not take longer than two to three hours if you are properly prepared. Careful separation of the cortical rind from the thalamo-cortical fiber tract will reduce the number of unwanted non-neuronal cells. To increase yields of neuronal cells triturate the pieces of the cortical tissue gently after the enzyme incubation step. This is imperative as it prevents unnecessary injury to cells and premature neuronal cell death. Since these cultures are maintained in the absence of glia feeder cells, they also offer an added advantage of growing cultures enriched in neurons.

In vivo Imaging and Therapeutic Treatments in an Orthotopic Mouse Model of Ovarian Cancer

Human cancer and response to therapy is better represented in orthotopic animal models. This paper describes the development of an orthotopic mouse model of ovarian cancer, treatment of cancer via oral delivery of drugs, and monitoring of tumor cell behavior in response to drug treatment in real time using in vivo imaging system. In this orthotopic model, ovarian tumor cells expressing luciferase are applied topically by injecting them directly into the mouse bursa where each ovary is enclosed. Upon injection of D-luciferin, a substrate of firefly luciferase, luciferase-expressing cells generate bioluminescence signals. This signal is detected by the in vivo imaging system and allows for a non-invasive means of monitoring tumor growth, distribution, and regression in individual animals. Drug administration via oral gavage allows for a maximum dosing volume of 10 mL/kg body weight to be delivered directly to the stomach and closely resembles delivery of drugs in clinical treatments. Therefore, techniques described here, development of an orthotopic mouse model of ovarian cancer, oral delivery of drugs, and in vivo imaging, are useful for better understanding of human ovarian cancer and treatment and will improve targeting this disease.

Isolation and Culture of Pulmonary Endothelial Cells from Neonatal Mice

Endothelial cells provide a useful research model in many areas of vascular biology. Since its first isolation 1, human umbilical vein endothelial cells (HUVECs) have shown to be convenient, easy to obtain and culture, and thus are the most widely studied endothelial cells. However, for research focused on processes like angiogenesis, permeability or many others, microvascular endothelial cells (ECs) are a much more physiologically relevant model to study 2. Furthermore, ECs isolated from knockout mice provide a useful tool for analysis of protein function ex vivo. Several approaches to isolate and culture microvascular ECs of different origin have been reported to date 3-7, but consistent isolation and culture of pure ECs is still a major technical problem in many laboratories. Here, we provide a step-by-step protocol on a reliable and relatively simple method of isolating and culturing mouse lung endothelial cells (MLECs). In this approach, lung tissue obtained from 6- to 8-day old pups is first cut into pieces, digested with collagenase/dispase (C/D) solution and dispersed mechanically into single-cell suspension. MLECS are purified from cell suspension using positive selection with anti-PECAM-1 antibody conjugated to Dynabeads using a Magnetic Particle Concentrator (MPC). Such purified cells are cultured on gelatin-coated tissue culture (TC) dishes until they become confluent. At that point, cells are further purified using Dynabeads coupled to anti-ICAM-2 antibody. MLECs obtained with this protocol exhibit a cobblestone phenotype, as visualized by phase-contrast light microscopy, and their endothelial phenotype has been confirmed using FACS analysis with anti-VE-cadherin 8 and anti-VEGFR2 9 antibodies and immunofluorescent staining of VE-cadherin. In our hands, this two-step isolation procedure consistently and reliably yields a pure population of MLECs, which can be further cultured. This method will enable researchers to take advantage of the growing number of knockout and transgenic mice to directly correlate in vivo studies with results of in vitro experiments performed on isolated MLECs and thus help to reveal molecular mechanisms of vascular phenotypes observed in vivo.

Here, we describe a protocol for isolation and culture of murine pulmonary endothelial cells. Generation of Mice Derived from Induced Pluripotent Stem Cells

The production of induced pluripotent stem cells (iPSCs) from somatic cells provides a means to create valuable tools for basic research and may also produce a source of patient-matched cells for regenerative therapies. iPSCs may be generated using multiple protocols and derived from multiple cell sources. Once generated, iPSCs are tested using a variety of assays including immunostaining for pluripotency markers, generation of three germ layers in embryoid bodies and teratomas, comparisons of gene expression with embryonic stem cells (ESCs) and production of chimeric mice with or without germline contribution2. Importantly, iPSC lines that pass these tests still vary in their capacity to produce different differentiated cell types2. This has made it difficult to establish which iPSC derivation protocols, donor cell sources or selection methods are most useful for different applications.
The most stringent test of whether a stem cell line has sufficient developmental potential to generate all tissues required for survival of an organism (termed full pluripotency) is tetraploid embryo complementation (TEC)3-5. Technically, TEC involves electrofusion of two-cell embryos to generate tetraploid (4n) one-cell embryos that can be cultured in vitro to the blastocyst stage6. Diploid (2n) pluripotent stem cells (e.g. ESCs or iPSCs) are then injected into the blastocoel cavity of the tetraploid blastocyst and transferred to a recipient female for gestation (see Figure 1). The tetraploid component of the complemented embryo contributes almost exclusively to the extraembryonic tissues (placenta, yolk sac), whereas the diploid cells constitute the embryo proper, resulting in a fetus derived entirely from the injected stem cell line.
Recently, we reported the derivation of iPSC lines that reproducibly generate adult mice via TEC1. These iPSC lines give rise to viable pups with efficiencies of 5-13%, which is comparable to ESCs3,4,7 and higher than that reported for most other iPSC lines8-12. These reports show that direct reprogramming can produce fully pluripotent iPSCs that match ESCs in their developmental potential and efficiency of generating pups in TEC tests. At present, it is not clear what distinguishes between fully pluripotent iPSCs and less potent lines13-15. Nor is it clear which reprogramming methods will produce these lines with the highest efficiency. Here we describe one method that produces fully pluripotent iPSCs and "all- iPSC" mice, which may be helpful for investigators wishing to compare the pluripotency of iPSC lines or establish the equivalence of different reprogramming methods.

Isolation and Culture of Mouse Primary Pancreatic Acinar Cells

This protocol permits rapid isolation (in less than 1 hr) of murine pancreatic acini, making it possible to maintain them in culture for more than one week. More than 20 x 106 acinar cells can be obtained from a single murine pancreas. This protocol offers the possibility to independently process as many as 10 pancreases in parallel. Because it preserves acinar architecture, this model is well suited for studying the physiology of the exocrine pancreas in vitro in contrast to cell lines established from pancreatic tumors, which display many genetic alterations resulting in partial or total loss of their acinar differentiation.

In this publication, we describe a rapid and convenient procedure for isolating and culturing primary pancreatic acinar cells from the murine pancreas. This method constitutes a valuable approach to study the physiology of fresh primary normal/untransformed exocrine pancreatic cells.

Quantifying Single Microvessel Permeability in Isolated Blood-perfused Rat Lung Preparation

The isolated blood-perfused lung preparation is widely used to visualize and define signaling in single microvessels. By coupling this preparation with real time imaging, it becomes feasible to determine permeability changes in individual pulmonary microvessels. Herein we describe steps to isolate rat lungs and perfuse them with autologous blood. Then, we outline steps to infuse fluorophores or agents via a microcatheter into a small lung region. Using these procedures described, we determined permeability increases in rat lung microvessels in response to infusions of bacterial lipopolysaccharide. The data revealed that lipopolysaccharide increased fluid leak across both venular and capillary microvessel segments. Thus, this method makes it possible to compare permeability responses among vascular segments and thus, define any heterogeneity in the response. While commonly used methods to define lung permeability require postprocessing of lung tissue samples, the use of real time imaging obviates this requirement as evident from the present method. Thus, the isolated lung preparation combined with real time imaging offers several advantages over traditional methods to determine lung microvascular permeability, yet is a straightforward method to develop and implement.

Indirect Immunofluorescence on Frozen Sections of Mouse Mammary Gland

Indirect immunofluorescence is used to detect and locate proteins of interest in a tissue. The protocol presented here describes a complete and simple method for the immune detection of proteins, the mouse lactating mammary gland being taken as an example. A protocol for the preparation of the tissue samples, especially concerning the dissection of mouse mammary gland, tissue fixation and frozen tissue sectioning, are detailed. A standard protocol to perform indirect immunofluorescence, including an optional antigen retrieval step, is also presented. The observation of the labeled tissue sections as well as image acquisition and post-treatments are also stated. This procedure gives a full overview, from the collection of animal tissue to the cellular localization of a protein. Although this general method can be applied to other tissue samples, it should be adapted to each tissue/primary antibody couple studied.

The indirect immunofluorescence protocol described in this article allows the detection and the localization of proteins in the mouse mammary gland. A complete method is given to prepare mammary gland samples, to perform immunohistochemistry, to image the tissue sections by fluorescence microscopy, and to reconstruct images.

En Face Detection of Nitric Oxide and Superoxide in Endothelial Layer of Intact Arteries

Endothelium-derived nitric oxide (NO) produced from endothelial NO-synthase (eNOS) is one of the most important vasoprotective molecules in cardiovascular physiology. Dysfunctional eNOS such as uncoupling of eNOS leads to decrease in NO bioavailability and increase in superoxide anion (O2.&#8722;) production, and in turn promotes cardiovascular diseases. Therefore, appropriate measurement of NO and O2.&#8722; levels in the endothelial cells are pivotal for research on cardiovascular diseases and complications. Because of the extremely labile nature of NO and O2.&#8722;, it is difficult to measure NO and O2.&#8722; directly in a blood vessel. Numerous methods have been developed to measure NO and O2.&#8722; production. It is, however, either insensitive, or non-specific, or technically demanding and requires special equipment. Here we describe an adaption of the fluorescence dye method for en face simultaneous detection and visualization of intracellular NO and O2.&#8722; using the cell permeable diaminofluorescein-2 diacetate (DAF-2DA) and dihydroethidium (DHE), respectively, in intact aortas of an obesity mouse model induced by high-fat-diet feeding. We could demonstrate decreased intracellular NO and enhanced O2.&#8722; levels in the freshly isolated intact aortas of obesity mouse as compared to the control lean mouse. We demonstrate that this method is an easy technique for direct detection and visualization of NO and O2.&#8722; in the intact blood vessels and can be widely applied for investigation of endothelial (dys)function under (physio)pathological conditions.

Immunofluorescence Analysis of Endogenous and Exogenous Centromere-kinetochore Proteins

"Centromeres" and "kinetochores" refer to the site where chromosomes associate with the spindle during cell division. Direct visualization of centromere-kinetochore proteins during the cell cycle remains a fundamental tool in investigating the mechanism(s) of these proteins. Advanced imaging methods in fluorescence microscopy provide remarkable resolution of centromere-kinetochore components and allow direct observation of specific molecular components of the centromeres and kinetochores. In addition, methods of indirect immunofluorescent (IIF) staining using specific antibodies are crucial to these observations. However, despite numerous reports about IIF protocols, few discussed in detail problems of specific centromere-kinetochore proteins.1-4 Here we report optimized protocols to stain endogenous centromere-kinetochore proteins in human cells by using paraformaldehyde fixation and IIF staining. Furthermore, we report protocols to detect Flag-tagged exogenous CENP-A proteins in human cells subjected to acetone or methanol fixation. These methods are useful in detecting and quantifying endogenous centromere-kinetochore proteins and Flag-tagged CENP-A proteins, including those in human cells.

Trans-inner Cell Mass Injection of Embryonic Stem Cells Leads to Higher Chimerism Rates

In an effort to increase efficiency in the creation of genetically modified mice via ES Cell methodologies, we present an adaptation to the current blastocyst injection protocol. Here we report that a simple rotation of the embryo, and injection through Trans-Inner cell mass (TICM) increased the percentage of chimeric mice from 31% to 50%, with no additional equipment or further specialized training. 26 different inbred clones, and 35 total clones were injected over a period of 9 months. There was no significant difference in either pregnancy rate or recovery rate of embryos between traditional injection techniques and TICM. Therefore, without any major alteration in the injection process and a simple positioning of the blastocyst and injecting through the ICM, releasing the ES cells into the blastocoel cavity can potentially improve the quantity of chimeric production and subsequent germline transmission.

Here we present a protocol to increase chimera production without the use of new equipment. A simple orientation change of the embryo for injection can increase the number of embryos produced, and potentially reduce the timeline to germline transmission.
