균주는 항생제의 부재 (대조군) 및 존재 (실험)에서 동일한 조건 (전단 + 효소 스트레스)에서 테스트됩니다. 각 채널의 위상 대비 및 형광 현미경 사진은 1 시간 동안 2 분마다 자동으로 촬영됩니다. 저항 지정은 그 때 대조군 채널에 있는 죽은 박테리아의 백분율을 대조채널에 있는 것과 비교하여 만들어집니다. 1 시간 후에, 세포 죽음 백분율이 1% 보다 큰 견본은 저항의 표시인 동안, 영향을 받기 쉬운 것으로 간주됩니다. 이 두 컷오프 사이에 떨어지는 백분율은 확정되지 않은 것으로 간주되며 샘플을 다시 테스트해야 합니다.

기계적/전단 응력은 고정된 박테리아를 통해 실온 매체의 흐름에 의해 생성됩니다. (참고: 미디어를 37°C로 온난화하는 것은 분석 결과에 큰 영향을 미치지 않습니다.) 자동 주사기 펌프는 1 ml/분의 유속으로 미세 유체 채널(200 μm x 400 μm)을 통해 미디어(사구 얼룩 +/-항생제 포함)를 강제하여 6.25kPa의 전단 력 또는 전단 속도 6,000초-1을제공합니다. 이 비율은 황색포도상구균에이전에 연구된 전단 응력과 같거나 초과합니다.
효소, 리소스타핀은 황색포도상구균 세포벽에 직접적인 손상을 일으키기 때문에 예비 실험을 위해 선택되었다. 리소스타핀(0.7 ng/ml)의 농도는 세균성 세포벽 손상을 유발하기에 충분했지만, 실험 기간 내에 항생제 없이 세균세포사멸을 유발하는 것만으로는 충분하지 않았다. 우리는 스트레스 기반 환경에서 항생제 감수성 검사를 위한 신속한 미세 유체 방법을 개발했습니다. 유체는 미세 유체 채널의 바닥에 고정 된 박테리아를 통해 고속으로 전달됩니다. 스트레스와 항생제의 존재에서, 박테리아의 취약 한 긴장 은 급속 하 게 죽는다. 그러나, 저항하는 박테리아는 이 스트레스가 많은 조건에서 살아남습니다. 이 방법의 뒤에 가설은 새로운: 항생제의 표적인 생화확적인 통로의 스트레스 활성화는, 항생제 감수성 시험을 가속화할 수 있습니다. 표준 항생제 감수성 검사 방법에 비해, 속도 제한 단계 - 세균 성장 - 항생제 적용 중에 생략된다. 이 방법의 기술적 구현은 표준 기술과 혁신적인 접근 방식의 조합입니다. 이 방법의 표준 부분에는 세균 배양 프로토콜, 폴리디메틸실록산(PDMS)의 미세 유체 채널 정의, 형광을 이용한 세포 생존 가능성 모니터링, 박테리아 계수에 대한 배치 이미지 처리 등이 포함됩니다. 이 방법의 혁신적인 부분은 기계적 응력 적용을 위한 배양 매체 흐름의 사용, 박테리아를 손상시키지 만 손상시키지 않는 효소의 사용, 세균 부착을 위한 미세어기 기판의 사용에 있다. 개발 된 플랫폼은 항생제 및 비 항생제 관련 약물 개발 및 테스트에 사용할 수 있습니다. 표준 세균현절 실험과 비교하여, 약물의 효과는 조절 기간 동안 반복적으로 켜고 끌 수 있다. 동일한 세균 집단의 반복적인 관찰은 동일한 실험의 과정을 통해 가능하다.
