Celcyclus controle controlepunten die zorgen voor nauwkeurige chromosoom segregatie voorkomen aneuploïdie en cel transformatie 1,2,3. In de huidige studie, vonden we dat inactivatie van PP2A-B56γ resulteerde in een verzwakte spindel vergadering checkpoint. Beeldvorming van de levende cel konden we observeren chromosomale verkeerde segregatie tijdens de mitose in PP2A-B56γ MEFs met nocodazole 9behandeld...

In het volgende protocol beschrijven we een handige methode om te visualiseren de chromosomale segregatie en de mitotische vooruitgang tijdens de celcyclus in muis embryonale fibroblasten met behulp van Histone 2B-GFP, BacMam 2.0 labeling en levende cel beeldvorming.
Celcyclus controle controlepunten controleren chromosoom segregatie en spelen een belangrijke rol bij de instandhouding van de genetische integriteit van de cel 1,2,3. Accumulatie van mis gescheiden chromosomen kan leiden tot aneuploïdie, dat is een kenmerk van de meest solide tumoren 4. Vandaar, detectie van achterblijvende chromosomen kan worden gebruikt als een methode om te studeren chromosomale instabiliteit.
Fluorescently label eiwitten kunnen worden gebruikt om te visualiseren live chromosoom segregatie en chromosomale achtergebleven maar de generatie van mCherry-gelabeld of H2B-GFP gelabeld eiwit vereist aanzienlijke kennis van gene levering en moleculaire biologie 5. Hier beschrijven we het gebruik van CellLight Histone2B-GFP BacMam 2.0 reagens, hierna genoemd CL-HB regent, omwille van de eenvoud. Dit reagens kan onmiddellijk worden gebruikt en dus elimineert bezorgdheid over vector kwaliteit en integriteit. Dit reagens hoeft bovendien niet het gebruik van potentieel schadelijke behandelingen of lipiden en kleurstof-laden chemicaliën. In tegenstelling tot conventionele fluorescerende labels, de CL-HB-regent vlekken onafhankelijk van de functie (dwz., membraanpotentiaal). De CL-HB-regent kan eenvoudig worden toegevoegd aan de cellen en eiwit expressie 's nachts ge¨ uncubeerd. De CL-HB-regent niet gerepliceerd in zoogdiercellen en kan worden gebruikt in de bioveiligheid niveau (BSL) 1 laboratorium-instellingen. Ook kan deze voorbijgaande transfectie worden gedetecteerd na overnachting incubatie gedurende maximaal 5 dagen, genoeg tijd om de meest dynamische cellulaire analyses uitvoeren.
Anderzijds kon chromosomale afwijkingen worden bestudeerd door verschillende technieken zoals stroom cytometry, immunohistochemistry of fluorescentie in situ hybridisatie (FISH) 6. Stroom cytometry kan worden gebruikt om te studeren aneuploïdie, die kan worden gemeten op basis van DNA-inhoud en de fase van cellen in de celcyclus. Hoewel stroom cytometry kan worden gebruikt voor het meten van aneuploïdie, biedt geen informatie op chromosomale verkeerde segregatie. VIS en immunohistochemistry technieken gebruik fluorescente sondes te binden aan DNA of chromosomen. Hoewel deze technieken een momentopname van de status van een bevolking van cellen bieden, laat ze niet levende cel imaging waardoor ontbreekt elke informatie verkregen via Spatio visualisatie van cytokinese in specifieke cellen gevolgd over een periode van tijd.
Dit protocol werd gebruikt voor het bestuderen van de achterblijvende chromosomen of chromosomale verkeerde segregatie in nocodazole behandeld muis embryonale fibroblasten (MEFs) van PP2A-B56γ-muizen geïsoleerd. In aanvulling op bovenstaande toepassing biedt dit protocol een eenvoudige tool om te etiketteren en visualiseren van chromosomale segregatie in verschillende celtypen, die kan worden gebruikt voor het bestuderen van de celcyclus verordening of chromosomale instabiliteit in tumorcellen. Bovendien, kan het ook worden gebruikt om te studeren chromosomale instabiliteit veroorzaakt door diverse drug behandelingen of de effecten van gene knock out resulterend in chromosomale verkeerde segregatie te bestuderen.

Chromosomen moeten worden betrouwbaar en uniform gescheiden in dochtercellen tijdens de mitotische celdeling. Trouw van chromosomale segregatie wordt beheerd door meerdere mechanismen waarin de spindel vergadering Checkpoint (SAC). De SAC is onderdeel van een complexe feedback-systeem dat verantwoordelijk is voor preventie van een cel vooruitgang door middel van mitose tenzij alle chromosomale kinetochoren hebben gehecht aan de spindel microtubuli. Chromosomale achterblijvende en abnormaal chromosoom segregatie is een indicator van disfunctionele celcyclus controle checkpoints en kan worden gebruikt voor het meten van de genomic stabiliteit van cellen te verdelen. Deregulering van de SAC kan resulteren in de omvorming van een normale cel tot een kwaadaardige cel door de accumulatie van fouten tijdens chromosomale segregatie. Uitvoering van de SAC en de vorming van de complexe kinetochoor zijn strak gereguleerd door interacties tussen kinases en fosfatase zoals eiwit fosfatase 2A (PP2A). Dit protocol beschrijft levende cel beeldvorming van achterblijvende chromosomen in muis embryonale fibroblasten geïsoleerd van muizen die had een knock-out van de regelgevende subeenheid van PP2A-B56γ. Deze methode overwint de tekortkomingen van andere celcyclus controle beeldvormingstechnieken zoals stroom cytometry of immunocytochemie waarmee slechts een momentopname van een cel cytokinese status, in plaats van een dynamische Spatio visualisatie van chromosomen tijdens de mitose.

MEFs van wild type en PP2A-B56γ-muizen waren zaadjes in een 2-well kamer cover glas en toegestaan om te koppelen. Op dag 2, MEFs zijn gesynchroniseerd met behulp van 0,1% FBS gedurende 24 uur. Op dag 3, MEFs in de media met 200 ng/mL nocodazole en 30 PPC CL-HB regentwere uncubeerd 18u op 37° C en 5% CO2.
