Cellen syklus kontroll sjekkpunkter som sikrer nøyaktig kromosom segregering hindre aneuploidy og celle transformasjon 1,2,3. I studien, fant vi at inaktivering av PP2A-B56γ resulterte i en redusert spindelen montering checkpoint. Levende celle imaging tillatt oss å observere chromosomal mis segregering under mitose i PP2A-B56γ MEFs behandlet med nocodazole 9.

I den følgende protokollen beskriver vi en praktisk metode for å visualisere chromosomal segregering og mitotisk progresjon i cellen syklus i mus embryonale fibroblaster Histone 2B-GFP, BacMam 2.0 merking og levende celle bildebehandling.
Cellen syklus kontroll sjekkpunkter overvåke kromosom segregering og spiller en viktig rolle i vedlikehold av genetisk integriteten til den celle 1,2,3. Akkumulering av mis segregerte kromosomer kan føre til aneuploidy, som er et kjennetegn på mest solide svulster 4. Påvisning av henger kromosomene kan derfor brukes som en metode for å studere chromosomal ustabilitet.
Fluorescently merket proteiner kan brukes å visualisere live kromosom segregering og chromosomal henger men generering av mCherry-merket eller H2B-GFP merket protein krever betydelig kunnskap om gen levering og molekylærbiologi 5. Her beskriver vi bruk av CellLight Histone2B-GFP BacMam 2.0 reagens, heretter kalt CL-HB regent, for enkelhets skyld. Denne reagensen kan brukes umiddelbart og således eliminerer bekymringer om vektor kvalitet og integritet. I tillegg krever denne reagensen ikke bruk av potensielt skadelige behandlinger eller lipider og fargestoff lasting kjemikalier. I motsetning til konvensjonelle fluorescerende etiketter, CL-HB regent flekker uavhengig funksjon (dvs., membran potensial). CL-HB regent kan bare legges til cellene og ruges overnatting i protein uttrykk. CL-HB regent replikerer ikke i pattedyrceller og kan brukes i biosikkerhet (BSL) 1 laboratorium innstillinger. Også kan denne forbigående transfection bli oppdaget etter overnatting inkubasjonstiden for opp til 5 dager, nok tid til å utføre mest dynamiske mobilnettet analyser.
Alternativt kunne kromosomavvik bli studert av ulike teknikker som flowcytometri, immunohistochemistry eller fluorescens i situ hybridisering (fisk) 6. Flowcytometri kan brukes til å studere aneuploidy, som kan måles basert på DNA innhold og fasen av celler i cellen syklus. Selv om flowcytometri kan brukes til å måle aneuploidy, gir det ikke informasjon på chromosomal mis raseskille. FISK og immunohistochemistry bruke fluorescerende sonder binde til DNA eller kromosomer. Disse teknikkene gir et øyeblikksbilde av statusen for en populasjon av celler, tillater de ikke levende celle imaging dermed mangler informasjon innhentet gjennom spatiotemporal visualisering av cytokinesis i bestemte celler fulgte over en tidsperiode.
Denne protokollen ble brukt til å studere henger kromosomer eller chromosomal mis segregering i nocodazole behandlet mus embryonale fibroblaster (MEFs) isolert fra PP2A-B56γ-mus. I tillegg til program over gir denne protokollen et enkelt verktøy for å merke og visualisere chromosomal segregering i ulike celletyper som kan brukes til å studere celle syklus regulering eller chromosomal ustabilitet i kreftceller. Dessuten, kan det også brukes til å studere chromosomal instability forårsaket av ulike medikamentelle behandlinger eller studere virkningene av genet banke ut resultere i chromosomal mis raseskille.

Kromosomene må være pålitelig og jevnt segregert i datter celler under mitotisk celledeling. Gjengivelse av chromosomal segregering styres av flere mekanismer som inkluderer spindelen montering Checkpoint (SAC). SAC er del av en kompleks feedback-systemet som er ansvarlig for forebygging av en celle fremgang gjennom mitose med mindre alle chromosomal kinetochores har lagt for å vri piskehale som henger. MEFs fra vill type og PP2A-B56γ-mus var sådd i en 2-vel kammer cover glass og lov til å feste. På dag 2, MEFs ble synkronisert med 0,1% FBS 24 h.
