1. 流体打击伤害
<ol><li>穿实验室外套或手术服、外科口罩、手术手套、头部覆盖物,并消毒与手术部位接触的所有工具和材料。</li>
	<li>麻醉10-12周大,雄性,威斯塔大鼠(350-400克),3%的旋脱剂和1L/min氧气在适合大鼠大小的感应室。将大鼠从感应室中取出,一旦它失去知觉,将其移到准备区域。将无菌的眼科膏放入两只眼睛。</li>
	<li>用电动剪子将毛发剪在老鼠头上,用#40刀片从眼睛上方到耳朵的刀底,以产生足够的手术场。从现场去除任何松散的、剪短的头发。</li>
	<li>通过在剃光头皮上涂抹 2% 氯西丁砂洗,然后加入 70% 乙醇,清洁手术部位。从中心开始,在远离切口部位的同心圆中向外移动。重复此过程 3 次。以同样的方式将 Betadine 溶液应用到现场,并允许干燥。</li>
	<li>将麻醉大鼠放入立体框架中,通过鼻塞酮将麻醉保持在2-3%异构-1 L/min氧气中。检查后肢的戒断反射损失和苍白反射的损失,以确保大鼠在麻醉的手术平面上。</li>
	<li>在整个手术过程中监测呼吸速率、心率、体温和氧饱和度。将心率保持在 300-400 bpm 之间,SpO2保持在 90% 以上。 注:连接到后脚的脉搏血氧仪可用于提供恒定读取的心率和SpO2。心率超过400 bpm表示大鼠没有充分麻醉。自我调节的加热垫,与直肠温度计结合,设定在37°C,可以放置在大鼠下整个手术,以保持体温。带光源的立体显微镜与光纤灯相结合,有助于可视化过程。</li>
	<li>使用 23 g 针头在切口前 10 - 15 分钟将盐酸盐酸注射到局部麻醉部位的头皮中。</li>
	<li>使用#10手术刀通过头皮的皮肤和肌肉进行1.5-2.5厘米的中线切口。收回皮肤和肌肉,露出头骨,并提供一个清晰的手术场。用无菌棉签反射从骨骼中消失的底层筋膜和脂肪组织。 注: 电烧灼装置可用于实现快速到基。</li>

	<li>在头骨上涂抹3%过氧化氢以干燥骨骼。 注:如果骨骼不够干燥,牙科水泥将无法正确粘附并形成固体密封。</li>
	<li>通过左骨创建一个直径为 5 mm 的颅骨切除部位。 注: 放置在连接到立体定向框架的电源钻中的颤音位有助于启动颅切除术。使用直径为 5 mm 的钻头,通过剩余的骨慢慢完成颅骨切除。当接近完成颅骨切除术时,反向旋转颤音,以防止底层的杜拉母体破裂。盘盘周围会变薄,轻轻按压时,头骨皮瓣会感觉松弛。</li>
	<li>用手术固化器和光滑的组织钳子去除骨瓣。 注:可能会出现一些出血,但通过用无菌棉签施加温和压力可以迅速实现止血。</li>
	<li>使用立体显微镜和照明来目视检查杜拉是否有任何破裂迹象。 颅骨切除部位的周长周围会留有一层薄薄的骨头。 用光滑的组织钳轻轻去除这个边缘,注意不要破裂杜拉。</li>
	<li>用70%乙醇擦拭头骨,去除任何骨尘,干燥头骨。</li>

	<li>使用注射器和针头向 Luer 锁轮毂注入无菌的不含防腐剂的人工脑脊髓液 (CSF) 溶液 (pH 7.4),以便在边缘顶部上方看到凸出的盐水。 注:当牙科水泥干燥时,溶液将保持杜拉湿润,并指示密封的完整性。如果溶液水平完全下降,则表示系统存在泄漏,必须拆下并更换 Luer 锁。</li>
	<li>一旦牙科水泥完全固化,停止气体麻醉,并将大鼠从立体框架中取出。</li>
	<li>将大鼠放在 FPI 设备旁边的平台上。</li>
	<li>FPI 器件具有一个弯曲的金属尖端,从储液罐末端的压力传感器延伸。将 12 厘米长的压力管固定在弯曲尖端的末端,并在 2 厘米公禄锁扭接连接器中端接另一端。通过将大鼠头骨上的中心线母端连接到雄性连接器,将大鼠固定到 FPI 设备。 注:确保连接牢固牢固,并且所有气泡均已从系统移除。</li>
	<li>将动物置于胸腔中,并反复检查退退反射的返回。一旦大鼠恢复戒断反射,但仍镇静,释放FPI装置的钟摆,导致单个20ms压力脉冲并诱发损伤。 注:在动物被深度麻醉时,不要诱发损伤,这一点很重要,因为这往往会导致由于神经原性肺水肿而增加的死亡率。所有设备都显示可变性。然而,在用于此实验的装置上,锤的 17° 角度放置会产生 2.2 - 2.3 个大气压脉冲。没有受伤的假动物接受所有相同的程序,除了实际的流体脉冲诱导伤害。</li>
	<li>受伤后立即断开大鼠与FPI装置的连接,将其置于胸腔回位,并通过鼻锥提供补充氧气(1升/分钟),直到自发呼吸恢复。呼吸暂停是受伤的预期后果。如有必要,通过袋阀面罩定期进行手动呼吸,直到大鼠开始自行自发呼吸。 注:通常,呼吸暂停持续不到 2 分钟。由于儿茶酚胺爆裂,在压力脉冲被给药后,立即观察到心率的瞬时快速上升(>500 bpm)。这可以通过连接到大鼠脚的脉搏血氧仪进行监测,并可以作为已发生严重伤害的可能指示器。</li>
	<li>持续监测大鼠并记录右反射返回的时间(四肢稳定。</li>
	<li>每只大鼠的大气压力脉冲大小应在± 0.05大气中。确认每个压力脉冲在示波器上产生平滑信号,振幅和持续时间一致。 注: 噪音信号可能表示系统中的气泡在传递伤害脉冲之前必须拆下。在这个实验中,产生严重伤害的大气压力脉冲通常会导致动物的纠正时间为30-60分钟。这一范围的纠正时间与死亡率约40-50%有关。</li>
	<li>作为支持性护理,在皮下施用10 mL的预付盐水。</li>
	<li>将大鼠返回到其家笼,让它至少恢复4小时。 注:当大鼠立即被置于麻醉下时,死亡率有所上升。</li>
</ol>2. 皮质脑电图电极的植入和视频-EEG记录
<ol><li>受伤后4小时,如前所述对大鼠进行麻醉,并将其放回立体定向框架,以拆下Luer锁轮毂和牙科水泥。 注:轮毂和水泥很容易在中等压力下脱落。拆下轮毂时,请仔细检查杜拉是否有任何破裂或损坏。对杜拉造成破坏的任何动物进行即时安乐死。</li>
	<li>在要钻孔5个导孔的每个位置,对头骨施加0.5%的盐酸乙酸乙烷(见图1)。 </li>
	<li>使用手持式 0.1 mm 钻头在头骨上钻孔。</li>
	<li>将不锈钢电极螺钉固定到以下位置的每个导孔中:将参考螺钉固定在小脑上的羔羊上。记录电极被放置在:1)在半球的侧边和罗斯尔到颅切除术;2) 在半球的边边和胆管到颅切除术;3) 在半球对侧和罗斯尔到颅切除术;4) 在半球对边和胆对颅切除术。</li>
	<li>用70%乙醇擦拭头骨,去除任何骨尘。</li>
	<li>用一层薄薄的无菌骨蜡覆盖颅骨切除术部位,以覆盖暴露的杜拉。</li>
	<li>将电极阵列连接到 5 个 EEG 电极,将彩色编码电极导线的外露端紧密地包裹在其指定的不锈钢电极螺钉上。 注:每根电极导线的两端位于底座连接器内的特定指定位置。</li>
	<li>准备一块骨水泥浆。</li>
	<li>将电极导线收集到底座下方的线圈中,用骨水泥将电线和底座固定到位。将底座固定在位置,直到骨水泥固化。 注:骨骼必须特别干燥,并且有任何残留的血液无效,以便获得适当的粘附,并防止过早去除变送器。</li>
	<li>在将动物从立体定向框架中取出之前,将带有新电池的无线发射器连接到底座上。</li>
	<li>将动物放在其家庭笼子里,并将笼子放在靠近接收器的位置,并放置在指定的摄像机中。启动视频/EEG 录制。</li>
</ol>3. 视频-EEG录音的收集
<ol><li>在收集 EEG 信号之前,对将容纳大鼠进行 EEG 收集的房间进行频率扫描,以识别任何潜在的干扰频率,以防止收集具有背景噪声的任何频率的 EEG 记录。</li>
	<li>将所有发射机设置为不受干扰的特定频率。</li>
	<li>设置每个可编程变送器的采样频率和输入范围。 注:这可以使用系统制造商提供的智能工具完成。发射机的最大采样速率为 1000 Hz,最大输入范围为 ±10 mV。在本实验中,分析了0.5Hz至30Hz之间的脑电图录音。因此,采样率设置为 250 Hz。我们通常观察到小于 1 mV 的振幅。因此,设定的输入范围为 ±2 mV。</li>
	<li>使用制造商提供的 EEG 收集软件连续录制视频-EEG,从受伤当天开始,通过独特的频率将每个无线发射器连接到特定接收器。 注:每个发射机接收器对能够监控4个单极EEG通道,以及X、Y和Z平面的加速度。EEG 数据可以写入存储服务器。视频数据应保存在链接到存储服务器的 NAS 设备上。EEG 分析软件根据存储服务器维护的时间同步视频和 EEG 录制。</li>
	<li>使用视频收集软件使用自己的 2 MP 分辨率摄像机 (1920 x 1080) 录制每只大鼠的视频,该摄像机配置为以 30 帧/秒的速度录制。 注:每台摄像机都有自己的红外照明,用于在夜间进行视频采集。</li>
	<li>将系统配置为每 24 小时自动将所有视频和 EEG 录像保存到存储服务器。视频产生相当大的文件。</li>
</ol>4. 视频/EEG分析
<ol><li>以 1/10 的分辨率将视频与每个 EEG 录制同步。为此,使用系统制造商的视频/EEG 分析软件创建带有 EEG 和视频精确时间的标记的元文件。</li>
	<li>手动筛选脑电图记录,以识别定义癫痫活动的索引事件。</li>
	<li>使用视频/EEG 分析软件和索引 EEG 事件,创建使用关键参数(即特定频段的功率、频段与总功率的比率、加速度阈值等)来定义特征的配置文件潜在的癫痫发作事件。</li>
