肠道胶质细胞因其在肠道稳态和疾病过程(包括术后并发症)中的作用而越来越受到认可。从紧急探查剖腹手术中恢复的马患者术后炎症风险很高,这凸显了建立可重复的马肠道神经胶质原代细胞培养用于研究的重要性。
马绞痛(急腹症)的术后炎症不仅会导致客户成本增加、患者不适和住院时间增加,而且在许多情况下,事实证明会危及生命。肠道神经胶质细胞是一种独特的肠道细胞群,因其在感知胃肠道环境和与周围细胞类型交流方面的作用而日益得到认可。肠道胶质细胞和肠上皮细胞之间的相互作用可能被证明对于确定马肠道神经胶质细胞如何改变粘膜屏障以调节健康和绞痛中的炎症至关重要。
为了研究这种相互作用,我们提出了一种从马空肠建立原代马肠肠胶质细胞培养物并将培养物暴露于已知存在于绞痛中的炎症状况的方法。原代肠道神经胶质细胞培养物来自因与绞痛无关的原因而被安乐死的成年马。显微解剖肠绒毛和固有层,露出粘膜下层。分离的粘膜下层用胶原酶、蛋白酶和牛血清白蛋白进行酶消化 2-3 h。接下来,包括离心、移液和细胞过滤器 (40-100 μm) 的机械消解,得到用于在 0.05 mg/mL 聚-L-赖氨酸包被的孔上铺板的沉淀,浓度为 ~400,000 个细胞/300 μL 培养基。
汇合和第一次传代后,然后将肠胶质细胞暴露于马重组 IL-1β (0、10、25 ng) 24 小时。为了模拟绞痛时的上皮-胶质细胞相互作用,将对照或处理的肠胶质细胞调节的培养基直接添加到融合的马空肠单层中,同时使用双电极 EndOhm 室测量跨上皮电阻 (TEER)。这些数据只是马肠道神经胶质细胞培养的众多潜在有影响力应用之一。
马绞痛是急诊会诊最常见的医疗主诉1。由于高达 17% 的马匹需要手术矫正,因此提高术后结果的努力应该是马医学研究的重中之重2。目前,术后绞痛患者患多种危及生命的疾病的风险很高,包括败血症/内毒素休克(12.3% 的患者)和术后肠梗阻(13.7% 的患者)3。尽管术后并发症的治疗取得了进展,但仍需要先进的治疗方法来预防或治疗这些疾病。
最近的研究强调了绞痛患者的局部和全身炎症状态4。例如,促炎蛋白,如肿瘤坏死因子 (TNF) α、白细胞介素 1β (IL-1β)、白细胞介素 6 (IL-6) 和单核细胞趋化蛋白-1,都已被证明在肠绞痛粘膜中的表达显著增加于正常肠组织4。从系统性的角度来看,已经证明肠道组织中 TNF α 的增加与术后鼻胃管反流大于 2 L 的风险增加相关,这是术后运动障碍的一般衡量标准4。众所周知,白细胞介素 IL-1β 和 TNF α 的给药能够诱导感染性休克的临床症状5。
