1. Vorbereitung am Tag vor der Dissektion
<ol><li>Eine Wolframnadel24 elektrolytisch schärfen, so dass der resultierende Durchmesser ca. 0,03 mm beträgt. Stellen Sie die gelieferte Spannung auf ca. 14 V ein und verwenden Sie 3,3 M NaOH. Das Schärfen sollte nicht länger als 1 oder 2 Minuten dauern. VORSICHT: NaOH ist stark korrosiv und verursacht Verbrennungen bei Kontakt mit der Haut. Tragen Sie beim Handling Handschuhe und Schutzbrillen und arbeiten Sie in einer Dunstabzugshaube. HINWEIS: Nach Gebrauch NaOH in einem Falcon-Rohr aus Polypropylen aufbewahren.</li>
	<li>Stellen Sie die vorbereiteten Bildmedien her. Kombinieren Sie in einem konischen 15-ml-Röhrchen 4,25 ml Schneider-Medien mit 750 μL fetalem Rinderserum (FBS, 15% Endkonzentration) und 27,5 μL Penicillin-Streptomycin (0,05 U/μL Penicillin, 0,05 μg/μL Endkonzentration). Lagern Sie dieses vorbereitete Bildmedium bei 4 °C.</li>
	<li>Stellen Sie Heptan-Klebstoff-Lösung her. Fügen Sie etwa 0,5 ml Heptan zu einer 20 ml verschließbaren Durchstechflasche hinzu, die mit etwa 20 cm doppelseitigem Klebeband gefüllt ist. Etwa eine Stunde lang auf einem Nutator schaukeln oder mit einer Pipettenspitze umrühren, bis eine Konsistenz zwischen Wasser und Glycerin erreicht ist. Diese Lösung von gelöstem Kleber hält mehrere Tage, bevor das Heptan verdunstet. Um die Lösung aufzufrischen, fügen Sie ein zusätzliches 0,5 ml Heptan und Gestein hinzu. HINWEIS: Eine effektive Heptankleberlösung kann unter Verwendung verschiedener Verhältnisse von Heptan zu Band sowie unterschiedlicher Schaukel- / Rührdauer hergestellt werden. Die oben genannten Spezifikationen sind lediglich Vorschläge zur Vorbereitung oder Auffrischung einer solchen geeigneten Lösung.</li>
</ol>2. Embryonenentnahme – 15–17 h vor der Dissektion
<ol><li>Fügen Sie frische Hefepaste zu einer Apfelsaft-Agarplatte25 hinzu. Stellen Sie einen Embryo-Sammelkäfig auf, indem Sie erwachsene Fliegen (weniger als 10 Tage alt) in eine leere Futterflasche geben, die mit kleinen Löchern26 perforiert ist. Verschließen Sie den Sammelkäfig mit der mit Hefe versehenen Agarplatte, die an die Flasche geklebt ist, und stellen Sie den Käfig mit der Tellerseite nach unten für 1 Stunde in einen 25 °C dunklen Inkubator. HINWEIS: Die Fliegen müssen ein Transgen exprimieren, das die Gonade fluoreszierend markiert, zum Beispiel six4-eGFP::Moesin 20, da die Dissektion von Embryonen unter einemstereofluoreszierenden Mikroskop stattfindet. Siehe Tabelle der Materialien für eine Liste der Genotypen, die zur Markierung der Gonade verwendet werden.</li>
	<li>Entfernen Sie den Käfig aus dem Inkubator und verwerfen Sie diese erste Sammlung. Ersetzen Sie es durch eine frisch gehegte Agarplatte und legen Sie den Käfig für 2 Stunden wieder in den Inkubator. HINWEIS: Die erste Embryonensammlung wird verwendet, um Weibchen von befruchteten, sich entwickelnden Embryonen zu befreien, um eine eng getaktete zweite Sammlung von Embryonen zu erreichen.</li>
	<li>Entfernen Sie die Agarplatte aus dem Käfig und legen Sie sie (mit Hefeseite nach oben) auf ein feuchtes Papiertuch in einem verschließbaren Kunststoffbehälter. Legen Sie diese feuchte Kammer in einen 25 ° C-Inkubator, um die Embryonen für 14,5 h zu altern (Endalter, 14,5-16,5 h nach der Eiablage). HINWEIS: Führen Sie unmittelbar vor Beginn von Schritt 2.4 die Schritte 3.1 und 3.2 aus.</li>
	<li>Entfernen Sie die Agarplatte aus dem Inkubator und fügen Sie mit einer Spritzflasche genügend Wasser in die Agarplatte ein, um die Hefepaste aufzulösen, indem Sie sie leicht mit einem Pinsel bürsten. Spülen Sie die Embryonen und die gelöste Hefe in einen kleinen Netzsiebkorb, der in einem Wiegeboot sitzt. Spülen Sie den Korb mit der Wasserspritzflasche ab, bis die meiste Hefepaste durch das Netz gefiltert ist.</li>
	<li>Entfernen Sie das Wasser vom Wiegeboot und legen Sie den Korb wieder hinein. Dechorionieren Sie die Embryonen, indem Sie sie mit einer Spritzflasche in eine 50% ige Bleichlösung tauchen. Die Bleichlösung sollte eine Tiefe von 3–5 mm haben. Halten Sie die Embryonen für ~ 2 min in Bleichmittel getaucht, mit gelegentlichem Wirbeln. VORSICHT: Bleichmittel ist korrosiv und kann Augen und Atemwege reizen oder schädigen. Tragen Sie Handschuhe und Schutzbrillen, während Sie mit Bleichmittel umgehen. HINWEIS: Führen Sie während dieser Zeit Schritt 3.3 so weit wie möglich aus. Der Dechorionationsfortschritt kann überprüft werden, indem der Korb unter ein Stereomikroskop gestellt und auf das Fehlen der dorsalen Anhängsel überprüft wird. Tauchen Sie die Embryonen bei Bedarf für zusätzliche Zeit in die Bleichlösung.</li>
	<li>Entsorgen Sie das Bleichmittel und spülen Sie die Embryonen im Korb gründlich mit Wasser aus einer Spritzflasche aus (für ~ 3 s, den Netzkorb mit Papiertüchern abtupfen; 2-3 Mal wiederholen).</li>
</ol>3. Vorbereitung auf den Tag der Sezierung
<ol><li>30 μL Insulin (10 mg/ml) in 1.500 μL präparierte Bildgebungsmedien (siehe Schritt 1.2) in einem 1,5 ml Eppendorf-Röhrchen (0,2 mg/ml Endkonzentration) geben. Gut mischen und das Rohr auf der Tischplatte lassen, um es auf Raumtemperatur auszugleichen.</li>
	<li>Bereiten Sie mit Kleber beschichtete Deckglasstreifen vor.<ol>
<li>Verwenden Sie ein Messer mit Diamantspitze, um ein 22 mm x 22 mm großes Deckglas in vier gleich große Streifen zu schneiden (Abbildung 1A).</li>
		<li>Verwenden Sie eine Pinzette, um einen Streifen aufzunehmen und insgesamt etwa 30 μL Heptankleberlösung auf beiden Seiten des Bandes zu verteilen (Abbildung 1B). Um eine gleichmäßige Schicht aus Leimrückständen zu erreichen, neigen Sie den Streifen in verschiedenen Winkeln, während das Heptan verdunstet.</li>
		<li>Lagern Sie den mit Leim überzogenen Streifen in einem leeren Schlitz einer Deckglasschachtel; Um seine Klebrigkeit zu erhalten, platzieren Sie den Streifen in einer schrägen, aber aufrechten Position und lehnen Sie sich an die Kanten des Kastens, um den Kontakt mit den Kastenoberflächen zu minimieren (Abbildung 1C). Schließen Sie die Box, damit Partikel in der Luft den Kleber nicht beschichten und seine Klebrigkeit verringern.</li>
	</ol>
</li>
	<li>Legen Sie die folgenden Gegenstände auf die Tischplatte: eine 6-Zoll-Pasteur-Pipette aus Glas, einen Objektträger, eine P200- und eine P1000-Pipette mit den entsprechenden Pipettenspitzen, Ringer-Lösung27 (pH-Wert eingestellt auf 7,3 mit NaOH) und eine unbedeckte, Poly-D-Lysin-beschichtete 35-mm-Bildschale.</li>
	<li>Übertragen Sie Embryonen aus dem Netzsiebkorb in ein kleines Uhrenglas, das mit 500–750 μL Heptan gefüllt ist.<ol>
<li>Die Seiten und den Boden des Korbes mit einem Taschentuch trocken tupfen. Befeuchten Sie einen Pinsel im Heptan, berühren Sie den Pinsel mit den Embryonen (die hydrophobe Vitellinmembran sollte an den Borsten haften) und tauchen Sie den Pinsel wieder in das Heptan im Uhrenglas (Embryonen sollten zu Boden sinken). HINWEIS: Die nächsten Schritte müssen so schnell wie möglich durchgeführt werden, um ein Austrocknen der Embryonen zu verhindern. Embryonen sollten nicht länger als 20 s der Luft ausgesetzt werden.</li>
	</ol>
</li>
	<li>Die Embryonen werden mit einer Pasteur-Pipette auf den Objektträger übertragen (Abbildung 1D). Ziehen Sie Embryonen langsam in die Pipette und beschränken Sie sie auf den schmalen Teil der Pipette. Pipettenembryonen langsam auf den Objektträger, so dass sie sich direkt in der Spitze der Pipette aggregieren, bevor sie auf den Objektträger fließen. Drehen Sie die Ecke eines Gewebewischtuchs in eine feine Spitze und leiten Sie Heptan von Embryonen auf dem Objektträger ab. Sie aggregieren und bedecken eine kleinere Fläche, wodurch es einfacher wird, sie im nächsten Schritt auf einem mit Leim bedeckten Streifen zu erfassen.</li>
	<li>Nehmen Sie mit einer Pinzette einen mit Leim bedeckten Streifen auf und berühren Sie ihn vorsichtig mit den Embryonen (Abbildung 1E). Legen Sie den Streifen in die Bildgebungsschüssel, mit der Embryonenseite nach oben und direkt außerhalb des Poly-D-Lysin-beschichteten Zentrums. Drücken Sie den Streifen mit einer Pinzette auf die Schale, um sicherzustellen, dass er an Ort und Stelle fixiert ist. Sofort die Schale mit 2–3,5 ml Ringer-Lösung überfluten; Tauchen Sie zuerst in die Embryonen ein, um zu verhindern, dass sie austrocknen (Abbildung 1F).</li>
</ol>4. Sezieren
HINWEIS: Diese Schritte müssen unter einem stereofluoreszierenden Mikroskop durchgeführt werden.
<ol><li>Devitellinisieren Sie 10-15 Embryonen. HINWEIS: Dies kann von zwei Embryonen für Anfänger bis zu fünfzehn Embryonen für Experten reichen.<ol>
<li>Wählen Sie Embryonen im Stadium 16 basierend auf der Darmmorphologie aus (Abbildung 2). In diesem Stadium haben Embryonen drei Darmverengungen, die vier gestapelte Darmsegmente erzeugen (Abbildung 2 B, B'). Um mit der Devitellinisierung zu beginnen, durchbohren Sie den ausgewählten Embryo an einem Ende, vorzugsweise am vorderen Ende, mit der Wolframnadel. Der Embryo kann aus seiner Vitellinmembran herausspringen, aber wenn nicht, schälte die Membran vom Embryo ab. HINWEIS: Embryonen, die zu jung sind, um sie zu sezieren, haben nicht regionalisierte Eingeweide (Abbildung 2C). Die Dissektion von Embryonen im Stadium 17 ist möglich, aber schwieriger als die Dissektion von Embryonen im Stadium 16, da sich die Kutikula in diesem Stadium zu entwickeln beginnt. Embryonen im Frühstadium 17 mit vier Darmsegmenten, die relativ zueinander verschoben sind (Abbildung 2D,D').</li>
		<li>Haken Sie die Nadel durch den Embryo in einer Region, die weit von den Gonaden entfernt ist. Übertragen Sie den hakenförmigen Embryo in die mit Polylysin beschichtete Region der Schale (von hier an einfach als "Deckblatt" bezeichnet) und ziehen Sie ihn gegen den Boden, bis er anhaftet. Wiederholen Sie diese Schritte und ordnen Sie devitellinisierte Embryonen in einer Reihe entlang der Oberseite des mit Polylysin beschichteten Deckglases an (Abbildung 3A), so dass unten viel Platz für eine weitere Sezierung bleibt. HINWEIS: Die Gonaden erscheinen als kleine sphärische Aggregate von Zellen, die je nach verwendetem Marker und Transgenkopienzahl hell fluoreszieren sollten. In diesem Stadium befinden sich die Gonaden lateral im Segment A5, bei etwa 70% -80% der Embryonenlänge von vorne. Während des gesamten Dissektionsprotokolls kann das Gewebe an der Nadel haften bleiben. Um die Nadel von den Ablagerungen zu befreien, heben Sie sie direkt über die Oberfläche der Ringer-Lösung. Die Devitellinisierung kann unordentlich sein, solange die eigentliche Gonade so wenig wie möglich gestört wird.</li>
	</ol></li>
	<li>Finesse die Gonaden aus dem Embryo und auf die Schale (Abbildung 3C,D).<ol>
<li>Filetieren Sie zuerst den Embryo, um sein Inneres freizulegen (Abbildung 3C). Schneiden Sie den Embryo von seiner Mitte durch und bewegen Sie die Nadel nach hinten zwischen den Gonaden. Necken Sie etwas inneres Gewebe heraus, da dieses viel besser an der Schale haften bleibt als die äußere Kutikula. Die Klebrigkeit des Gewebes ermöglicht es den nächsten Manipulationen, die Gonade zu enthüllen und sie am Deckglas haften zu lassen. HINWEIS: Wenn das Gewebe nicht an der Schale haftet, versuchen Sie, es an einen nicht kontaminierten Bereich des beschichteten Deckglases zu locken. Die äußeren Bereiche des Deckglases werden besonders klebrig sein. Wie in Schritt 4.1.2 erwähnt, spielt es keine Rolle, ob die Dissektionsmanipulationen zu einem verstümmelten Embryokadaver führen, solange die Gonaden unversehrt bleiben.</li>
		<li>Verwenden Sie die Nadel, um eine Gonade zu schneiden, bis ein Stück Gewebe, einschließlich der Gonade, vom verbleibenden Kadaver getrennt ist. Ziehen Sie dieses Gewebe mit der Nadel in einen frischen Bereich aus beschichtetem Deckglas und locken Sie es gegen den Boden, bis es an der Schale haftet. HINWEIS: Die beste Bildgebung wird für die Fälle erreicht, in denen die Gonade selbst direkt am Deckglas befestigt wird, anstatt indirekt über darüber liegendes Gewebe befestigt zu werden. Versuchen Sie daher, wenn das Gewebe vom Kadaver weggeführt wird, dass die Gonade zuerst den Deckslip berührt, anstatt Fremdgewebe.</li>
		<li>Entfernen Sie so viel umgebendes Gewebe wie möglich aus der Gonade (Abbildung 3D), indem Sie das anhaftende Gewebe vorsichtig mit der Nadel von der Gonade wegziehen. Vermeiden Sie es, die Gonade direkt zu berühren, da sie beschädigt werden könnte (Abbildung 4E). Um sicherzustellen, dass die Gonade ausreichend an der Schale haftet, bewegen Sie die Nadel in einer sanften kreisförmigen Bewegung um die Gonade - wenn sie sich bewegt, berühren Sie die Nadel mit dem verbleibenden anhaftenden Gewebe und richten Sie die Gonade auf einen frischen Bereich aus klebrigem Deckglas. Wiederholen Sie diesen Vorgang, bis keine Bewegung der Gonade mehr erkennbar ist.</li>
		<li>Kehren Sie zum Embryokadaver zurück und sezieren Sie die zweite Gonade. Wiederholen Sie diesen Vorgang an so vielen Embryonen wie möglich, überschreiten Sie jedoch NICHT 25 Minuten. Die Lebensfähigkeit des Gewebes wird in Ringers Lösung nach mehr als ~ 40-45 Minuten beeinträchtigt.</li>
	</ol></li>
	<li>Nachdem die Dissektionen abgeschlossen sind, verwenden Sie einen unauslöschlichen Marker, um Registrierungsmarkierungen am äußeren Rand der Bildschale hinzuzufügen, um ihre Ausrichtung während der Dissektion aufzuzeichnen.</li>
	<li>Entfernen Sie den mit Leim beschichteten Streifen von der Schale.<ol>
<li>Führen Sie den unteren Stift einer Pinzette vorsichtig unter den Streifen ein, umschließen und neigen Sie den Streifen langsam nach oben, um ihn mit so wenig Schwappen der Ringerlösung wie möglich aus der Schale zu befreien, um die Störung der anhaftenden Gonaden zu minimieren. HINWEIS: Der Streifen sollte von den sezierten Gonaden weggeneigt werden.</li>
	</ol></li>
	<li>Tragen Sie die Schale vorsichtig zum bildgebenden Mikroskop, so dass ein Schwappen der Ringerlösung vermieden wird.</li>
</ol>5. Bildgebung
<ol><li>Legen Sie die abbildende Schale in den Bühnenhalter und verwenden Sie die Eintragungszeichen, um die Schale in der ungefähren Ausrichtung wie bei der Dissektion zu platzieren. Identifizieren und fokussieren Sie mit Hilfe der Hellfeldmikroskopie und eines Objektivs mit geringem Stromverbrauch (~ 10x) jedes Stück Gewebe, das auf dem Deckglas haftet. Schalten Sie die Okulareinstellungen um, um die Fluoreszenz anzuzeigen, und scannen Sie mit den binokularen Okularen systematisch die Schale und markieren Sie die Position jeder Gonade in der Bildgebungssoftware (siehe Materialtabelle). HINWEIS: Stellen Sie sicher, dass die Gonaden im Sichtfeld zentriert sind, bevor Sie Positionen markieren.</li>
	<li>Entfernen Sie vorsichtig die gesamte Bühnenhalterbaugruppe mit der abbildenden Schale in der Halterung, und legen Sie die Baugruppe auf die Werkbank.</li>
	<li>Ersetzen Sie die Lösung des Ringers durch die vorbereiteten bildgebenden Medien, die Insulin enthalten (siehe Schritte 1.2 und 3.1).<ol>
<li>Verwenden Sie einen P1000, um alle Ringer-Lösungen von der inneren oberen Leiste der Bildschale zu entfernen (pipettieren Sie Ringer nicht aus dem zentralen Deckglasbereich). Als nächstes wechseln Sie zu einem P200 und platzieren Sie seine Spitze direkt unter der Oberfläche der verbleibenden Ringer's in der zentralen Region. Entfernen Sie vorsichtig 50–100 μL Ringer's; Entfernen Sie NICHT die gesamte Lösung.</li>
		<li>Ziehen Sie ~ 200 μL Bildmedien auf und platzieren Sie die P200-Spitze wieder direkt unter der Oberfläche der verbleibenden Ringer-Medien, fügen Sie diese Bildgebungsmedien langsam hinzu. Als nächstes fügen Sie die verbleibenden Bildmedien (~ 1.300 μL) in die Schale hinzu, beginnend am äußersten Rand der oberen Kante. Bewegen Sie sich beim Pipettieren in Richtung der zentralen Kuppel der Flüssigkeit und verschmelzen Sie schließlich die beiden, indem Sie die Pipettenspitze über beide streichen. Legen Sie den Deckel auf die Schüssel. HINWEIS: Bildgebende Medien sollten den gesamten Innendurchmesser der Schale mit einer Tiefe von etwa 2 mm abdecken, um eine Verdunstung zu verhindern (Abbildung 4D).</li>
	</ol>
</li>
	<li>Schalten Sie das Mikroskop auf ein Objektiv mit höherer Leistung (63x, 1,2 NA) um, wenden Sie die richtige Tauchflüssigkeit basierend auf dem verwendeten Objektiv an (Tauchflüssigkeitstyp und Brechungsindex für das Objektiv erforderlich) und ersetzen Sie dann die Tischhalterbaugruppe. Verwenden Sie die Hellfeldmikroskopie, um sich auf das Gewebe zu konzentrieren, das am Boden der Bildgebungsschale haftet. HINWEIS: Wenn die Bühne nicht bewegt wurde, sollte die letzte Gonade in Sicht kommen, sobald das Ziel fokussiert ist.</li>
	<li>Gehen Sie durch jede markierte Gonadenposition und passen Sie diese Position bei Bedarf an. Wählen Sie aus, welche Gonaden basierend auf Bildklarheit, Gonadengeschlecht usw. abgebildet werden sollen. Passen Sie die Bildgebungseinstellungen an (Mehrkanal, Belichtungszeiten, Laserintensitäten, Inkremente der Z-Serie, Zeitraffer mit geeigneten Intervallen usw.). Beginnen Sie mit der Bildgebung. HINWEIS: Männliche Gonaden können durch das Vorhandensein sowohl einer Nische als auch am gegenüberliegenden Ende der Gonade identifiziert werden, einem Cluster kleiner, stark kreisförmiger somatischer Zellen, die als männlich-spezifische somatische Gonadenvorläufer (msSGPs) bezeichnet werden. Wenn der Fluoreszenzmarker six4-eGFP::moesin verwendet wird, sind die Nischenzellen die zweithellsten Zellen in der Gonade, wobei die hellsten die msSGPs sind. In diesem Stadium haben weibliche Gonaden weder eine Nische noch msSGPs.</li>

Die Drosophila melanogaster testis hat als Paradigma für unser Verständnis vieler dynamischer zellulärer Prozesse gedient. Studien dieses Modells haben Aufschluss über die Stammzellteilungsregulation 1,2,3, die Keimzellentwicklung4,5, die piRNA-Biologie 6,7,8 und die Nischenstammzell-Signalisierungsereignisse 9,10,11,12,13 gegeben. Dieses Modell ist vorteilhaft, weil es genetisch behandelbar ist14,15 und eines der wenigen ist, bei denen wir Stammzellen in ihrer natürlichen Umgebung live darstellenkönnen 3,16,17,18. Die Live-Bildgebung dieses Modells war jedoch auf adultes Gewebe und frühe Embryonalstadien beschränkt, was eine Lücke in unserem Wissen über die Gonadendynamik im späten Embryo hinterlässt, dem genauen Stadium, in dem sich die Nische zum ersten Mal bildet und zu funktionieren beginnt.
Die embryonale Gonade im Spätstadium ist eine Kugel, die aus somatischen Nischenzellen im vorderen Bereich und Keimzellen besteht, die von somatischen Gonadenzellen in hintereren Regioneneingestreut werden 19. Dieses Organ kann live in vivo bis zum frühen Embryonalstadium 17 17,20,21 abgebildet werden. Eine weitere Bildgebung wird durch die Einleitung großflächiger Muskelkontraktionen verhindert. Diese Kontraktionen sind so stark, dass sie die Gonade aus dem Bildgebungsrahmen drücken, und eine solche Bewegung kann nicht mit einer Bildgebungssoftware korrigiert werden. Unser Labor ist daran interessiert, die Mechanismen der Nischenbildung aufzudecken, die während dieser schwer fassbaren Zeit für die Live-Bildgebung auftritt. Daher haben wir einen Ex-vivo-Ansatz entwickelt, um die Gonade ab dem Embryonalstadium 16 live abzubilden, was die Untersuchung der Zelldynamik während dieser entscheidenden Phase der Gonadenentwicklung erleichtert. Frühere Arbeiten aus unserem Labor zeigen, dass diese Ex-vivo-Bildgebung in vivo Gonadenentwicklung originalgetreu rekapituliert17. Diese Technik ist die erste und einzige ihrer Art für die embryonale Gonade Drosophila.
Hier stellen wir das Dissektionsprotokoll vor, das für die Ex-vivo-Live-Bildgebung der Gonade in späten Embryonalstadien erforderlich ist. Dieses Protokoll kann mit pharmakologischen Behandlungen oder transgener Manipulation spezifischer Zelllinien innerhalb der Gonade kombiniert werden. Mit dieser Technik haben wir die Schritte der Stammzellnischenbildung erfolgreich abgebildet17. Dieser bildgebende Ansatz ist daher für das Gebiet der Stammzellbiologie von entscheidender Bedeutung, da er die Visualisierung der Anfangsstadien der Nischenbildung in Echtzeit in seiner natürlichen Umgebung ermöglicht15,17. Während diese Methode für das Gebiet der Stammzellbiologie von Vorteil ist, ist sie zusätzlich anwendbar, um dynamische Prozesse zu visualisieren, die während dieses Entwicklungszeitpunkts in der Gonade auftreten, einschließlich zellulärer Umlagerungen 22, Zelladhäsion2, 12, 23 und Zellmigration23. Large-scale Zebrafish Embryonic Heart Dissection for Transcriptional Analysis

The zebrafish embryonic heart is composed of only a few hundred cells, representing only a small fraction of the entire embryo. Therefore, to prevent the cardiac transcriptome from being masked by the global embryonic transcriptome, it is necessary to collect sufficient numbers of hearts for further analyses. Furthermore, as zebrafish cardiac development proceeds rapidly, heart collection and RNA extraction methods need to be quick in order to ensure homogeneity of the samples. Here, we present a rapid manual dissection protocol for collecting functional/beating hearts from zebrafish embryos. This is an essential prerequisite for subsequent cardiac-specific RNA extraction to determine cardiac-specific gene expression levels by transcriptome analyses, such as quantitative real-time polymerase chain reaction (RT-qPCR). The method is based on differential adhesive properties of the zebrafish embryonic heart compared with other tissues; this allows for the rapid physical separation of cardiac from extracardiac tissue by a combination of fluidic shear force disruption, stepwise filtration and manual collection of transgenic fluorescently labeled hearts.

Live-imaging of the Drosophila Pupal Eye

Inherent processes of Drosophila pupal development can shift and distort the eye epithelium in ways that make individual cell behavior difficult to track during live cell imaging. These processes include: retinal rotation, cell growth and organismal movement. Additionally, irregularities in the topology of the epithelium, including subtle bumps and folds often introduced as the pupa is prepared for imaging, make it challenging to acquire in-focus images of more than a few ommatidia in a single focal plane. The workflow outlined here remedies these issues, allowing easy analysis of cellular processes during Drosophila pupal eye development. Appropriately-staged pupae are arranged in an imaging rig that can be easily assembled in most laboratories. Ubiquitin-DE-Cadherin:GFP and GMR-GAL4&#8211;driven UAS-&#945;-catenin:GFP are used to visualize cell boundaries in the eye epithelium 1-3. After deconvolution is applied to fluorescent images captured at multiple focal planes, maximum projection images are generated for each time point and enhanced using image editing software. Alignment algorithms are used to quickly stabilize superfluous motion, making individual cell behavior easier to track.

Dissection and Culture of Mouse Embryonic Kidney

The goal of this protocol is to describe a method for the dissection, isolation, and culture of mouse metanephric rudiments. 
During mammalian kidney development, the two progenitor tissues, the ureteric bud and the metanephric mesenchyme, communicate and reciprocally induce cellular mechanisms to eventually form the collecting system and the nephrons of the kidney. As mammalian embryos grow intrauterine and therefore are inaccessible to the observer, an organ culture has been developed. With this method, it is possible to study epithelial-mesenchymal interactions and cellular behavior during kidney organogenesis. Furthermore, the origin of congenital kidney and urogenital tract malformations can be investigated. After careful dissection, the metanephric rudiments are transferred onto a filter that floats on culture medium and can be kept in a cell culture incubator for several days. However, one must be aware that the conditions are artificial and could influence the metabolism in the tissue. Also, the penetration of test substances could be limited due to the extracellular matrix and basal membrane present in the explant.
One main advantage of organ culture is that the experimenter can gain direct access to the organ. This technology is cheap, simple, and allows a large number of modifications, such as the addition of biologically active substances, the study of genetic variants, and the application of advanced imaging techniques.

Long-term Live Imaging of Drosophila Eye Disc

Live imaging provides the ability to continuously track dynamic cellular and developmental processes in real time. Drosophila larval imaginal discs have been used to study many biological processes, such as cell proliferation, differentiation, growth, migration, apoptosis, competition, cell-cell signaling, and compartmental boundary formation. However, methods for the long-term ex vivo culture and live imaging of the imaginal discs have not been satisfactory, despite many efforts. Recently, we developed a method for the long-term ex vivo culture and live imaging of imaginal discs for up to 18 h. In addition to using a high insulin concentration in the culture medium, a low-melting agarose was also used to embed the disc to prevent it from drifting during the imaging period. This report uses the eye-antennal discs as an example. Photoreceptor R3/4-specific m&#948;0.5-Ga4 expression was followed to demonstrate that photoreceptor differentiation and ommatidial rotation can be observed during a 10 h live imaging period. This is a detailed protocol describing this simple method.

Bioengineering of Humanized Bone Marrow Microenvironments in Mouse and Their Visualization by Live Imaging

Human hematopoietic stem cells (HSCs) reside in the bone marrow (BM) niche, an intricate, multifactorial network of components producing cytokines, growth factors, and extracellular matrix. The ability of HSCs to remain quiescent, self-renew or differentiate, and acquire mutations and become malignant depends upon the complex interactions they establish with different stromal components. To observe the crosstalk between human HSCs and the human BM niche in physiological and pathological conditions, we designed a protocol to ectopically model and image a humanized BM niche in immunodeficient mice. We show that the use of different cellular components allows for the formation of humanized structures and the opportunity to sustain long-term human hematopoietic engraftment. Using two-photon microscopy, we can live-image these structures in situ at the single-cell resolution, providing a powerful new tool for the functional characterization of the human BM microenvironment and its role in regulating normal and malignant hematopoiesis.

Live Imaging of Mouse Secondary Palate Fusion

The fusion of the secondary palatal shelves to form the intact secondary palate is a key process in mammalian development and its disruption can lead to cleft secondary palate, a common congenital anomaly in humans. Secondary palate fusion has been extensively studied leading to several proposed cellular mechanisms that may mediate this process. However, these studies have been mostly performed on fixed embryonic tissues at progressive timepoints during development or in fixed explant cultures analyzed at static timepoints. Static analysis is limited for the analysis of dynamic morphogenetic processes such a palate fusion and what types of dynamic cellular behaviors mediate palatal fusion is incompletely understood. Here we describe a protocol for live imaging of ex vivo secondary palate fusion in mouse embryos. To examine cellular behaviors of palate fusion, epithelial-specific Keratin14-cre was used to label palate epithelial cells in ROSA26-mTmGflox reporter embryos. To visualize filamentous actin, Lifeact-mRFPruby reporter mice were used. Live imaging of secondary palate fusion was performed by dissecting recently-adhered secondary palatal shelves of embryonic day (E) 14.5 stage embryos and culturing in agarose-containing media on a glass bottom dish to enable imaging with an inverted confocal microscope. Using this method, we have detected a variety of novel cellular behaviors during secondary palate fusion. An appreciation of how distinct cell behaviors are coordinated in space and time greatly contributes to our understanding of this dynamic morphogenetic process. This protocol can be applied to mutant mouse lines, or cultures treated with pharmacological inhibitors to further advance understanding of how secondary palate fusion is controlled.

Here, we present a protocol for live imaging of mouse secondary palate fusion using confocal microscopy. This protocol can be used in combination with a variety of fluorescent reporter mouse lines, and with pathway inhibitors for mechanistic insight. Nervous system development involves a sequential series of events that are coordinated by several signaling pathways and regulatory networks. Many of the proteins involved in these pathways are evolutionarily conserved between mammals and other eukaryotes, such as the fruit fly Drosophila melanogaster, suggesting that similar organizing principles exist during the development of these organisms. Importantly, Drosophila has been used extensively to identify cellular and molecular mechanisms regulating processes that are required in mammals including neurogenesis, differentiation, axonal guidance, and synaptogenesis. Flies have also been used successfully to model a variety of human neurodevelopmental diseases. Here we describe a protocol for the step-by-step microdissection, fixation, and immunofluorescent localization of proteins within the adult Drosophila brain. This protocol focuses on two example neuronal populations, mushroom body neurons and retinal photoreceptors, and includes optional steps to trace individual mushroom body neurons using Mosaic Analysis with a Repressible Cell Marker (MARCM) technique. Example data from both wild-type and mutant brains are shown along with a brief description of a scoring criteria for axonal guidance defects. While this protocol highlights two well-established antibodies for investigating the morphology of mushroom body and photoreceptor neurons, other Drosophila brain regions and the localization of proteins within other brain regions can also be investigated using this protocol.

Dissection and Staining of Drosophila Pupal Ovaries

Unlike adult Drosophila ovaries, pupal ovaries are relatively difficult to access and examine due to their small size, translucence, and encasing within a pupal case. The challenge of dissecting pupal ovaries also lies in their physical location within the pupa: the ovaries are surrounded by fat body cells inside the pupal abdomen, and these fat cells must be removed to allow for proper antibody staining. To overcome these challenges, this protocol utilizes customized Pasteur pipets to extract fat body cells from the pupal abdomen. Moreover, a chambered coverglass is used in place of a microcentrifuge tube during the staining process to improve visibility of the pupae. However, despite these and other advantages of the tools used in this protocol, successful execution of these techniques may still involve several days of practice due to the small size of pupal ovaries. The techniques outlined in this protocol could be applied to time course experiments in which ovaries are analyzed at various stages of pupal development.

Dissection of the Drosophila Pupal Retina for Immunohistochemistry, Western Analysis, and RNA Isolation

The Drosophila pupal retina provides an excellent model system for the study of morphogenetic processes during development. In this paper, we present a reliable protocol for the dissection of the delicate Drosophila pupal retina. Our surgical approach utilizes readily-available microdissection tools to open pupae and precisely extract eye-brain complexes. These can be fixed, subjected to immunohistochemistry, and retinas then mounted onto microscope slides and imaged if the goal is to detect cellular or subcellular structures. Alternatively, unfixed retinas can be isolated from brain tissue, lysed in appropriate buffers and utilized for protein gel electrophoresis or mRNA extraction (to assess protein or gene expression, respectively). Significant practice and patience may be required to master the microdissection protocol described, but once mastered, the protocol enables relatively quick isolation of mainly undamaged retinas.

Live Imaging and Analysis of Muscle Contractions in Drosophila Embryo

Coordinated muscle contractions are a form of rhythmic behavior seen early during development in Drosophila embryos. Neuronal sensory feedback circuits are required to control this behavior. Failure to produce the rhythmic pattern of contractions can be indicative of neurological abnormalities. We previously found that defects in protein O-mannosylation, a posttranslational protein modification, affect the axon morphology of sensory neurons and result in abnormal coordinated muscle contractions in embryos. Here, we present a relatively simple method for recording and analyzing the pattern of peristaltic muscle contractions by live imaging of late stage embryos up to the point of hatching, which we used to characterize the muscle contraction phenotype of protein O-mannosyltransferase mutants. Data obtained from these recordings can be used to analyze muscle contraction waves, including frequency, direction of propagation and relative amplitude of muscle contractions at different body segments. We have also examined body posture and taken advantage of a fluorescent marker expressed specifically in muscles to accurately determine the position of the embryo midline. A similar approach can also be utilized to study various other behaviors during development, such as embryo rolling and hatching.
