Cannulation is the most fundamental procedure for establishing DHCA in rats. Before cannulation, soaking the artery with 0.5 mL of 2% lidocaine will make it easier to cannulate. After cannulation, heparinization with 500 IU/kg heparin via the external jugular vein is necessary to avoid microthrombus formation17. Deep hypothermic circulatory arrest (DHCA) has been used in cardiac surgery since 19531. DHCA involves reducing the patient&#39;s core temperature to profoundly hypothermic levels (15-22 &#176;C) before globally interrupting the blood flow to the body2. The circulatory arrest can provide a relatively bloodless operating field. Deep hypothermia decreases the metabolism, especially in the brain and myocardium, which is an effective method of protection against ischemia3. DHCA is commonly applied during surgeries for complex congenital heart disease, aortic arch disease, and even renal or adrenal tumors with a vena cava thrombus4,5. Therefore, establishing DHCA animal models provides an important reference for the refinement of the procedure and the prevention of complications in clinical settings.
Although models can be established with canines6, rabbits7, and other animals, it is preferable to use rats because of their operability and low cost. The DHCA rat model was described for the first time in 2006 by Jungwirth et al.8. It was found that the duration of circulatory arrest had an impact on the neurologic outcomes. Since then, DHCA rat models have been investigated broadly. It has been clarified that DHCA could provoke systemic inflammatory response syndrome (SIRS)9. In subsequent studies, pharmacologists found that the DHCA-related neuroinflammation induced by SIRS could be attenuated by resveratrol10 and triptolide11. Our team also found that DHCA-related neuroinflammation could be attenuated by inhibiting the cold-inducible RNA-binding protein12. In the cardiovascular system, superoxide dismutase has a cardioprotective effect on ischemia/reperfusion (I/R) injuries during DHCA13. These results expanded the understanding of DHCA-related pathophysiologic processes and offered new directions for improving the outcomes of DHCA. However, the results regarding endotoxemia, oxidative stress, and autophagy after DHCA are inconclusive. DHCA uses the same operational technology as the cardiopulmonary bypass (CPB)14, but its management strategy is different, and the steps to generate DHCA differ across various teams8,9,10,11. The present study aims to provide a method for establishing the DHCA procedure in rats.

Deep hypothermic circulatory arrest (DHCA) is routinely applied during surgeries for complex congenital heart disease and aortic arch disease. The present study aims to provide a method for establishing DHCA in rats. To evaluate the impact of the DHCA process on vital signs, a normal temperature cardiopulmonary bypass (CPB) rat model without circulatory arrest was used as a control. As expected, DHCA led to a significant decrease in body temperature and mean arterial blood pressure. The blood gas analysis indicated that DHCA increased lactic acid levels but did not influence the blood pH and the concentrations of hemoglobin, hematocrit, Na+, Cl&#8722;, K+, and glucose. Furthermore, compared with the normal temperature CPB rats, the results of the transmission electron microscopy showed a mild increase in hippocampal autophagosomes in the DHCA rats.

As the control group, the normal temperature CPB (NtCPB) rats without circulatory arrest showed a stable mean arterial blood pressure (MAP) and body temperature during the whole procedure, while the MAP of the DHCA rats decreased during the cardiac arrest (p &#60; 0.01, Figure 3A). Deep hypothermic circulatory arrest, DHCA is routinely applied during surgeries of complex congenital heart disease and aortic arch disease. The present study aims to provide a method for establishing DHCA in rats.The protocols received an institutional review and got approval from the Institutional Animal Care and Use Committee, Fuwai Hospital, Chinese Academy of Medical Sciences. Preparatory work. Ensure CPB circuits contain a reservoir modified from morphs dropper, roller pump, heat exchanger, membrane oxygenator, connecting tube, and a water tank connected the circuit and makes 12 milliliter of hydroxy starch with one milliliter heparin sodium and one milliliter saline prime the circuit with 40 milliliter priming solution with the roller pump gently rotating.Two, anesthesia and cannulation. Connect the tube with the ventilator and side parameters. Put an electric heating blanket and the right and face the right with tape.Apply ophthalmic ointment to the eyes to prevent dryness. Shave the hair on the right inguinal region, right cervical region and the tail with the shaver. Then disinfect the skin with aldehyde and alcohol.Cut the skin and the left inguinal region and dissect the muscle and tissue soft bone to expose the left femoral vein and artery. Separate the artery carefully. Cannulate 22 gauge intravenous catheter into the left femoral artery.Ligated artery and catheter with silk. Use saline containing heparin to flush the cannula to avoid clotting. Connect the catheter with the press sensor to monitor blood pressure.Cut the skin of the tail and then use a scalpel to cut the superficial specia of the tail artery to expose the tail artery which is in the middle of the surgical field. Cannulate the tail artery with the 22 gauge intravenous catheter. Ligate artery and catheter with silk.Use saline containing heparin to flush the catheter to avoid clotting. Cut the skin of the right jugular of vein then separate muscle and tissue to expose the vein. Insert a 16 gauge homemade multiage intravenous catheter into the right external jugular of vein and put it into the right inferior vena cava or the right atrium carefully.Administrate heparin sodium where the right external vein cover each cannulated region with moist balls to avoid contamination. Three, connect the circuit with the catheter in the tail artery and keep the pump low rate one to two milliliter per minute. Then connect the reservoir with the catheter in the right external jugular vein.Make sure there is always a blood level of about one centimeter in the reservoir. Turn on the water tank and set the water temperature at 37 centigrade first. After the blood pressure is stable, gently increase the pump of flow up to 80 to 100 milliliter per minute.Four, cooling. Set the room temperature to around 20 centigrade. Put ice cubes in this disposable gloves and then put them on the right head and sides.The temperature of the tank was suggested to real time according to the rectal temperature of the rice. Change the relevant parameters of the ventilator properly according to the results of blood gas analysis. Five, deep hypothermia circulatory arrest.Then the rectal temperature drop to 15 to 20th centigrade. Stop the roller pump.Keep the reservoir in contact with the environment and the drain the blood slowly from the external jugular vein to the reservoir. Pay attention to the blood pressure reform. After there is no blood pressure and heartbeat, stop the drainage and keep the reservoir closed.Turn off the ventilator. Six, warm-up and reperfusion. Remove all the disposable gloves and increase the room temperature to 25 centigrade.Restore the memory oxygen inter ventilation while keeping the venous drainage tube clipping. Turn on the roller pump to make sure the blood in the reservoir is slowly back to the right body. Turn on the ventilator after blood level in the reservoir remain one centimeter.Using the drainage tube and drain the blood from the right atrium to the reservoir slowly. Turn on the heating lamp, the heating pad and the water tank. Set the temperature of water tank to 25 centigrade.Then adjust it to left temperature timely. Remove the heating lamp after the rectal temperature returned to 34 centigrade. Seven, weaning off CPB.Slowly and gradually reduce the roller of pump flow rate and adjust the vena genus speed into the flow rate reduce to one milliliter per minute. Keep the reservoir in contact with the environment. Infuse the remaining blood in a circuit with a flow rate of one milliliter per minute.Stop the memory oxygenation and the roller pump. Euthanized after a period of mechanical ventilation and a deep anesthesia. Representative results.As the control group, the normal temperature CPB, NtCPB, rat. Without the circulatory arrest showed a stable mean artery blood pressure MAP and body temperature during the whole procedure. While the MAP of DHCA rats decreased during the cardiac arrest.The temperature of DHCA rats dropped fast during the cooling phase and recovered gradually during the revolving phase. The weaning of DHCA circuit the temperature of DHCA rats returned to normal. The above data indicated that our DHCA procedure imitated the actual temperature and MAP changes of patients in clinics.The effect of DHCA processor and rise was investigated by blood gas analysis. After the whole blood contact with the primary solution, the concentration of hemoglobin was higher than six gram per decilitter in both groups. The weaning off the DHCA surface, it increased to nine gram per decilitter because of the reinfusing of remaining blood in the CPB circuit into the right.The hematocrit, HCT, showed the similar tendency to HB.After DHCA reperfusion lactate acid increased fast which was more pronounced in the DHCA group. pH decreased after the DHCA procedure, which was most likely the result of lactate acid accumulation. During the whole experiment, the concentration of sodium, chloride, potassium ions, and the glucose did not show significant differences at each 10 point.This result suggests that DHCA only induced the increased to lactate acid but did not influence the blood pH and the concentration of hemoglobin, hematocrit, sodium ions, chloride ions, potassium ions, and glucose. In order to evaluate the impact of DHCA on neuronal autophagy, we used the transmission electro microscope and surprisingly found and increased the number of autophagosomes in the hippocampus of DHCA rats. The above data indicated that our DHCA model imitated actual temperature and MAP changes of patients in the clinics.This model can be applied to investigate systemic inflammatory response syndrome, neuroinflammation, et cetera.

Composite tissue allotransplantation (CTA) now represents a valid therapeutic option after the loss of a hand, forearm or digits and has become a novel therapeutic entity in reconstructive surgery. However, long term high-dose multi-drug immunosuppressive therapy is required to ensure graft survival, bearing the risk of serious side effects which halters broader application. Further progression in this field may depend on better understanding of basic immunology and ischemia reperfusion injury in composite tissue grafts.
To date, orthotopic hind limb transplantation in rats has been the preferred rodent model for reconstructive transplantation (RT), however, it is an extremely demanding procedure that requires extraordinary microsurgical skills for reattachment of vasculature, bones, muscles and nerves.
We have introduced the vascular cuff anastomosis technique to this model, providing a rapid and reliable approach to rat hind limb transplantation. This technique simplifies and shortens the surgical procedure and enables surgeons with basic microsurgical experience to successfully perform the operation with high survival and low complication rates. The technique seems to be well suited for immunological as well as ischemia reperfusion injury (IRI) studies.

Diagnosis of Ecto- and Endoparasites in Laboratory Rats and Mice

Internal and external parasites remain a significant concern in laboratory rodent facilities, and many research facilities harbor some parasitized animals. Before embarking on an examination of animals for parasites, two things should be considered. One: what use will be made of the information collected, and two: which test is the most appropriate. Knowing that animals are parasitized may be something that the facility accepts, but there is often a need to treat animals and then to determine the efficacy of treatment. Parasites may be detected in animals through various techniques, including samples taken from live or euthanized animals. Historically, the tests with the greatest diagnostic sensitivity required euthanasia of the animal, although PCR has allowed high-sensitivity testing for several types of parasite. This article demonstrates procedures for the detection of endo- and ectoparasites in mice and rats. The same procedures are applicable to other rodents, although the species of parasites found will differ.

This article describes various procedures for screening rats and mice to detect endo- or ectoparasitism. Several diagnostic assays will be demonstrated, both those suitable for use on live animals and those used after euthanasia of the animal. Establishment of Epstein-Barr Virus Growth-transformed Lymphoblastoid Cell Lines

Infection of B cells with Epstein-Barr virus (EBV) leads to proliferation and subsequent immortalization, resulting in establishment of lymphoblastoid cell lines (LCL) in vitro. Since LCL are latently infected with EBV, they provide a model system to investigate EBV latency and virus-driven B cell proliferation and tumorigenesis1. LCL have been used to present antigens in a variety of immunologic assays2, 3. In addition, LCL can be used to generate human monoclonal antibodies4, 5 and provide a potentially unlimited source when access to primary biologic materials is limited6, 7.
A variety of methods have been described to generate LCL. Earlier methods have included the use of mitogens such as phytohemagglutinin, lipopolysaccharide8, and pokeweed mitogen9 to increase the efficiency of EBV-mediated immortalization. More recently, others have used immunosuppressive agents such as cyclosporin A to inhibit T cell-mediated killing of infected B cells7, 10-12.
The considerable length of time from EBV infection to establishment of cell lines drives the requirement for quicker and more reliable methods for EBV-driven B cell growth transformation. Using a combination of high titer EBV and an immunosuppressive agent, we are able to consistently infect, transform, and generate LCL from B cells in peripheral blood. This method uses a small amount of peripheral blood mononuclear cells that are infected in vitroclusters of cells can be demonstrated. The presence of CD23 with EBV in the presence of FK506, a T cell immunosuppressant. Traditionally, outgrowth of proliferating B cells is monitored by visualization of microscopic clusters of cells about a week after infection with EBV. Clumps of LCL can be seen by the naked eye after several weeks. We describe an assay to determine early if EBV-mediated growth transformation is successful even before microscopic clusters of cells can be demonstrated. The presence of CD23hiCD58+ cells observed as early as three days post-infection indicates a successful outcome.

Establishment of an In vitro System to Study Intracellular Behavior of Candida glabrata in Human THP-1 Macrophages

A cell culture model system, if a close mimic of host environmental conditions, can serve as an inexpensive, reproducible and easily manipulatable alternative to animal model systems for the study of a specific step of microbial pathogen infection. A human monocytic cell line THP-1 which, upon phorbol ester treatment, is differentiated into macrophages, has previously been used to study virulence strategies of many intracellular pathogens including Mycobacterium tuberculosis. Here, we discuss a protocol to enact an in vitro cell culture model system using THP-1 macrophages to delineate the interaction of an opportunistic human yeast pathogen Candida glabrata with host phagocytic cells. This model system is simple, fast, amenable to high-throughput mutant screens, and requires no sophisticated equipment. A typical THP-1 macrophage infection experiment takes approximately 24 hr with an additional 24-48 hr to allow recovered intracellular yeast to grow on rich medium for colony forming unit-based viability analysis. Like other in vitro model systems, a possible limitation of this approach is difficulty in extrapolating the results obtained to a highly complex immune cell circuitry existing in the human host. However, despite this, the current protocol is very useful to elucidate the strategies that a fungal pathogen may employ to evade/counteract antimicrobial response and survive, adapt, and proliferate in the nutrient-poor environment of host immune cells.

Complete Thymectomy in Adult Rats with Non-invasive Endotracheal Intubation

Thymectomy in neonatal rodents is an established and reliable procedure for immunological studies. However, in adult rats, complications of hemorrhage and pneumothorax from pleural disruption can result in a significant mortality rate. This protocol is a simple method of rat thymectomy that utilizes a mini-sternotomy and endotracheal intubation. Intubation is accomplished with a non-invasive and easily reproducible method and allows for positive pressure ventilation to prevent pneumothorax and a controlled airway that allows sufficient time for careful thymus dissection to minimize pleural disruption. A 1.5 cm sternal incision decreases contact with mediastinal vessels and pleura, while still providing full visualization of the thymus. Following exposure of the mediastinum, the thymus is removed by blunt dissection under magnification. The pleural space is then sealed by suture closure of the pre-tracheal muscles followed by the application of surgical glue. The thorax is then closed by suture closure of the sternum, followed by suture closure of the skin. All thymectomies were complete as evidenced by immunohistochemical (IHC) staining of mediastinal tissue, and absence of na&#239;ve T-cells by flow cytometry, and the procedure had a 96% survival rate. This method is suitable when complete thymectomy with minimal complications is desired for further immunological studies in athymic adult rats.

Prevention of Heat Stress Adverse Effects in Rats by Bacillus subtilis Strain

This study was designed to evaluate the protective effect of the Bacillus subtilis strain against complications related to heat stress. Thirty-two Sprague-Dawley rats were used in this study. Animals were orally treated twice a day for two days with B. subtilis BSB3 strain or PBS. The next day after the last treatment, each group was divided and two experimental groups (one treated with PBS and one treated with B. subtilis) were placed at 45 oC for 25 min. Two control groups stayed for 25 min at room temperature. All rats were euthanized and different parameters were analyzed in all groups. Adverse effects of heat stress are registered by the decrease of villi height and total mucosal thickness in the intestinal epithelium; translocation of bacteria from the lumen; increased vesiculation of erythrocytes and elevation of the lipopolysaccharides (LPS) level in the blood. The protective efficacy of treatment is evaluated by prevention of these side effects. The protocol was set up for the oral treatment of rats with bacteria for prevention of heat stress complications, but this protocol can be modified and used for other routes of administration and for analysis of different compounds.

Stenosis of the Inferior Vena Cava: A Murine Model of Deep Vein Thrombosis

Deep vein thrombosis (DVT) and its devastating complication, pulmonary embolism, are a severe health problem with high mortality. Mechanisms of thrombus formation in veins remain obscure. Lack of mobility (e.g., after surgery or long-haul flights) is one of the main factors leading to DVT. The pathophysiological consequence of the lack of mobility is blood flow stagnation in venous valves. Here, a model is described that mimics such flow disturbance as a thrombosis-driving factor. In this model, partial flow restriction (stenosis) in the inferior vena cava (IVC) is created. Closure of about 90% of the IVC lumen for 48 h results in development of thrombi structurally similar to those in humans. The similarities are: i) most of the thrombus volume is red, i.e., consists of red blood cells and fibrin, ii) presence of a white part (lines of Zahn), iii) non-denuded endothelial monolayer, iv) elevated plasma D-Dimer levels, and v) possibility to prevent thrombosis by low molecular weight heparin. Limitations include variable size of thrombi and the fact that a certain percentage of wild-type mice (0 - 35%) may not produce a thrombus. In addition to visual observation and measurement, thrombi may be visualized by non-invasive technologies, such as ultrasonography, which allows for monitoring the dynamics of thrombus development. At shorter time points (1 - 6 h), intravital microscopy may be applied to directly observe events (e.g., recruitment of cells to the vessel wall) preceding thrombus formation. Use of this method by several teams around the world has made it possible to uncover basic mechanisms of DVT initiation and identify potential targets that might be beneficial for its prevention.

Deep Vein Thrombosis Induced by Stasis in Mice Monitored by High Frequency Ultrasonography

Venous thrombosis is a common condition affecting 1 - 2% of the population, with an annual incidence of 1 in 500. Venous thrombosis can lead to death through pulmonary embolism or results in the post-thrombotic syndrome, characterized by chronic leg pain, swelling, and ulceration, or in chronic pulmonary hypertension resulting in significant chronic respiratory compromise. This is the most common cardiovascular disease after myocardial infarction and ischemic stroke and is a clinical challenge for all medical disciplines, as it can complicate the course of other disorders such as cancer, systemic disease, surgery, and major trauma.
Experimental models are necessary to study these mechanisms. The stasis model induces consistent thrombus size and a quantifiable amount of thrombus. However, it is necessary to systematically ligate side branches of the inferior vena cava to avoid variability in thrombus sizes and any erroneous data interpretation. We have developed a non-invasive technique to measure thrombus size using ultrasonography. Using this technique, we can assess thrombus development and resolution over time in the same animal. This approach limits the number of mice required for quantification of venous thrombosis consistent with the principle of replacement, reduction, and refinement of animals in research. We have demonstrated that thrombus weight and histological analysis of thrombus size correlate with measurement obtained with ultrasonography. Therefore, the current study describes how to induce deep vein thrombosis in mice using the inferior vena cava stasis model and how to monitor it using high frequency ultrasound.

Deep Dermal Injection As a Model of Candida albicans Skin Infection for Histological Analyses

The skin is an extremely extended organ of the body and, due to this large surface, it is continuously exposed to microorganisms. Skin damage can easily lead to infections in the dermis which can, in turn, result in the dissemination of pathogens into the bloodstream. Understanding how the immune system fights infections at the very early stage and how the host can eliminate the pathogens is an important step to set the base for future therapeutic interventions. Here we describe a model of Candida albicans infection that can visualize the processes that occur early during an infection, including when the pathogen has passed the epithelial barrier, as well as the immune response elicited by the C. albicans invasion. We used this infection model to perform histological analyses that show the immune cells that infiltrate the skin as well as the presence and localization of the pathogen. Samples collected after the infection can be processed for RNA extraction.

Establishment of Viral Infection and Analysis of Host-Virus Interaction in Drosophila Melanogaster

Virus spreading is a major cause of epidemic diseases. Thus, understanding the interaction between the virus and the host is very important to extend our knowledge of prevention and treatment of viral infection. The fruit fly Drosophila melanogaster has proven to be one of the most efficient and productive model organisms to screen for antiviral factors and investigate virus-host interaction, due to powerful genetic tools and highly conserved innate immune signaling pathways. The procedure described here demonstrates a nano-injection method to establish viral infection and induce systemic antiviral responses in adult flies. The precise control of the viral injection dose in this method enables high experimental reproducibility. Protocols described in this study include the preparation of flies and the virus, the injection method, survival rate analysis, the virus load measurement, and an antiviral pathway assessment. The influence effects of viral infection by the flies' background were mentioned here. This infection method is easy to perform and quantitatively repeatable; it can be applied to screen for host/viral factors involved in virus-host interaction and to dissect the crosstalk between innate immune signaling and other biological pathways in response to viral infection.
