In this work, we described a protocol to prepare rat sciatic nerves for ex vivo neurophysiology. Tissue extraction takes approximately 30 min, including animal handling, anesthesia, culling, and dissection, while nerve cleaning, placement in the bath, and electrode implantation should require an additional 30 min before recording can be started. The current understanding of fundamental nerve function as modeled in silico is lacking in several aspects, notably with respect to the effects of nerve tissue compartmentalization outside of the soma, axon, and dendrites. Axon-myelin interactions are still poorly understood as evidenced by the fact that even detailed computational nerve models such as MRG1 (for mammalian nerves) that adequately capture conventional electrical stimulation response, do not capture other experimentally observed behaviors such as high-frequency block carryover2 or secondary onset response3.
This protocol provides a method to efficiently investigate neurophysiological processes at the nerve level in an acute small laboratory animal model, using a standardized preparation protocol to isolate the nerve, control its environment, and remove it from an in vivo context to an ex vivo context. This will prevent other body processes or anesthetics used by in vivo nerve stimulation protocols to alter nerve behavior and confound measured results or their interpretation4,5. This enables the development of more realistic models focusing solely on effects specific to nerve tissues that are poorly understood. This protocol is also useful as a testbed for new nerve stimulation and recording electrode materials and geometries, as well as new stimulation paradigms such as high-frequency block2,3. Variations of this technique have been used previously to study nerve physiology in tightly controlled conditions6, for example, to measure ion channel dynamics and properties or the effects of local anesthetics7.
This technique provides several advantages compared to alternatives such as acute in vivo small animal experimentation8. The technique obviates the need to maintain anesthesia depth as the tissue has been extracted from the body, reducing the amount of required equipment such as an anesthetic diffuser, oxygen concentrator, and heating pad. This simplifies the experimental protocol, reducing the risk of mistakes. As anesthetics can potentially alter nerve function4, this technique ensures that measures will not be confounded by side effects from these anesthetic compounds. Finally, this technique is more appropriate than acute in vivo experiments when studying the effects of neurotoxic compounds such as tetrodotoxin, which would kill an anesthetized animal by paralysis.
Peripheral nerve sections are a unique ex vivo system since there is a high chance that the fibers responsible for recorded neural signals do not contain any soma. As these would normally be located, for motor neurons, in the spine, and for sensory neurons in the dorsal root ganglia next to the spine, the preparation of a section of the mammalian nerve can be roughly modeled as a collection of tubular membranes with ion channels, open at both ends9. Metabolism is maintained by the mitochondria located in the axon at the time of tissue dissection10. Suturing of the open ends of the axolemma is encouraged after extraction to close them and thereby help maintain existing ionic gradients across the membrane, which are essential for normal nerve function.
To maintain tissue homeostasis outside the body, several environmental variables must be tightly controlled. These are temperature11, oxygenation12, osmolarity, pH13,14, and access to glucose to maintain metabolism. For this protocol, the approach is to use a modified Krebs-Henseleit buffer15,16 (mKHB) continuously aerated with a mixture of oxygen and carbon dioxide. The mKHB is in the family of cardioplegic buffers6,17 used to preserve dissected tissues outside of the body, for example, in ex vivo experiments. These buffers do not contain any hemoglobin, antibiotics, or antifungals and are, therefore, only suitable for preparations involving small amounts of tissue for a limited time. pH control was achieved with the carbonate and carbon dioxide redox pair, requiring constant aeration of the buffer with carbon dioxide to maintain pH equilibrium. This is to avoid using other common buffering agents such as HEPES, which can modify nerve cell function18. To oxygenate the buffer and provide pH control, a mixture of 5% carbon dioxide in oxygen called carbogen (95% O2, 5% CO2) was used. A heating stirrer was used for temperature control of a buffer container, and the buffer was perfused through a nerve bath, and then recirculated to the starting container. A typical experiment would last 6-8 h before the nerve loses its viability and no longer responds sufficiently to stimulation for measures to be representative of healthy tissue.
To optimize the signal-to-noise ratio, silver-chloride electrodes were used for recording, which were prepared according to previously described methods19. For stimulation, a combination of commercial off-the-shelf platinum cuff electrodes and custom-made conductive polymer cuff electrodes can be used. Conductive polymer cuff electrodes have notably higher charge capacities, which are useful when stimulating the nerve using high amplitude waveforms20.
The stimulator used in this protocol has been previously described20. Documentation, design files, and software scripts to use it are publicly available21. Other stimulators can be used to execute this protocol; however, the custom stimulator is also capable of high-frequency alternative current (HFAC) block2,20, which enables a wider range of neurophysiology experiments. To use HFAC block, conductive elastomer cuffs are recommended to avoid damage to the nerve. Conductive elastomer nerve cuffs are soft and fully polymeric electrode arrays produced from conductive elastomers as the conductive component and polydimethylsiloxane as the insulation22. Devices were manufactured in a bipolar configuration using conventional laser microfabrication techniques.

Ex vivo preparations enable the study of many neurophysiological processes in isolation from the rest of the body while preserving local tissue structure. This work describes the preparation of rat sciatic nerves for ex vivo neurophysiology, including buffer preparation, animal procedures, equipment setup and neurophysiological recording. This work provides an overview of the different types of experiments possible with this method. The outlined method aims to provide 6 h of stimulation and recording on extracted peripheral nerve tissue in tightly controlled conditions for optimal consistency in results. Results obtained using this method are A-fibre compound action potentials (CAP) with peak-to-peak amplitudes in the millivolt range over the entire duration of the experiment. CAP amplitudes and shapes are consistent and reliable, making them useful to test and compare new electrodes to existing models, or the effects of interventions on the tissue, such as the use of chemicals, surgical alterations, or neuromodulatory stimulation techniques. Both conventional commercially available cuff electrodes with platinum-iridium contacts and custom-made conductive elastomer electrodes were tested and gave similar results in terms of nerve stimulus strength-duration response.

Representative results that can be obtained with this protocol are the consistent compound action potentials from A-type nerve fibers within the sciatic nerve. These action potentials typically have a peak-to-peak amplitude of approximately 1 mV at the electrode and therefore 100 mV once amplified (Figure 2). This protocol enables the observation of many neurophysiological phenomena that aren't well understood, such as nerve block carryover after high frequency/high amplitude current injection, while preventing interference from other processes in a living body. This technique yields highly repeatable, robust results and is more practical than in vivo electrophysiology, because complex variables such as depth of anesthesia don't have to be controlled and therefore can't influence measurements. After a euthanization procedure under anesthetic, place a female rat on the dissection table, hold the ankle firmly between the thumb, index finger, and middle finger, and sever the calcaneal tendon using 12-centimeter straight blunt scissors.Using fine forceps and scissors, make careful incisions through the muscle layers near the middle of the back of the leg until the sciatic nerve is exposed. Once the nerve is visible, moisturize the cavity using ice-cold modified Krebs-Henseleit buffer, or MKHB, to prevent the nerve from drying out. Using hemostats, pull the flaps of skin on each side and maintain the incision open for finer dissection.Starting from the calcaneal tendon incision location, using fine scissors, interrupt the muscle on the medial side of the leg to free the nerve. Continue to maintain moisture levels in the area with ice-cold MKHB. As the nerve is exposed while moving up the leg, dissect the overlying muscle tissue.Free the nerve nearer to the spine from connective tissues until reaching the spinal cleft, at which point there is a kink in the nerve. Do not clean the nerve yet, as speed is essential at this stage. To make the dissection easier, using forceps, gently pull the end of the nerve near the ankle.Then, using fine scissors, severe the nerve close to the spine. Place the dissected nerve in a 15-milliliter centrifuge tube filled with MKHB and place the tube back on the ice until the start of the cleaning procedure. Fill the coated Petri dish halfway with chilled oxygenated MKHB, then place one dissected sciatic nerve in the dish and pin both ends of the nerve to the dish such that the nerve is straight without kinks, torsion, or twists.Using 6-0 silk sutures or fine thread, tie a double knot around each end of the nerve to prevent cytosol leakage into the buffer. Place the knots just next to the insect pins on the side closer to the center of the nerve to prevent leakage from nerve tissue into the buffer. Under a microscope, using two-millimeter angled spring scissors and fine forceps, remove fat, blood vessels, and muscle tissue from the nerve.Prune any nerve branches that will not be used in the stimulation and recording. After every five minutes of cleaning, replace the buffer with fresh, chilled, oxygenated MKHB. After cleaning, place the nerve back into the transport tube filled with buffer and rest the tube on ice.Prepare a clean, dual-chamber nerve bath. Using standard laboratory boss heads and grippers, place the nerve bath below the level of the two-liter bottle placed on the heating stirrer. Connect the drain of the bath to the peristaltic pump inlet.Connect the outlet of the peristaltic pump to a tube leading back to the buffer bottle. Connect the bath inlet to a tube with an adjustable flow valve and put the tube inside the bottle. Use a three-way valve with a syringe connected to the middle outlet to help with priming the tube as a siphon for gravity-assisted buffer inflow.Prime the siphon by drawing the syringe until buffer flows into it. Configure the valve such that the buffer flow rate into the bath is five to six millimeters per minute. The flow rate can be increased initially to fill the bath.Once the bath buffer level has reached the drain, place the nerves in the bath. Using an insect pin, secure the end of the nerve at the corner of the buffer-filled bath chamber. Using 45-degree angled fine forceps and pinching the nerve only at the ends, carefully thread the nerve to be stimulated through the hole in the partition between the two bath chambers.Using an insect pin, secure the other end of the nerve in the oil chamber of the bath, ensuring that the nerve is straight without being stretched and is free of kinks and twists. Using silicone grease, make a seal to prevent buffer leakage from the buffer chamber into the oil chamber Fill the oil chamber with silicone or mineral oil. Place the silver/silver chloride recording electrode hooks in the oil bath chamber and secure them using boss heads and grippers.Then, drape the portion of the nerve in the oil bath over the hooks without pulling the nerve taut. Adjust or repair the silicone grease seal if any leakage is observed after moving the nerve. Connect the reference silver/silver chloride electrode to the amplifier ground and place the electrode in the buffer-filled bath chamber by securing it using a laboratory gripper.After preparing a clean nerve cuff electrode for stimulation, place the electrode in the buffer-filled bath chamber. Using forceps or fine tweezers with blunt or angled tips, open the electrode in the bath to wet the inside of the cuff. If bubbles remain, use a fine syringe to draw buffer from the bath and force the bubbles out of the cuff.Using a tweezer, gently open the cuff and slide it under the nerve. Close the cuff around the nerve, avoiding any kinking or twisting of the nerve. Connect the stimulation electrode and the current return electrode to the stimulator.If using a square platinum sheet as the current return electrode, position the sheet away from the nerve in the bath. Secure both the electrodes'lead with tape. Connect the stimulator TTL signal output to channel four of the oscilloscope.On the oscilloscope screen, press the Trigger channel tab and specify channel four as the triggering channel. Set the trigger level to one volt using the level knob. On the oscilloscope, set time resolution to one millisecond per division and voltage resolution to 10 millivolts per division.Center the trigger reference in time and set the trigger level to one volt. After connecting the stimulator to the computer, turn on the stimulator by connecting the battery power supply to the power input. Then, start the MATLAB software on the computer.Execute the custom MATLAB script. Then, open the indicated MATLAB script. Edit the script and set the parameters stimulator pulse amplitude to minus 300 microamperes, stimulator pulse width to 300 microseconds, stimulator number of pulses to 10, and stimulator time between pulses to one second.Start the stimulation protocol by clicking on Run in the software. The compound action potential, or CAP, had a peak-to-peak amplitude of one millivolt at the electrode and 100 millivolts when amplified. The typical nerve excitability for standard platinum nerve cuffs and custom-made conductive elastomer nerve cuffs are shown here.The CAP obtained ex vivo over a day of experiments using both sciatic nerves is shown here. Minimal CAP amplitude reduction was observed with the right sciatic nerve. In contrast, the left sciatic nerve CAP amplitude at approximately three millivolts was similar to that of the right sciatic nerve at the start of the experiment more than six hours after nerve extraction.Emphasize precision and repeatability in dissection, cleaning, and implementation technique. Pay attention to what is happening in the bath. If the nerve signal is lost, it may simply be that the electrodes are not in the good contact or that there is saline in the bath oil partition, and not that the nerve is died.First time users might be tempted to rush, especially during the dissection phase. This will likely lead to mistakes and damage the nerve. Precision and consistency are key here and will lead to higher quality results.Special attention should also be given to consistency in buffer preparation.

Axoplasm Isolation from Rat Sciatic Nerve

Isolation of pure axonal cytoplasm (axoplasm) from peripheral nerve is crucial for biochemical studies of many biological processes. In this article, we demonstrate and describe a protocol for axoplasm isolation from adult rat sciatic nerve based on the following steps: (1) dissection of nerve fascicles and separation of connective tissue; (2) incubation of short segments of nerve fascicles in hypotonic medium to release myelin and lyse non-axonal structures; and (3) extraction of the remaining axon-enriched material. Proteomic and biochemical characterization of this preparation has confirmed a high degree of enrichment for axonal components.

Reproducible Mouse Sciatic Nerve Crush and Subsequent Assessment of Regeneration by Whole Mount Muscle Analysis

Regeneration in the peripheral nervous system (PNS) is widely studied both for its relevance to human disease and to understand the robust regenerative response mounted by PNS neurons thereby possibly illuminating the failures of CNS regeneration1. Sciatic nerve crush (axonotmesis) is one of the most common models of peripheral nerve injury in rodents2. Crushing interrupts all axons but Schwann cell basal laminae are preserved so that regeneration is optimal3,4. This allows the investigator to study precisely the ability of a growing axon to interact with both the Schwann cell and basal laminae4. Rats have generally been the preferred animal models for experimental nerve crush. They are widely available and their lesioned sciatic nerve provides a reasonable approximation of human nerve lesions5,4. Though smaller in size than rat nerve, the mouse nerve has many similar qualities. Most importantly though, mouse models are increasingly valuable because of the wide availability of transgenic lines now allows for a detailed dissection of the individual molecules critical for nerve regeneration6, 7. Prior investigators have used multiple methods to produce a nerve crush or injury including simple angled forceps, chilled forceps, hemostatic forceps, vascular clamps, and investigator-designed clamps8,9,10,11,12. Investigators have also used various methods of marking the injury site including suture, carbon particles and fluorescent beads13,14,1. We describe our method to obtain a reproducibly complete sciatic nerve crush with accurate and persistent marking of the crush-site using a fine hemostatic forceps and subsequent carbon crush-site marking. As part of our description of the sciatic nerve crush procedure we have also included a relatively simple method of muscle whole mount we use to subsequently quantify regeneration.

In this report we describe a method to crush mouse sciatic nerve. This method uses readily available hemostatic forceps and easily and reproducibly produces complete sciatic nerve crush. In addition, we describe a method to prepare muscle whole mounts suitable for analysis of nerve regeneration after sciatic nerve crush.

In Vivo Electrophysiological Measurements on Mouse Sciatic Nerves

Electrophysiological studies allow a rational classification of various neuromuscular diseases and are of help, together with neuropathological techniques, in the understanding of the underlying pathophysiology1. Here we describe a method to perform electrophysiological studies on mouse sciatic nerves in vivo.
The animals are anesthetized with isoflurane in order to ensure analgesia for the tested mice and undisturbed working environment during the measurements that take about 30 min/animal. A constant body temperature of 37 &#176;C is maintained by a heating plate and continuously measured by a rectal thermo probe2. Additionally, an electrocardiogram (ECG) is routinely recorded during the measurements in order to continuously monitor the physiological state of the investigated animals.
Electrophysiological recordings are performed on the sciatic nerve, the largest nerve of the peripheral nervous system (PNS), supplying the mouse hind limb with both motoric and sensory fiber tracts. In our protocol, sciatic nerves remain in situ and therefore do not have to be extracted or exposed, allowing measurements without any adverse nerve irritations along with actual recordings. Using appropriate needle electrodes3 we perform both proximal and distal nerve stimulations, registering the transmitted potentials with sensing electrodes at gastrocnemius muscles. After data processing, reliable and highly consistent values for the nerve conduction velocity (NCV) and the compound motor action potential (CMAP), the key parameters for quantification of gross peripheral nerve functioning, can be achieved.

In Vivo Gene Transfer to Schwann Cells in the Rodent Sciatic Nerve by Electroporation

The formation of the myelin sheath by Schwann cells (SCs) is essential for rapid conduction of nerve impulses along axons in the peripheral nervous system. SC-selective genetic manipulation in living animals is a powerful technique for studying the molecular and cellular mechanisms of SC myelination and demyelination in vivo. While knockout/knockin and transgenic mice are powerful tools for studying SC biology, these methods are costly and time consuming. Viral vector-mediated transgene introduction into the sciatic nerve is a simpler and less laborious method. However, viral methods have limitations, such as toxicity, transgene size constraints, and infectivity restricted to certain developmental stages. Here, we describe a new method that allows selective transfection of myelinating SCs in the rodent sciatic nerve using electroporation. By applying electric pulses to the sciatic nerve at the site of plasmid DNA injection, genes of interest can be easily silenced or overexpressed in SCs in both neonatal and more mature animals. Furthermore, this in vivo electroporation method allows for highly efficient simultaneous expression of multiple transgenes. Our novel technique should enable researchers to efficiently manipulate SC gene expression, and facilitate studies on SC development and function.

In Vivo Electrophysiological Measurement of the Rat Ulnar Nerve with Axonal Excitability Testing

Electrophysiology enables the objective assessment of peripheral nerve function in vivo. Traditional nerve conduction measures such as amplitude and latency detect chronic axon loss and demyelination, respectively. Axonal excitability techniques &#34;by threshold tracking&#34; expand upon these measures by providing information regarding the activity of ion channels, pumps and exchangers that relate to acute function and may precede degenerative events. As such, the use of axonal excitability in animal models of neurological disorders may provide a useful in vivo measure to assess novel therapeutic interventions. Here we describe an experimental setup for multiple measures of motor axonal excitability techniques in the rat ulnar nerve.
The animals are anesthetized with isoflurane and carefully monitored to ensure constant and adequate depth of anesthesia. Body temperature, respiration rate, heart rate and saturation of oxygen in the blood are continuously monitored. Axonal excitability studies are performed using percutaneous stimulation of the ulnar nerve and recording from the hypothenar muscles of the forelimb paw. With correct electrode placement, a clear compound muscle action potential that increases in amplitude with increasing stimulus intensity is recorded. An automated program is then utilized to deliver a series of electrical pulses which generate 5 specific excitability measures in the following sequence: stimulus response behavior, strength duration time constant, threshold electrotonus, current-threshold relationship and the recovery cycle.
Data presented here indicate that these measures are repeatable and show similarity between left and right ulnar nerves when assessed on the same day. A limitation of these techniques in this setting is the effect of dose and time under anesthesia. Careful monitoring and recording of these variables should be undertaken for consideration at the time of analysis.

Use of In Vivo Single-fiber Recording and Intact Dorsal Root Ganglion with Attached Sciatic Nerve to Examine the Mechanism of Conduction Failure

Single-fiber recording has been a classical and effective electrophysiological technique over the last few decades because of its specific application for nerve fibers in the central and peripheral nervous systems. This method is particularly applicable to dorsal root ganglia (DRG), which are primary sensory neurons that exhibit a pseudo-unipolar structure of nervous processes. The patterns and features of the action potentials passed along axons are recordable in these neurons. The present study uses in vivo single-fiber recordings to observe the conduction failure of sciatic nerves in complete Freund&#8217;s adjuvant (CFA)-treated rats. As the underlying mechanism cannot be studied using in vivo single-fiber recordings, patch-clamp-recordings of DRG neurons are performed on preparations of intact DRG with the attached sciatic nerve. These recordings reveal a positive correlation between conduction failure and the rising slope of the after-hyperpolarization potential (AHP) of DRG neurons in CFA-treated animals. The protocol for in vivo single fiber-recordings allows the classification of nerve fibers via the measurement of conduction velocity and monitoring of abnormal conditions in nerve fibers in certain diseases. Intact DRG with attached peripheral nerve allows observation of the activity of DRG neurons in most physiological conditions. Conclusively, single-fiber recording combined with electrophysiological recording of intact DRGs is an effective method to examine the role of conduction failure during the analgesic process.

In order to obtain high-quality electron micrographs that can yield the degree of morphological detail necessary for quantitative analysis of pre-synaptic vesicle distribution, optimal specimen preparation is critical. Chemical fixation is the first step in the process of specimen preparation, and of utmost importance to preserve fine ultrastructure. Vascular fixation with a glutaraldehyde-formaldehyde solution, followed by treatment of vibratome-sectioned specimens with osmium tetroxide, stabilizes the maximum number of molecules, especially proteins and lipids, and results in superior conservation of ultrastructure. Tissue is then processed with counterstaining, sequential dehydration and resin-embedding. En bloc staining with uranyl acetate (i.e., staining of vibratome-sectioned tissue before resin embedding) enhances endogenous contrast and stabilizes cell components against extraction during specimen processing. Contrast can be further increased by applying uranyl acetate as a post-stain on ultrathin sections. Double-staining of ultrathin sections with lead citrate after uranyl acetate treatment also improves image resolution, by intensifying electron-opacity of nucleic acid-containing structures through selective binding of lead to uranyl acetate. Transmission electron microscopy is a powerful tool for characterization of the morphological details of synaptic vesicles and quantification of their size and spatial organization in the terminal bouton. However, because it uses fixed tissue, transmission electron microscopy can only provide indirect information regarding living or evolving processes. Therefore, other techniques should be considered when the main objective is to study dynamic or functional aspects of synaptic vesicle trafficking and exocytosis.

We describe procedures for processing newborn rat brain tissue to obtain high-resolution electron micrographs for morphometric analysis of synaptic vesicle distribution at nerve terminals. The micrographs obtained with these methods can also be used to study the morphology of a number of other cellular components and their dimensional structural relationships.

Ex Vivo Oculomotor Slice Culture from Embryonic GFP-Expressing Mice for Time-Lapse Imaging of Oculomotor Nerve Outgrowth

Accurate eye movements are crucial for vision, but the development of the ocular motor system, especially the molecular pathways controlling axon guidance, has not been fully elucidated. This is partly due to technical limitations of traditional axon guidance assays. To identify additional axon guidance cues influencing the oculomotor nerve, an ex vivo slice assay to image the oculomotor nerve in real-time as it grows towards the eye was developed. E10.5 IslMN-GFP embryos are used to generate ex vivo slices by embedding them in agarose, slicing on a vibratome, then growing them in a microscope stage-top incubator with time-lapse photomicroscopy for 24-72 h. Control slices recapitulate the in vivo timing of outgrowth of axons from the nucleus to the orbit. Small molecule inhibitors or recombinant proteins can be added to the culture media to assess the role of different axon guidance pathways. This method has the advantages of maintaining more of the local microenvironment through which axons traverse, not axotomizing the growing axons, and assessing the axons at multiple points along their trajectory. It can also identify effects on specific subsets of axons. For example, inhibition of CXCR4 causes axons still within the midbrain to grow dorsally rather than ventrally, but axons that have already exited ventrally are not affected.

3D Kinematic Analysis for the Functional Evaluation in the Rat Model of Sciatic Nerve Crush Injury

Compared to the Sciatic Functional Index (SFI), kinematic analysis is a more reliable and sensitive method for performing functional evaluations of sciatic nerve injury rodent models. In this protocol, we describe a novel kinematic analysis method that uses a three-dimensional (3D) motion capture apparatus for functional evaluations using a rat sciatic nerve crush injury model. First, the rat is familiarized with treadmill walking. Markers are then attached to the designated bone landmarks and the rat is made to walk on the treadmill at the desired speed. Meanwhile, the posterior limb movements of the rat are recorded using four cameras. Depending on the software used, marker tracings are created using both automatic and manual modes and the desired data are produced after subtle adjustments. This method of kinematic analysis, which uses a 3D motion capture apparatus, offers numerous advantages, including superior precision and accuracy. Many more parameters can be investigated during the comprehensive functional evaluations. This method has several shortcomings that require consideration: The system is expensive, can be complicated to operate, and may produce data deviations due to skin shifting. Nevertheless, kinematic analysis using a 3D motion capture apparatus is useful for performing functional anterior and posterior limb evaluations. In the future, this method may become increasingly useful for generating accurate assessments of various traumas and diseases.

Ex Vivo Pressurized Hippocampal Capillary-Parenchymal Arteriole Preparation for Functional Study

From subtle behavioral alterations to late-stage dementia, vascular cognitive impairment typically develops following cerebral ischemia. Stroke and cardiac arrest are remarkably sexually dimorphic diseases, and both induce cerebral ischemia. However, progress in understanding the vascular cognitive impairment, and then developing sex-specific treatments, has been partly limited by challenges in investigating the brain microcirculation from mouse models in functional studies. Here, we present an approach to examine the capillary-to-arteriole signaling in an ex vivo hippocampal capillary-parenchymal arteriole (HiCaPA) preparation from mouse brain. We describe how to isolate, cannulate, and pressurize the microcirculation to measure arteriolar diameter in response to capillary stimulation. We show which appropriate functional controls can be used to validate the HiCaPA preparation integrity and display typical results, including testing potassium as a neurovascular coupling agent and the effect of the recently characterized inhibitor of the Kir2 inward rectifying potassium channel family, ML133. Further, we compare the responses in preparations obtained from male and female mice. While these data reflect functional investigations, our approach can also be used in molecular biology, immunochemistry, and electrophysiology studies.
