1. 시약의 준비

	<li>항체 용액: β2 integrin의 고친화성 형태를 보고하는 알로피코시아닌(APC)-복합 단클론 항체 mAb24(mAb24-APC) 120μL를 호중구 배지 10mL에 희석하여 1.2μg/mL mAb24-APC 용액을 만듭니다.</li>

</ol>2. 인간 혈액에서 호중구 분리
<ol><li>혈청학적 피펫을 사용하여 15mL 원심분리 튜브에 시판되는 밀도 구배 배지( 재료 표 참조) 8mL 위에 4mL의 혈액을 조심스럽게 층층합니다(그림 2A).</li>
	<li>20°C에서 30-50분 동안 550× g 에서 혈액을 원심분리합니다. 로터를 점진적으로 감속합니다(감속 계수 1). 참고: 적혈구에서 호중구( 그림 2B의 밴드 2)를 성공적으로 분리하는 것은 기증자마다 다를 수 있습니다. 대부분의 기증자의 경우 30분의 원심분리로 충분합니다. 그러나 일부 기증자의 경우 분리가 성공하지 못할 경우 추가로 10-30분의 원심분리가 필요할 수 있습니다(그림 2C).</li>
	<li>플라즈마(상단의 노란색 액체)와 단핵 전지(상단의 흐린 띠; 도 2B의 PBMC 밴드)를 1mL 피펫으로 사용하였다.</li>

	<li>혈구계를 사용하여 세포 수를 계산합니다. 전형적으로, 1 내지 4 × 107 호중구는 8 mL의 인간 혈액으로부터 얻어졌다. 알림: 호중구 현탁액에 적혈구 오염이 있을 수 있습니다. 적혈구는 대부분의 분석에 큰 영향을 미치지 않으며 호중구 활성화/프라이밍을 예방할 수 있다27. 정확한 호중구 농도를 얻기 위해 계산하기 전에 적혈구를 용해해야 합니다. 적혈구를 용해하기 위해 10-30초 동안 891μL의 탈이온수에 세포 현탁액 10μL를 첨가한 다음 99μL의 10× PBS를 추가하여 삼투압의 균형을 유지하여 호중구의 용해를 방지합니다.</li>
	<li>호중구 배지를 추가하여 세포 밀도를 6.25 × 105 cells/mL로 조정합니다.</li>
</ol>3. 384웰 플레이트의 준비
<ol><li>1.5mL 원심분리 튜브에 1mL의 항체 용액(1.2μg/mL mAb24-APC)과 0.2mL의 호중구 배지를 결합하여 1μg/mL mAb24-APC 용액을 만듭니다. 그런 다음 이 혼합물 25μL를 384웰 플레이트의 음성 대조군 웰( 그림 1의 파란색 웰)에 추가합니다.</li>
	<li>15mL 원심분리 튜브에 9mL의 항체 용액을 21.6μL의 fMLP 용액(100μM) 및 1.7784mL의 호중구 배지와 혼합하여 1μg/mL mAb24-APC 및 200nM fMLP를 포함하는 용액을 만듭니다. 다음으로, 이 혼합물 25μL를 384웰 플레이트( 그림 1의 빨간색 및 파란색 웰)의 양성 대조군 및 테스트 웰에 추가합니다.</li>

</ol>4. 세포의 처리
<ol><li>16채널 피펫을 사용하여 각 웰에 20μL의 호중구 현탁액을 추가합니다(384웰 플레이트의 각 컬럼에 대해 5-50μL 범위). 피펫을 사용하여 5-10회 부드럽게 혼합하고 각 피펫팅 사이에 일정한 시간 간격을 유지합니다. 참고: 웰의 최종 농도는 다음과 같습니다: 호중구 2.5 × 105 cells/mL(모든 웰); mAb24-APC 0.5μg/mL(모든 웰); fMLP 100nM(포지티브 대조군 및 테스트 웰); 화합물 10μM(테스트 웰).</li>
	<li>300rpm의 Shaker에서 실온(RT)에서 10분 동안 배양합니다.</li>

</ol>5. 유세포 분석

	<li>피복 유체 용기가 채워져 있고 폐기물 용기가 비어 있고 기기에 제대로 연결되어 있는지 확인하십시오.</li>
	<li>384웰 플레이트를 유세포 분석기의 샘플 로더에 로드합니다. 플레이트의 A1 웰이 로더의 A1 표시와 정렬되는지 확인하십시오.</li>
	<li>소프트웨어를 열고 새 실험을 만듭니다. 384 well 을 선택하고 원하는 형광단을 선택합니다. 그런 다음 플레이트 설정(Plate setup)을 클릭하고 드래그하여 모든 웰을 선택한 다음 샘플( Sample)을 클릭합니다. "High throughput(높은 처리량)" 스위치를 켜고 rate 패널에서 High(높음 )를 선택합니다.<ol><li>볼륨 상자에 50을 입력합니다. 홀수 열에서 샘플을 선택한 다음 "교반" 스위치를 활성화합니다. 마지막으로 오른쪽 패널에서 샘플의 이름을 지정합니다.</li>

	<li>유세포 분석기는 각 웰에서 ~50μL의 1000-3000 호중구를 순차적으로 샘플링합니다. 전체 384웰 플레이트를 완성하는 데 약 3시간이 걸립니다.</li>
	<li>모든 샘플을 완료한 후 다음 단계를 위해 .fcs 파일을 내보냅니다.</li>
</ol>6. 데이터 분석
<ol><li>유세포 분석 데이터 분석 소프트웨어를 엽니다( 재료 표 참조). 모든 .fcs 파일이 포함된 폴더를 선택하여 소프트웨어 창으로 드래그한 다음 손을 떼면 데이터가 소프트웨어로 가져옵니다.</li>
	<li>그림 3과 같이 팝업 창에서 FSC 및 SSC28,29,30을 기반으로 하는 단일 호중구를 게이트하는 샘플 하나를 두 번 클릭합니다. 게이트 행을 선택하고 폴더로 끈 다음 놓아 해당 폴더의 모든 샘플에 게이팅을 적용합니다.</li>
	<li>소프트웨어의 테이블 편집기 기능을 사용하여 각 웰에서 호중구의 APC 평균 형광 강도(MFI)를 계산하고 내보낼 수 있습니다. Edit(편집) 탭에서 Add Column(열 추가)을 클릭합니다. 통계량 탭에서 중앙값을 선택하고, 게이트 모집단 호중구를 선택하고, APC를 매개변수로 선택합니다. 확인 버튼을 클릭합니다.</li>
	<li>"Table editor" 탭으로 돌아가서 "Group" 탭에서 All Samples 를 선택하고 Create table 버튼을 누릅니다. 생성된 테이블을 표시하는 새 창이 나타납니다. 텍스트, CSV 또는 Excel 파일로 저장합니다.</li>
	<li>내보낸 표를 열고 양성 및 음성 대조군 샘플에 대한 APC MFI의 평균값과 표준 편차를 계산합니다(그림 4).</li>
	<li>다음 방정식을 사용하여 플레이트의 Z'-계수(Z')를 계산합니다. Z' = 1 - 3(σ p + σ n)/(μp - μn), 여기서σ p와 σ n은 각각 양성 및 음성 대조군의 표준 편차이고,μ p 및 μn은 각각 양성 대조군과 음성 대조군의 평균입니다(31). 참고: Z' 계수는 양성 및 음성 제어 분포의 분리를 나타냅니다. β2 인테그린은 이전에도 염증성 질환의 표적이 된 적이 있습니다. 인테그린 αLβ2를 직접 표적으로 하는 단클론 항체인 에팔리주맙(Efalizumab)은 건선5를 치료하기 위해 개발되었습니다. 그러나 에팔리주맙은 JC 바이러스 재활성화로 인한 진행성 다발성 백질뇌병증이라는 치명적인 부작용으로 인해 철회되었다 6,7. 새로운 항염증 인테그린 기반 요법은 부작용을 최소화하기 위해 백혈구의 항감염 기능을 유지하는 것을 고려해야 합니다. 에팔리주맙의 부작용은 혈류 내 단일클론항체의 장기적 순환으로 인해 발생할 수 있으며, 이는 장기적으로 면역 기능을 저해할 수 있다8. 최근 연구에 따르면 에팔리주맙은 αLβ2 교차결합과 α4 인테그린의 원치 않는 내재화를 매개하여 부작용에 대한 대안적 설명을 제공한다9. 따라서 수명이 짧은 저분자 길항제는 이 문제를 피할 수 있습니다.

Many inflammatory diseases are characterized by the infiltration of neutrophils at the site of swelling or injury1. To infiltrate these tissues, neutrophils must complete the neutrophil recruitment cascade, which involves arrest to the endothelium, extravasation across the vessel wall, and recruitment into the tissue2. Circulating neutrophils need &#946;2 integrin activation to complete this cascade, especially for the arrest phase. Thus, integrin-inhibiting drugs that reduce neutrophil adhesion, extravasation, and recruitment may effectively treat inflammatory diseases3,4.
&#946;2 integrins have been targeted for inflammatory diseases before. Efalizumab, a monoclonal antibody directly targeting integrin &#945;L&#946;2, was developed to treat psoriasis5. However, efalizumab was withdrawn due to its lethal side effect - progressive multifocal leukoencephalopathy resulting from JC virus reactivation6,7. New anti-inflammatory integrin-based therapies should consider maintaining the anti-infection functions of leukocytes to minimize side effects. The side effects of efalizumab might be due to the prolonged circulation of monoclonal antibodies in the bloodstream, which could inhibit immune functions in the long term8. A recent study shows that efalizumab mediates &#945;L&#946;2 crosslinking and the unwanted internalization of &#945;4 integrins, providing an alternative explanation for the side effects9. Thus, short-lived, small-molecule antagonists might avoid this problem.
A high-throughput method to screen small-molecule &#946;2 integrin antagonists using human neutrophils is presented here. &#946;2 integrin activation requires conformational changes of the integrin ectodomain to gain access to and increase its binding affinity to its ligand. In the canonical switchblade model, the bent-closed integrin ectodomain first extends to an extended-closed conformation and then opens its headpiece to a fully activated extended-open conformation10,11,12,13. There is also an alternative pathway that starts from the bent-closed to bent-open and extended-open, eventually14,15,16,17,18,19. The conformation-specific antibody mAb24 binds to an epitope in the human &#946;2-I-like domain when the headpiece of the ectodomain is open20,21,22,23.
Here, mAb24-APC is used to determine whether the &#946;2 integrins are activated. To activate neutrophils and integrin, N-formylmethionyl-leucyl-phenylalanine (fMLP), a bacterial-derived short chemotactic peptide that can activate neutrophil &#946;2 integrins24, is used as a stimulus in this protocol. When fMLP binds to the Fpr1 on neutrophils, downstream signaling cascades involving G-proteins, phospholipase C&#946;, and phosphoinositide 3-kinase &#947; are activated. These signaling events ultimately result in integrin activation via the inside-out signaling pathway18,25. Besides small molecule antagonists that directly bind to &#946;2 integrins and prevent conformational changes of integrin activation26, compounds that can inhibit components in the &#946;2 integrin inside-out activation signaling pathway would also be detected with this method. Automated flow cytometers enable high-throughput screening. Identifying new antagonists may not only deepen our understanding of integrin physiology but also provide translational insight into integrin-based anti-inflammation therapy.

This protocol aims to establish a method for identifying small molecular antagonists of &#946;2 integrin activation, utilizing conformational-change-reporting antibodies and high-throughput flow cytometry. The method can also serve as a guide for other antibody-based high-throughput screening methods. &#946;2 integrins are leukocyte-specific adhesion molecules that are crucial in immune responses. Neutrophils rely on integrin activation to exit the bloodstream, not only to fight infections but also to be involved in multiple inflammatory diseases. Controlling &#946;2 integrin activation presents a viable approach for treating neutrophil-associated inflammatory diseases. In this protocol, a monoclonal antibody, mAb24, which specifically binds to the high-affinity headpiece of &#946;2 integrins, is utilized to quantify &#946;2 integrin activation on isolated primary human neutrophils. N-formylmethionyl-leucyl-phenylalanine (fMLP) is used as a stimulus to activate neutrophil &#946;2 integrins. A high-throughput flow cytometer capable of automatically running 384-well plate samples was used in this study. The effects of 320 chemicals on &#946;2 integrin inhibition are assessed within 3 h. Molecules that directly target &#946;2 integrins or target molecules in the G protein-coupled receptor-initiated integrin inside-out activation signaling pathway can be identified through this approach.
