ヒト前立腺には、分泌機能を有する明るい上皮と、その根底にある基底細胞と、異常な神経内分泌細胞成分が含まれています。上皮細胞は、この場合、生体内で比較的静止している前立腺幹細胞の稀な集団から生成され、生涯を通じて腺恒常性を維持するための修復システムとして作用する。多くの進歩にもかかわらず、前立腺幹細胞の同定および機能的単離は、この分野における主要な課題であり続ける。幹細胞バイオマーカーは、フローサイトメトリーと組み合わせた細胞表面マーカーベースの方法論を含む、幹細胞研究2、3、4に一般的に使用される。しかしながら、濃縮および単離の結果は、マーカーの組み合わせおよび抗体特異性5、6の機能として大きく異なり、単離された細胞の同一性に関する疑問を提起する。幹細胞様細胞濃縮のためのもう一つの広く用いられているアプローチは、三次元(3D)回転楕円体培養物2、3、4である。常駐幹細胞はニッチ拘束を伴う生体内で比較的静止しているが、それらは3Dマトリックス培養(対称と非対称の両方)で細胞分裂を受け、系統コミットメント7、8に向けて急速に再生する幹細胞および前駆細胞の両方を生成する。形成された回転楕円体は、初期段階および後期前駆細胞を含む系統コミットメントの様々な段階で幹細胞と前駆細胞の両方を含む異種混合物である。したがって、スフェロイド全体を用いたアッセイは幹細胞を排他的ではなく、独特の幹細胞特性の同定は決定的ではない。したがって、前立腺幹細胞を識別し、娘の前駆体から分離するアッセイを作成することが重要です。この目的に向かって、現在のプロトコルの目標は、ヒト前立腺組織からの幹細胞の効率的な同定と単離を可能にするアッセイシステムを確立し、その生物学的機能の堅牢な下流分析を行うことです。
長期5-ブロモ-2'-デオキシウリジン(BrdU)標識保持は、その長期倍増時間9、10に基づいて幹細胞のインビボおよびインビトロ系統トレースに広く使用されている。本明細書に記載される前立腺幹細胞同定および単離のための現在のアプローチは、混合上皮集団内の相対的な静止特性および標識保持特性に基づいている。さらに、不死鎖DNA仮説に基づいて、幹細胞のみが非対称細胞分裂を受けることができる。幹細胞は、古い親のDNAを含む娘細胞を表し、コミットされた娘細胞である前駆細胞は、新たに合成されたDNAを受け取ります。上記のユニークな幹細胞特性は、一次培養における親幹細胞におけるBrdU標識を行い、3Dアンカーフリー回転楕円体培養への転写時にBrdU-ウォッシュアウト後にそのラベルを追跡するために利用される。一次前立腺上皮細胞の大部分は、2D培養において基底および通過増幅表現型を保持する一方で、3Dシステム3、12への転移時に対応する培養媒体を有するスフェロイドまたは完全に分化したオルガノイドの形成によって証明されるように、多能性幹細胞の補充および維持の稀な集団もある。現在のプロトコルでは、HPrEC前立腺回転楕円体またはプロスタスフィアベースのBrdU、CFSE、またはファーレッド保持アッセイを使用し、続いて蛍光活性化細胞選別(FACS)を使用して、単一細胞レベル13でスフェロイド中の標識保持幹細胞を同定する。
重要なことに、系統へのコミットメントを有する前駆細胞と比較して、早期スフェロイド内の標識保持細胞の幹細胞特性をさらに確認した。これらには、幹細胞不斉分裂、インビトロ回転楕円体形成能および生体内組織再生能、オートファジー活性の上昇、増強リボソーム生体形成および代謝活性の低下が含まれる。続いて、RNAAseq分析を行った。標識保持スフェロイド細胞において微分発現した遺伝子は、ヒト前立腺幹細胞の新規バイオマーカーとなる可能性が認められた。この回転楕円体ベースの標識保持アプローチは、少数の癌幹細胞を同様に同定する癌検体に適用され得る。以下に提示する、ヒト原発性前立腺上皮細胞(HPrEC)を例に用いたプロスタスフィアベースの標識保持アッセイである。

成体幹細胞研究の進歩にもかかわらず、組織標本からの幹細胞の同定と単離は依然として大きな課題である。常駐幹細胞は成体組織のニッチ拘束を伴う比較的静止しているが、アンカーフリー三次元(3D)培養では細胞周期に入り、対称細胞分裂と非対称細胞分裂の両方を受け、幹と前駆体の両方を生み出す細胞。後者は急速に増殖し、系統コミットメントの様々な段階で主要な細胞集団であり、異種回転楕円体を形成する。原発性正常ヒト前立腺上皮細胞(HPrEC)を用いて、スフェロイドベースの標識保持アッセイを用いて、スフェロイド開始幹細胞の同定と機能的単離を単一細胞分解能で可能にする。
HPrECまたは細胞株は、自己更新幹細胞を含むすべての分裂細胞のDNAへの組み込みを可能にするために、BrdUで10日間培養された2次元(2D)である。3D培養への転移時に5日間洗い流し、その間、幹細胞は非対称分裂を介して自己更新し、回転楕円体形成を開始する。比較的静止した娘幹細胞はBrdU標識の親DNAを保持するが、娘の前駆者は急速に増殖し、BrdU標識を失う。BrdUは、FACSによる生きた幹細胞分離を可能にするCFSEまたはファーレッドのプロ染料で置き換えることができます。幹細胞の特徴は、インビトロ回転楕円体形成、生体組織再生アッセイ、およびそれらの対称/非対称細胞分裂を文書化することによって確認される。単離された標識保持幹細胞は、RNA-seq、ChIP-seq、単一細胞捕捉、代謝活性、プロテオームプロファイリング、免疫細胞化学、オルガノイド形成、および生体内組織再生を含む下流の分子および生物学的研究によって厳密に尋問することができる。重要なことに、このマーカーフリー機能性幹細胞分離アプローチは、新鮮な癌標本および癌細胞株から幹細胞細胞を複数の器官から識別し、広い適用性を示唆している。がん幹細胞様細胞バイオマーカーの同定、がん幹細胞様細胞を標的とする医薬品のスクリーニング、がんの新しい治療標的の発見に利用できます。

原発正常ヒト前立腺上皮細胞は、フィブロネクチン被覆培養皿に入れられ、細胞増殖は2D培養中に維持される(図1a)。基底膜マトリックスで3D培養に移ると、分化した上皮細胞はゆっくりと死ぬ。前立腺幹細胞だけがアンカーフリー培養で生き残ることができ、5日間で回転楕円体を形成する(図1b)。

幹細胞の分離は、現在の方法を使用した前駆細胞の共分離による主要な課題であり続けています。我々は、静止幹細胞を混合前駆物質から分離するための新しいバイオマーカーフリーアッセイを開発した。このスフェロイドベースのラベル保持アッセイの主な利点は、CFSEまたはファーレッド標識を使用してFACSによって娘の前駆細胞から生きた幹細胞を拡大および分離することができる。従来の治療法ではほとんどの前立腺癌細胞が根絶されますが、がん幹細胞は治療抵抗のために残り、病気の進行を促進します。幹細胞様細胞分離は、効果的な疾患寛解のために治療的に共同標的化することができる新しい遺伝子の同定を可能にする。当社のラベル保持アッセイは、様々な組織や細胞からの正常な幹細胞分離、および癌幹細胞研究に適用可能です。重要なことに、このアッセイは、乳癌および大腸癌などの他の上皮細胞癌に適用可能である。100ミリメートルの培養皿を2.5マイクログラムフィブロネクチン溶液の2ミリリットルでコーティングし、室温で一晩インキュベートします。その後、溶液を吸引し、培養皿をバイオセーフティキャビネット内で45分間空気乾燥させます。前立腺上皮細胞増殖培地をフィブロネクチンコーティング皿に9ミリリットル加え、摂氏37度の二酸化炭素インキュベーターで温かく保ちます。37°Cの水浴で凍結したヒト前立腺上皮細胞のバイアルを1バイアルで解凍し、10ミリリットルの温かい培地で細胞を再懸濁する。5分間500回gで細胞懸濁液を遠心分離する。その後、吸引し、上清を捨てます。細胞を温かい培養培地の1ミリリットルに再懸濁し、1ミリリットルの懸濁液を、あらかじめ温めた培地でフィブロネクチンコーティング皿に直接移す。その後、摂氏37度で二酸化炭素インキュベーターに皿をインキュベートします。細胞に共同標識を付ける場合は、テキスト原稿に記載されている10日間の2D培養でBrdU標識細胞を調製する。その後、5つのマイクロモルCFSEまたはファーレッドを加え、30分間細胞をインキュベートします。インキュベーション後、慎重に色素で培地を吸引し、5ミリリットルの温かいPBSで細胞を2回洗浄する。次に、原稿の指示に従って05%トリプシンEDTAで酵素消化を行う。500回gで細胞を5分間遠心し、上清を捨てます。細胞を氷冷、1対1の基体膜マトリックスおよび培養培地ミックス中に再懸濁し、3Dプロスフィア形成を行った。3D基質マトリックスシステムでプロスフィア培養を調製するには、基膜マトリックスを摂氏4度で一晩解凍し、使用前に氷の上に保管します。その後、氷冷基膜マトリックスの1ミリリットルを同量の氷冷培養培地にそっと加えます。ピペットは上下に混ざり、気泡を導入しないように注意してください。氷冷1対1基膜マトリックスと培養培地ミックス中のヒト前立腺上皮細胞の5倍を50~1/4回再懸濁し、合計体積500マイクロリットルを混合する。各ウェルの下縁に溶液をピペットし、均等に混合物を分配するためにプレートを旋回します。プレートを37°Cの二酸化炭素インキュベーターに30分間置き、マトリックスを固めます。その後、リングを乱さないように、井戸あたり1ミリリットルの温かい培養培地で覆います。CFSE標識プロスタフェアを収穫するには、2ミリリットルのディスパーゼとピペットを上下に置き換えて数回混合します。37°Cでプレートを30分間インキュベートしてマトリックスを消化し、球体混合物を15ミリリットルの遠心分離管に集めます。500回gで球を5分間遠心し、上清を捨てます。ペレットを暖かい05%トリプシンEDTAの500マイクロリットルに再懸濁し、1.5ミリリットルの遠心分離管に移します。37°Cで5分間、球をインキュベートし、10%FBSで500マイクロリットルの温かいPBSを加えます。26ゲージの針で1ミリリットルの注射器を通して、球体を完全に解体します。5分間500回gで細胞を遠心分離する。その後、吸引し、上清を捨てます。1ミリリットルの温かい培養培地で細胞を1ミリリットルに再懸濁し、ヨウ化プロピジウム1ミリグラムで死んだ細胞を染色し、室温で1分間インキュベートする。遠心分離を繰り返し、上清を捨てます。その後、1ミリリットルの温かい媒体で細胞を洗います。遠心分離を繰り返し、上清を捨てます。温かい培養培地の1ミリリットルで細胞を再懸濁し、35ミクロンの孔サイズの細胞ストレーナースナップキャップを介してそれらを濾過することによって、5ミリリットルポリスチレンラウンドボトムチューブで細胞を収集します。トリプシン分散CFSE標識プロスタ球細胞のFACS分析を行う。非標識セルを負のコントロールとして使用し、ゲートを設定するための正のコントロールとして CFSE 標識セルを使用して、サンプルを実行して、分数が設定された CFSE 高セルおよび CFSE 低セルのサブオーバジストを並べ替えて収集します。一次正常ヒト前立腺上皮細胞をフィブロネクチンコーティング培養皿に入れ、細胞増殖を2D培養で維持した。基体膜マトリックスを用いた3D培養物に移ると、分化した上皮細胞はゆっくりと消滅し、前立腺幹細胞だけが残った。2D培養における前立腺上皮細胞の二重標識は、3D培養におけるスフェロイド形成に続いて、同じ標識保持細胞におけるBrdU、CFSE、およびファーレッドの共局在化を示した。二重免疫染色は、標識保持細胞がサイトケラチンタンパク質ケラチン14の低レベルを示し、細胞結合タンパク質E-カドヘリンを減少させ、幹細胞初期マーカータンパク質Wnt10bおよびALDH1A1を増加させ、オートファジータンパク質LC3を増加させ、およびミオシンIIBを増加させることを示した。さらに、スフェロイド系のラベル保持アッセイは、前立腺癌検体における癌幹細胞様細胞の検出に成功した。CFSE標識保持幹細胞様癌細胞およびヒト前立腺癌検体由来のスフェロイドは、非標識前駆細胞に対してEカドヘリンタンパク質の減少を示した。このプロトコルを試みる際には、培地調製用の液体状のマトリックスを維持することが重要である。4度で一晩保存し、マトリックスを分配する前に氷冷PBSでピペットの先端を冷やします。スフェロイドベースのラベル保持アッセイを用いた幹細胞の単離後、単細胞RNAシーケンシングおよびプロテオーム解析を行い、幹細胞内の特定の遺伝子および新規バイオマーカーを同定することができる。スフェロイドベースの標識保持アッセイによる生きた癌幹細胞の単離は、研究者が癌幹細胞由来の腫瘍化をインビトロで成長させ、新しい抗癌薬をスクリーニングすることを可能にする。
